A comparison between a direct immunofluorescence assay (DFA) and the shell-vial culture (SVC) was conducted to evaluate their efficacies according to the quality and origin of the sample and the type of herpes simplex (HSV) responsible for the infection. The SVC detected all 58 HSV-infected samples, while the DFA detected only 49 (84.5%) positive samples. The DFA detected HSV type 1 in 22 of 89 samples (24.7%) and HSV type 2 in 27 of 96 samples (28.1%). Compared with the SVC, the DFA had a sensitivity of 75.8% for HSV type 1 and 93.1% for HSV type 2. The sensitivity of the DFA depends on the quality of the sample. Thus, while DFA is recommendable as a screening method, the SVC remains the method of choice for obtaining the maximum diagnostic yield from the sample.  相似文献

We examined the differentiation and resorptive function of osteoclasts (OC) cultured on the slices of calcified bone, decalcified bone and hyaline cartilage, and found that OC differentiation depends on the co-cultured substratum, as well as osteoblast-derived factors. Bone marrow-derived macrophages (BMM) were formed from marrow cells of 5 week old ddY mice and cultured for 3 days on freeze-dried slices of calcified bone, decalcified bone or cartilage, all prepared from rabbit costal bone. BMM cultured on calcified bone slices exhibited tartrate-resistant acid phosphatase (TRAP) activity and were structurally characterized by multinucleation and ruffled border development. However, on decalcified bone slices, BMM seldom became multinucleated and exhibited weak TRAP activity. BMM cultured on cartilage slices were mononuclear, devoid of TRAP activity and structurally resembled mononuclear phagocytes. In SEM observations of co-cultured slices, resorption lacunae were formed only on calcified bone slices, and not on slices of decalcified bone and cartilage. Our results, therefore, indicated that BMM could differentiate into functional OC only on calcified bone slices, suggesting a key role of calcified components in the bone matrix for the terminal OC differentiation. Then, we cultured BMM on the same slices with yeast particles. In cultures with yeast particles, BMM exhibited intense TRAP activity, developed a ruffled border-like structure and formed resorption lacunae even on decalcified bone and cartilage slices. Vacuolar-type H+-ATPase was strongly expressed along the ruffled border membranes of these OC. Only the BMM that had not incorporated yeast particles developed a ruffled border, whereas the BMM that had incorporated yeast particles did not become multinucleated and lacked a ruffled border structure. Thus, our results further suggest that, even on uncalcified substrata, the terminal differentiation of BMM into functional OC is induced by an unidentified external stimulus, which may be contained in the cell membrane of yeast particles.  相似文献

Most experts agree that it is too early to say how quantum computers will eventually be built, and several nanoscale solid-state schemes are being implemented in a range of materials. Nanofabricated quantum dots can be made in designer configurations, with established technology for controlling interactions and for reading out results. Epitaxial quantum dots can be grown in vertical arrays in semiconductors, and ultrafast optical techniques are available for controlling and measuring their excitations. Single-walled carbon nanotubes can be used for molecular self-assembly of endohedral fullerenes, which can embody quantum information in the electron spin. The challenges of individual addressing in such tiny structures could rapidly become intractable with increasing numbers of qubits, but these schemes are amenable to global addressing methods for computation.  相似文献

Graphene and other 2D materials, such as molybdenum disulfide, have been increasingly used in electronics, composites, and biomedicine. In particular, MoS2 and graphene hybrids have attracted a great interest for applications in the biomedical research, therefore stimulating a pertinent investigation on their safety in immune cells like macrophages, which commonly engulf these materials. In this study, M1 and M2 macrophage viability and activation are mainly found to be unaffected by few‐layer graphene (FLG) and MoS2 at doses up to 50 µg mL?1. The uptake of both materials is confirmed by transmission electron microscopy, inductively coupled plasma mass spectrometry, and inductively coupled plasma atomic emission spectroscopy. Notably, both 2D materials increase the secretion of inflammatory cytokines in M1 macrophages. At the highest dose, FLG decreases CD206 expression while MoS2 decreases CD80 expression. CathB and CathL gene expressions are dose‐dependently increased by both materials. Despite a minimal impact on the autophagic pathway, FLG is found to increase the expression of Atg5 and autophagic flux, as observed by Western blotting of LC3‐II, in M1 macrophages. Overall, FLG and MoS2 are of little toxicity in human macrophages even though they are found to trigger cell stress and inflammatory responses.  相似文献

