Electrostatic attraction between α and β globin chains holds the subunits together in a tetrameric human hemoglobin molecule (α2β2). Compared to normal globin chains, the affinity of a mutant chain to its partner globin might be different in genetic variants of hemoglobin. This leads to an unequal abundance of normal and variant hemoglobin in heterozygous samples, even though the rates of synthesis of both the normal and variant chains are the same. The aforementioned affinities across various globin chains might be assessed by quantification of the different forms of the tetramers present in a variant hemoglobin sample. In the present study, by exploiting mass differences between globin chains, differently populated hemoglobin tetramers present in hemoglobin (Hb) Beckman, a β variant (βA135D), were structurally characterized. The relative populations of dissymmetric tetramers (α2β2, α2ββV, and α2βV2) indicated that both β and βV have different affinities towards the α globin chain. Conformational dynamics analyzed from hydrogen/deuterium exchange kinetics of the three peptide fragments of Hb Beckman in its oxy state displayed molecular insight into its functional abnormality. However, in comparison to normal hemoglobin (α2β2), the point mutation did not show any change in the collision cross‐sections of the functionally active conformers of the variant hemoglobin molecules (α2ββV and α2βV2).  相似文献

Cationic surfactant cetyltrimethylammonium bromide (CTAB) and cetylpyridinium bromide (CPB) bind to the calf thymus DNA (ct‐DNA) like anionic biopolymers electrostatically, and establish equilibrium in aqueous medium at pH 7. At low concentration, ct‐DNA does not interact with anionic surfactants, sodium dodecylsulfate (SDS) and sodium dodecylbenzylsulfonate (SDBS). However, in the ground state, anionic surfactant is found to clearly establish equilibrium with ct‐DNA‐bound cationic surfactant whereby the same surfactant–DNA isosbestic point reappears. We herein report a detail ratiometric binding of CPB with ct‐DNA, and interaction of anionic SDBS with DNA‐bound CPB in comparison with the combined ct‐DNA–CTAB–SDS system. Compaction of ct‐DNA in presence of CPB and its decompaction using anionic SDBS is also studied in comparison with CTAB–SDS combination. The techniques used are tensiometry, spectrophotometry, viscometry, cyclic voltammetry, circular dichroism, isothermal titration calorimetry, and density functional theory (DFT)‐based computational calculations. The size and surface charge density of the surfactant headgroups and the phosphate group in DNA have a contributing role in the DNA compaction–decompaction phenomenon.  相似文献

BACKGROUND: Low energy and less expensive membrane based separation of acetic acid‐water mixtures would be a better alternative to conventional separation processes. However, suitable acid resistant membranes are still lacking. Thus, the objective of the present study was to develop mixed matrix membrane (MMM) which would allow high flux and water selectivity over a wide range of feed concentrations of acid in water. RESULTS: Three MMMs, namely PANBA0.5, PANBA1.5 and PANBA3 were made by emulsion copolymerization of acrylonitrile (AN) and butyl acrylate (BA) with 5.5:1 comonomer ratio and in situ incorporation of 0.5, 1.5 and 3 wt%, sodium montmorilonite (Na‐MMT) nanofillers, respectively. For a feed concentration of 99.5 wt% of acid in water the membranes show good permeation flux (2.61, 3.19, 3.97 kg m?2 h?1 µm?1, for PANBA0.5, PANBA1.5 and PANBA3 membrane, respectively) and very high separation factors for water (1473, 1370, 1292 for PANBA0.5, PANBA1.5 and PANBA3 membrane, respectively) at 30 °C. Similarly for a dilute acid–water solution, i.e. for 71.6 wt% acid the membrane showed a very high thickness normalize flux (8.67, 9.44, 11.56 kg m?2 h?1 µm?1, for PANBA0.5, PANBA1.5 and PANBA3 membrane, respectively) and good water selectivity (101.7, 95.3, 79 for PANBA0.5, PANBA1.5 and PANBA3 membrane, respectively) at the same feed temperature. The permeation ratio, permeability, diffusion coefficient and activation energy for permeation of the membranes were also estimated. CONCLUSION: Unlike most of the reported membranes, the present MMMs allowed high flux and selectivity over a wide range of feed concentrations. Castor oil has gained momentous attention as a valuable bio-based monomer and a potential alternative to the current petrobased polyol for synthesizing polyurethane due to the presence of inherent hydroxyl group. In spite of its huge potentiality very little has been reviewed regarding the development of polyurethane from castor oil. This review thus highlights the recent trends and development in the field of polyurethane and its nanocomposite based on castor oil including its biodegradability and weatherability studies. Further, this review also provides an insight regarding the utilization of castor oil based polyurethane and its nanocomposite for coating application.  相似文献

