Aluminium/ammonium perchlorate (40/60 wt.%) is a pyrotechnic composition widely used as an igniter charge for small calibre rocket motors. Five different types of aluminium powders with specific surface area varying from 0.52 m2/g to 6.00 m2/g have been studied to evaluate the influence of specific surface area on the ignition characteristics of the charge. While the peak pressure varies from 35.5 ksc to 54.5 ksc (0.30 g in 48.0 cm3 vol.), the corresponding time to Pmax varies from 11.2 ms to 1.6 ms showing higher reactivity of the mix with increase in aluminium surface area. This order of reactivity agrees with the order of reactivity determined from the DTG curves of the metal oxidation by comparing the peak height to half width ratio. However, the total gas generated by all the charges is approximately the same and the difference in peak pressure is caused by the difference in the rate of reaction and after about 30 ms all the compositions exhibit the same order of temperature and pressure. The charge has got an ignition temperature of about 640 K and the ignition is preceded by the exothermic decomposition of ammonium perchlorate. The activation energy varies from 92.2 kJ/mol to 127.6 kJ/mol and the corresponding pre-exponential factor from 9.7 × 106 min−1 to 7.1 × 109 min−1 and they exhibit the kinetic compensation effect with an isokinetic temperature of 647 K.  相似文献

Loss of chromosome 10q is a frequently observed genetic defect in prostate cancer. Recently, the PTEN/MMAC1 tumor suppressor gene was identified and mapped to chromosome 10q23.3. We studied PTEN structure and expression in 4 in vitro cell lines and 11 in vivo xenografts derived from six primary and nine metastatic human prostate cancers. DNA samples were allelotyped for eight polymorphic markers within and surrounding the PTEN gene. Additionally, the nine PTEN exons were tested for deletions. In five samples (PC3, PC133, PCEW, PC295, and PC324), homozygous deletions of the PTEN gene or parts of the gene were detected. PC295 contained a small homozygous deletion encompassing PTEN exon 5. In two DNAs (PC82 and PC346), nonsense mutations were found, and in two (LNCaP and PC374), frame-shift mutations were found. Missense mutations were not detected. PTEN mRNA expression was clearly observed in all cell lines and xenografts without large homozygous deletions, showing that PTEN down-regulation is not an important mechanism of PTEN inactivation. The high frequency (60%) of PTEN mutations and deletions indicates a significant role of this tumor suppressor gene in the pathogenesis of prostate cancer.  相似文献

1. The serotonin type 1A (5-HT1A) receptors are members of a superfamily of seven transmembrane domain receptors that couple to GTP-binding regulatory proteins (G-proteins). We have studied the modulation of agonist binding to 5-HT1A receptors from bovine hippocampus by metal ions and guanine nucleotide. 2. Bovine hippocampal membranes containing the 5-HT1A receptor were isolated. These membranes exhibited high-affinity binding sites for the specific agonist [3H]OH-DPAT. 3. The agonist binding is inhibited by monovalent cations Na+, K+, and Li+ in a concentration-dependent manner. Divalent cations such as Ca2+, Mg2+, and Mn2+, on the other hand, show more complex behavior and induce enhancement of agonist binding up to a certain concentration. The effect of the metal ions on agonist binding is strongly modulated in the presence of GTP-gamma-S, a nonhydrolyzable analogue of GTP, indicating that these receptors are coupled to G-proteins. 4. To gain further insight into the mechanisms of agonist binding to bovine hippocampal 5-HT1A receptors under these conditions, the binding affinities and binding sites have been analyzed by Scatchard analysis of saturation binding data. Our results are relevant to ongoing analyses of the overall regulation of receptor activity for G-protein-coupled seven transmembrane domain receptors.  相似文献

Peptide toxins have proved to be useful agents, both in discriminating between different components of native calcium channel currents and in the molecular isolation and designation of their cloned channel counterparts. Here, we describe the isolation and characterization of the biochemical and physiological properties of a novel 74-amino acid peptide toxin (DW13.3) extracted from the venom of the spider Filistata hibernalis. The subtype specificity of DW13.3 was investigated using calcium channel currents recorded from two separate expression systems and several different cultured mammalian cell preparations. Overall, DW13.3 potently blocked all native calcium channel currents studied, with the exception of T-type currents recorded from GH3 cells. Examination of transiently expressed calcium channels in oocytes showed that DW13.3 had the highest affinity for alpha1A, followed by alpha1B > alpha1C > alpha1E. The affinity of DW13.3 for alpha1B N-type currents varied by 10-fold between expressed channels and native currents. Although block occurred in a similar 1:1 manner for all subtypes, DW13.3 produced a partial block of both alpha1A currents and P-type currents in cerebellar Purkinje cells. Selective occlusion of the P/Q-type channel ligand omega-conotoxin MVIIC (but not omega-agatoxin IVA) from its binding site in Purkinje neurons suggests that DW13.3 binds to a site close to the pore of the channel. The inhibition of different subtypes of calcium channels by DW13.3 reflects a common \"macro\" binding site present on all calcium channels except T-type.  相似文献

