Molecular analyses were carried out on four preflight and six postflight International Space Station (ISS)-associated potable water samples at various stages of purification, storage, and transport, to ascertain their associated microbial diversities and overall microbial burdens. Following DNA extraction, PCR amplification, and molecular cloning procedures, rDNA sequences closely related to pathogenic species of Acidovorax, Afipia, Brevundimonas, Propionibacterium, Serratia, and others were recovered in varying abundance. Retrieval of sequences arising from the iodine (biocide)-reducing Delftia acidovorans in postflight waters is also of concern. Total microbial burdens of ISS potable waters were derived from data generated by an ATP-based enumeration procedure, with results ranging from 0 to 4.9 x 10(4) cells/ml. Regardless of innate biases in sample collection and analysis, such circumstantial evidence for the presence of viable, intact pathogenic cells should not be taken lightly. Implementation of new cultivation approaches and/or viability-based assays are requisite to confirm such an occurrence.  相似文献

Optical designs often specify both surface form and centering (tilt and lateral displacement) tolerances on aspheric surfaces. In contrast to spherical surfaces, form and centering errors are coupled for aspheric surfaces. Current standards do not specify how to interpret such tolerances, and in particular they do not define the position of an aspheric surface that has form errors. The straightforward definition that uses the best-fit surface position that minimizes rms error has subtle problems. The best-fit surface position for aspheric surfaces is influenced by power error and can be highly sensitive to surface form errors when the derivative of aspheric departure is small. We analyze the conditions under which form and centering tolerances may be considered compatible when the best-fit surface-position definition is used. We propose alternative definitions of surface position that do not suffer from the same problems and consider their consequences for optical design and metrology.  相似文献

Laser ablation has been used ex situ to create metal nanoparticles for introduction into a reactive pyrolysis flame. By prior synthesis of the metal nanoparticles, the effects of the reactive gases can be clearly separated from the pyrolysis chemistry of a solvent carrier, as when nebulized solutions are used. Moreover, varying reactivity issues associated with particle growth and size are bypassed. Our results show that Fe selectively reacts with CO to produce nanotubes, whereas Ni selectively reacts with C2H2 to produce nanofibers. These observations are interpreted through the donation and withdrawal of electron density between the adsorbate's molecular orbitals and surface atoms of the metal nanoparticle. The rate of reaction of Ni with only C2H2 is found to be greater than the rate with C2H2 and CO. This suggests that CO inhibits the Ni-catalyzed reaction.  相似文献

The electrical measurements required during the primary calibrations of laboratory standard microphones by the reciprocity method can be influenced by power line interference. Because of this influence, the protocols of international inter-laboratory key comparisons of microphone calibrations usually have not included measurements at power line frequencies. Such interference has been observed in microphone output voltage measurements made with a microphone pressure reciprocity calibration system under development at NIST. This system was configured for a particular type of standard microphone in such a way that measurements of relatively small signal levels, which are more susceptible to the effect of power line interference, were required. This effect was investigated by acquiring microphone output voltage measurement data with the power line frequency adjusted to move the frequency of the interference relative to the center frequency of the measurement system passband. These data showed that the effect of power line interference for this system configuration can be more than one percent at test frequencies harmonically related to the power line frequency. These data also showed that adjusting the power line frequency to separate the interference and test frequencies by as little as 1.0 Hz can reduce the effect of the interference by at least an order of magnitude. Adjustment of the power line frequency could enable accurate measurements at test frequencies that otherwise might be avoided.  相似文献

A new method for imaging the vibration mode of an object is investigated. The radiation force of ultrasound is used to scan the object at a resonant frequency of the object. The vibration of the object is measured by laser and the resulting acoustic emission from the object is measured by a hydrophone. It is shown that the measured signal is proportional to the value of the mode shape at the focal point of the ultrasound beam. Experimental studies are carried out on a mechanical heart valve and arterial phantoms. The mode images on the valve are made by the hydrophone measurement and confirmed by finite-element method simulations. Compared with conventional B-scan imaging on arterial phantoms, the mode imaging can show not only the interface of the artery and the gelatin, but also the vibration modes of the artery. The images taken on the phantom surface suggest that an image of an interior artery can be made by vibration measurements on the surface of the body. However, the image of the artery can be improved if the vibration of the artery is measured directly. Imaging of the structure in the gelatin or tissue can be enhanced by small bubbles and contrast agents.  相似文献

Electro-optic (EO) modulation devices, which utilize an external electric field to modulate a beam of optical radiation, are strongly affected by parasitic effects, which change the polarization state of the optical beam. As a result, very small changes in the birefringence or optical path length within the EO material can result in very large fluctuations of the amplitude and phase of the optical modulation signal. A method of actively analyzing the modulated beam is described and demonstrated, which eliminates these fluctuations and keeps the modulation device stably operating at its peak responsivity. Applications to electric field detection and measurements are discussed.  相似文献

We report the first in vitro enzymatic synthesis of paramagnetic and antiferromagnetic nanoparticles toward magnetic ELISA reporting. With our procedure, alkaline phosphatase catalyzes the dephosphorylation of l-ascorbic-2-phosphate, which then serves as a reducing agent for salts of iron, gadolinium, and holmium, forming magnetic precipitates of Fe45±14Gd5±2O50±15 and Fe42±4Ho6±4O52±5. The nanoparticles were found to be paramagnetic at 300 K and antiferromagnetic under 25 K. Although weakly magnetic at 300 K, the room-temperature magnetization of the nanoparticles found here is considerably greater than that of analogous chemically-synthesized LnxFeyOz (Ln = Gd, Ho) samples reported previously. At 5 K, the nanoparticles showed a significantly higher saturation magnetization of 45 and 30 emu/g for Fe45±14Gd5±2O50±15 and Fe42±4Ho6±4O52±5, respectively. Our approach of enzymatically synthesizing magnetic labels reduces the cost and avoids diffusional mass-transfer limitations associated with pre-synthesized magnetic reporter particles, while retaining the advantages of magnetic sensing.  相似文献