In the present study, we report a simple method to synthesize silver (Ag)‐polypyrrole (PPy)/graphene (Gr) nanocomposite as efficient electrode materials for supercapacitor application. The probable interaction between Ag nanoparticles with both PPy and Gr were characterized by FTIR, UV–visible, and Raman spectroscopies. The morphological analysis confirmed that the Gr sheets are uniformly coated by PPy and in the coated Gr sheets there is the presence of Ag nanoparticles. The Ag‐PPy/Gr nanocomposite achieved the highest specific capacitance of 472 F/g at a 0.5 A/g current density. Better energy and power density also obtained for the nanocomposite. The presence of both Ag nanoparticles and Gr is the main reason for the enhancement of the electrochemical properties of the nanocomposite. Based on the superior electrochemical properties, the nanocomposite can be used for next‐generation supercapacitor electrode material. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. Dielectric elastomer actuators (DEAs) have been studied widely in recent years for artificial muscle applications, but their implementation into production is limited due to high operating voltages required. The actuation behavior of dielectric elastomer under an applied electric field is predicted by Maxwell's pressure and thickness strain equations. According to these equations, the best electromechanical response is achieved when the relative permittivity is high and elastic modulus is low. The potential source for additives increasing the relative permittivity of rubbers can be vegetable powders that have much higher dielectric constant than common elastomers. In the present research, the dielectric and actuation properties of polyacrylate rubber (ACM) were studied after the addition of different vegetable‐based fillers such as potato starch, corn starch, garlic, and paprika. The results were compared to ACM filled with barium titanate. The compounds containing vegetable fillers showed higher relative dielectric permittivity at 1 Hz frequency than the compounds containing barium titanate due to higher interfacial polarization. The actuation studies showed that lower electric fields are required to generate certain actuation forces when the starches and garlic are used in the rubber instead of barium titanate. The present report studies on the flow pattern transitions during vertical air water downflow through millichannels (0.83 ≤ Eötvös no. ≤ 20.63). Four basic flow patterns namely falling film flow, slug flow, bubbly flow, and annular flow are observed in the range of experimental conditions studied and their range of existence has been noted to vary with tube diameter and phase velocities. Based on experimental observations, phenomenological models are proposed to predict the transition boundaries between adjacent patterns. These have been validated with experimental flow pattern maps from the present experiments. In the present article a facile synthesis of cerium oxide nanoparticles (CNPs) encapsulated in PLGA microparticles is reported. The release kinetics of the CNPs from the PLGA matrix was investigated under acidic, basic and near-neutral pH. A diffusion model was applied to determine the diffusivity of the CNPs from the PLGA matrix. The morphology of the degraded PLGA particles was characterized by high resolution SEM. Superoxide dismutase (SOD) mimetic activity was retained in released CNPs for a longer period of time (~90 days) under different pH. PLGA encapsulated CNP showed excellent biocompatibility. This study demonstrates a potential strategy to deliver CNPs using biodegradable PLGA that ensures a slow release of the CNPs over a long period of time. Thus, the synthesized PLGA encapsulated CNPs could find potential applications in tissue engineering like bone remodelling and regeneration, and protection from disorders caused by neurodegeneration.  相似文献

Multiwalled carbon nanotube (MWCNT)/alumina (Al2O3) nanocomposites containing CNT from 0.15 vol.% to 2.4 vol.% have been successfully fabricated by simple wet mixing of as-received commercial precursors followed by pressureless sintering. Extent of densification of nanocomposites sintered at low temperature (e.g. 1500 °C) was <90%, but increased up to ∼99% when sintered at 1700 °C and offered superior performance compared to pure Al2O3. Nanocomposites containing 0.3 vol.% MWCNT and sintered at 1700 °C for 2 h in Argon led to ∼23% and ∼34% improvement in hardness and fracture toughness, respectively, than monolithic Al2O3. In addition, the highest improvement (∼20%) in bending strength was obtained for 0.15 vol.% MWCNT/Al2O3 nanocomposite compared to pure Al2O3. Weibull analysis indicated reliability of nanocomposites increased up to 0.3 vol.% MWCNT, whereas, beyond that loading consistency was the same as obtained for pure Al2O3. Detailed microstructure and fractographic analysis were performed to assess structure-property relationship of present nanocomposites.  相似文献