The characteristics of objective lenses and Ca2+-sensitive probes were examined for imaging with a two-photon laser-scanning microscope (TP-LSM). The brightness of the images of beads taken by different objectives greatly varied and depended predominantly on their numerical aperture (NA) and less on transmittance and chirping effects. Lateral and axial resolutions, dx and dz, defined as the half decay length of fluorescence intensity of the image of a spherical bead (0.3 m) were 0.12 and 0.42 microm (objective; 40x/0.75). They are far better than those of confocal microscopes (0.3 and 1.5 microm, respectively) measured similarly (Kuba et al., 1994). dx linearly increased with an increase in 1/NA, while dz linearly increased with an increase in n/(NA)2 (n, refractive index) except for an objective of large NA (1.3). The coverslip compensation of objective lenses greatly affected the shape of the X-Z scanned images of 5.0 microm beads as well as resolutions, indicating a large effect of spherical aberration. Two-photon excitation spectra of Ca2+-sensitive fluorescent probes, indo-1, fura-2 and Oregon Green BAPTA-1, lied in a wavelength range shorter than twice that activated by one-photon absorption, while emission spectra were unchanged. Three-dimensional images of a cultured hippocampal neurone loaded with Oregon Green BAPTA-1 showed fine structures of spines, dendrites and axons, while imaging with FM1-43 localized presynaptic boutons and demonstrated synaptic vesicle turnover. Dyes bleached little during the recording of 100 sectioned images. These characteristics of TP-LSM as well as its ability to image deeper tissues provide excellent means to study dynamic, spatial changes in intracellular substances and structures. To achieve the good performance of a TP-LSM, however, the relevant usage of appropriate objectives and fluorescent probes are required.  相似文献

The feasibility of intragenerically characterizing bifidobacteria by a comparison of a short region within the recA gene was tested. An approximately 300 bp fragment of the recA gene was PCR-amplified from six species from the genus Bifidobacterium using primers directed to two universally conserved regions of the recA gene. A phylogenetic analysis of the sequenced recA products compared favorably to classification based on the 16S rRNA sequences of the species tested. To apply this rapid methodology to unknown human intestinal bifidobacteria, 46 isolates were randomly chosen from the feces of four subjects and initially characterized by RFLP analysis of a PCR-amplified region of their 16S RNA genes. From a representative of the dominant RFLP family in each of the subjects, the recA segment was PCR-amplified, sequenced and phylogenetically analyzed. All four isolates were found to be related to one another and to B. longum and B. infantis. These results illustrate that the recA gene may be useful for intrageneric phylogenetic analysis as well as for the identification of unknown fecal bifidobacteria.  相似文献

The recently published ‘FE–Meshfree’ QUAD4 element is extended to geometrical non‐linear analysis. The shape functions for this element are obtained by combining meshfree and finite element shape functions. The concept of partition of unity (PU) is employed for the purpose. The new shape functions inherit their higher order completeness properties from the meshfree shape functions and the mesh‐distortion tolerant compatibility properties from the finite element (FE) shape functions. Updated Lagrangian formulation is adopted for the non‐linear solution. Several numerical example problems are solved and the performance of the element is compared with that of the well‐known Q4, QM6 and Q8 elements. The results show that, for regular meshes, the performance of the element is comparable to that of QM6 and Q8 elements, and superior to that of Q4 element. For distorted meshes, the present element has better mesh‐distortion tolerance than Q4, QM6 and Q8 elements. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

A highly vascular petroclival meningioma supplied by tentorial branches of the internal carotid artery was embolized by temporary balloon occlusion of the parent vessel distal to the tumor, followed by obliteration of the tumor vascularity with polyvinyl alcohol particles. Subsequently, in vivo proton spectroscopy showed necrosis of a large portion of the tumor and helped determine the timing of surgery. Both innovative techniques considerably facilitated the subsequent radical excision of the tumor with no neurological morbidity.  相似文献

Polymer blend electrolytes comprising of poly(vinyl acetate), poly(vinylidene fluoride-co-hexafluoropropylene), LiClO4, and EC-based plasticizer combinations (EC + PC, EC + GBL, EC + DMP, EC + DBP, EC + DEC) are prepared by solvent casting technique. Ionic conductivities of the electrolytes are determined by ac impedance studies in the temperature range 303–363 K. Among the various combinations of plasticizers, EC + PC added complex exhibits maximum ionic conductivity of the order of 10−4 S cm−1 and the temperature-dependent ionic conductivity plots seem to obey the VTF relation. The structural and complex formations of the prepared samples have been confirmed by X-ray diffraction analysis. DSC technique is used to study the thermal behaviour. The surface images of the sample having maximum ionic conductivity are analyzed with the help of SEM and AFM techniques.  相似文献
