An electrochemical genosensor for the detection of nucleic acid sequences specific of Legionella pneumophila is reported. An immobilized thiolated hairpin probe is combined with a sandwich-type hybridization assay, using biotin as a tracer in the signaling probe, and streptavidin-alkaline phosphatase as reporter molecule. The activity of the immobilized enzyme was voltammetrically determined by measuring the amount of 1-naphthol generated after 2 min of enzymatic dephosphorylation of 1-naphthyl phosphate. The sensor allows discrimination between L. pneumophila and L. longbeachae with high sensitivity under identical assay conditions (no changes in stringency). A limit of detection of 340 pM L. pneumophila DNA, and a linear relationship between the analytical signal and the logarithm of the target concentration to 2 muM were obtained. Experimental results show the superior sensitivity and selectivity of the hairpin-based assay when compared with analogous sandwich-type assays using linear capture probes.  相似文献

Single wall carbon nanotube (SWCNT) architecture when employed as conducting scaffolds in a TiO2 semiconductor based photoelectrochemical cell can boost the photoconversion efficiency by a factor of 2. Titanium dioxide nanoparticles were dispersed on SWCNT films to improve photoinduced charge separation and transport of carriers to the collecting electrode surface. The shift of approximately 100 mV in apparent Fermi level of the SWCNT-TiO2 system as compared to the unsupported TiO2 system indicates the Fermi level equilibration between the two systems. The interplay between the TiO2 and SWCNT of attaining charge equilibration is an important factor for improving photoelectrochemical performance of nanostructured semiconductor based solar cells. The feasibility of employing a SWCNT-TiO2 composite to drive the water photoelectrolysis reaction has also been explored.  相似文献

Microtubule targeting agents (MTAs) have been exploited mainly as anti-cancer drugs because of their impact on cellular division and angiogenesis. Additionally, microtubules (MTs) are key structures for intracellular transport, which is frequently hijacked during viral infection. We have analyzed the antiviral activity of clinically used MTAs in the infection of DNA and RNA viruses, including SARS-CoV-2, to find that MT destabilizer agents show a higher impact than stabilizers in the viral infections tested, and FDA-approved anti-helminthic benzimidazoles were among the most active compounds. In order to understand the reasons for the observed antiviral activity, we studied the impact of these compounds in motor proteins-mediated intracellular transport. To do so, we used labeled peptide tools, finding that clinically available MTAs impaired the movement linked to MT motors in living cells. However, their effect on viral infection lacked a clear correlation to their effect in motor-mediated transport, denoting the complex use of the cytoskeleton by viruses. Finally, we further delved into the molecular mechanism of action of Mebendazole by combining biochemical and structural studies to obtain crystallographic high-resolution information of the Mebendazole-tubulin complex, which provided insights into the mechanisms of differential toxicity between helminths and mammalians.  相似文献

Experience has proved that interactive applications delivered through Digital TV must provide personalized information to the viewers in order to be perceived as a valuable service. Due to the limited computational power of DTV receivers (either domestic set-top boxes or mobile devices), most of the existing systems have opted to place the personalization engines in dedicated servers, assuming that a return channel is always available for bidirectional communication. However, in a domain where most of the information is transmitted through broadcast, there are still many cases of intermittent, sporadic or null access to a return channel. In such situations, it is impossible for the servers to learn who is watching TV at the moment, and so the personalization features become unavailable. To solve this problem without sacrificing much personalization quality, this paper introduces solutions to run a downsized semantic reasoning process in the DTV receivers, supported by a pre-selection of material driven by audience stereotypes in the head-end. Evaluation results are presented to prove the feasibility of this approach, and also to assess the quality it achieves in comparison with previous ones.

Pazos-Arias   received the Ph.D. degree in Computer Science from the Madrid University of Technology (UPM) in 1995, and worked with Alcatel Laboratories in Madrid prior to joining the University of Vigo. He is the founder and director of the Networking & Software Engineering Group, which is currently involved with several projects related to DTV middleware and applications. Jorge García-Duque   received the Ph.D. degree in Computer Science from the University of Vigo in 2000. His research is focused on the deployment of information services over heterogeneous networks of consumer devices. Manuel Ramos-Cabrer   received the Ph.D. degree in Computer Science from the University of Vigo in 2000. His research interests include the application of artificial intelligence techniques to personalization systems. Alberto Gil-Solla   received the Ph.D. degree in Computer Science from the University of Vigo in 2000. His research is currently involved with different aspects of middleware design and interactive multimedia services. Rebeca P. Díaz-Redondo   received the Ph.D. degree in Computer Science from the University of Vigo in 2002. Her research is now focused on interactive DTV applications playing a central role in the control of smart home environments. Ana Fernández-Vilas   received the Ph.D. degree in Computer Science from the University of Vigo in 2002. Uveal melanoma (UM) is a malignant tumor that arises in the melanocytes of the uveal tract. It is the most frequent eye cancer, and despite new therapeutic approaches, prognosis is still poor, with up to 50% of patients developing metastasis with no efficient treatment options available. In contrast to cutaneous melanoma, UM is considered an “immune-cold” tumor due to the low mutational burden and the unique immunosuppressive microenvironment. To gain insight into the role of the UM microenvironment in regard to prognosis and metastatic progression, we have performed a pool analysis characterizing the UM microenvironment by using a bioinformatic approach. A variety of scores based on gene expression measuring stromal infiltration were calculated and used to assess association with prognosis. As a result, the highest immune and stromal scores were associated with poor prognosis. Specifically, stromal cells (fibroblasts and endothelial cells), T cells CD8+, natural killer (NK) cells, and macrophages M1 and M2 infiltration were associated with poor prognosis. Contrary to other tumors, lymphocytic infiltration is related to poor prognosis. Only B cells were associated with more favorable prognosis. UM samples scoring high in both angiogenesis (Angio) and antigen presentation (AP) pathways showed a poor prognosis suggesting an additive role of both functions. Almost all these tumors exhibited a chromosome 3 monosomy. Finally, an enrichment analysis showed that tumors classified as high Angio-high AP also activated metabolic pathways such as glycolysis or PI3K-AKT-MTOR. In summary, our pool analysis identified a cluster of samples with angiogenic and inflammatory phenotypes exhibiting poor prognosis and metabolic activation. Our analysis showed robust results replicated in a pool analysis merging different datasets from different analytic platforms.  相似文献

The Transient Receptor Vanilloid 1 (TRPV1) or capsaicin receptor is a nonselective cation channel, which is abundantly expressed in nociceptors. This channel is an important transducer of several noxious stimuli, having a pivotal role in pain development. Several TRPV1 studies have focused on understanding its structure and function, as well as on the identification of compounds that regulate its activity. The intracellular roles of these channels have also been explored, highlighting TRPV1′s actions in the homeostasis of Ca2+ in organelles such as the mitochondria. These studies have evidenced how the activation of TRPV1 affects mitochondrial functions and how this organelle can regulate TRPV1-mediated nociception. The close relationship between this channel and mitochondria has been determined in neuronal and non-neuronal cells, demonstrating that TRPV1 activation strongly impacts on cell physiology. This review focuses on describing experimental evidence showing that TRPV1 influences mitochondrial function.  相似文献