Pasteurized whole, 2%, and skim milk were inoculated with Listeria monocytogenes Scott A and treated with high-voltage pulsed electric field (PEF). The effects of milk composition (fat content) and PEF parameters (electric field strength, treatment time, and treatment temperature) on the inactivation of the bacterium were studied. No significant differences were observed in the inactivation of L. monocytogenes Scott A in three types of milk by PEF treatment. With treatment at 25 degrees C, 1- to 3-log reductions of L. monocytogenes were observed. PEF lethal effect was a function of field strength and treatment time. Higher field strength or longer treatment time resulted in a greater reduction of viable cells. A 4-log reduction of the bacterium was obtained by increasing the treatment temperature to 50 degrees C. Results indicate that the use of a high-voltage PEF is a promising technology for inactivation of foodborne pathogens.  相似文献

This paper presents a new method for accurately estimating the pose (position and orientation) of a mobile robot by registering a segment-based local map observed from the current robot pose and a global map. The method works in a two-stage procedure. First, the orientation is determined by aligning the local and global map through a voting process based on a generalized Hough transform. Second, it uses a coarse-to-fine approach for selecting candidate positions and a weighted voting scheme to determine the degree of overlap of the two maps at each of these poses. Unlike other methods previously proposed, this approach allows us to uncouple the problem of estimating the robot orientation and the robot position which may be useful for some applications. In addition it can manage environments described by many (possibly short) segments. This paper presents some experimental results based on our mobile robot RAM-2 that show the accuracy and the robustness of the proposed method even for poor quality maps and large dead-reckoning errors.  相似文献

Scene registration of a pair of three-dimensional (3D) range images is a 6D optimization problem usually required in mobile robotics. This problem is different from object registration, since all scan directions and depths may contain relevant data, and because farther regions are sampled with lower densities. The paper proposes an efficient scene matching method based on the concept of coarse binary cubes. An integer objective function is defined as the number of coincident cubes between both scans. This is a metric of the degree of overlap that does not employ point distances. Its value is obtained without actually using any 3D grid data structure, with a computational complexity of order O(n), where n represents the number of laser points. This objective function is optimized with a globalized version of the downhill Simplex algorithm to avoid local minima. Experimental results are presented from indoor and outdoor environments with different degrees of structuring. The effect of cube size and the number of vertices on registration performance has been analyzed. Besides, experiments show that the proposed method achieves similar accuracy as iterative closest points (ICP) and normal distribution transform (NDT), while it improves both computation time and robustness against initial misalignments.  相似文献

We prepared membranes based on a liquid-crystalline side-chain polyether obtained by chemical modification of commercial poly(epichlorohydrin) (PECH) with dendrons. This polymer exhibited a columnar structure, which could form an ion channel in the inner part. The columns were successfully oriented by taking advantage of surface interactions between the polymer and hydrophilic substrates, as confirmed by X-ray diffraction analysis (XRD), environmental scanning electron microscopy (ESEM) and optical microscopy between crossed polars (POM). Column orientation was found to be crucial for effective transport: the oriented membranes exhibited proton transport comparable to that of Nafion® N117 and no water uptake. An increase in sodium ion concentration in the feed phase suggested a proton/cation antiport. On the contrary, no proton transport was detected on unoriented membranes based on the same liquid-crystalline side-chain polyether or on unmodified PECH.  相似文献

The use of the solar photo-Fenton process is proposed to degrade Paracetamol in water in order to form biodegradable reaction intermediates which can be finally removed with a downstream biological treatment. Firstly, biodegradability enhancement with photo-Fenton treatment time has been evaluated; the minimum mineralization level should be at least 18.6% where Paracetamol has been degraded and biodegradability efficiency is higher than 40%. 20 mg L(-1) of Fe(2+) and 200 mg L(-1) of H(2)O(2) were selected in a lab-scale study looking at Paracetamol's degradation rate and organic carbon mineralization rate. As a result of scaling up the process at a pilot plant, 157.5 mg L(-1) of Paracetamol (~1 mM) was treated in 25 min of photo-Fenton treatment achieving the desired biodegradability. A further economic evaluation shows how the proposed treatment strategy markedly increases plant efficiency, resulting in an 83.33% reduction in reagent cost and a 79.11% reduction in costs associated with reaction time. Total cost is reduced from 3.4502 ?m(3) to 0.7392 ?m(3).  相似文献

In reliability engineering and practice an important role is played by those units whose life characteristics change with time. The case is herewith considered where change underlies a nonhomogeneous Markov model. Simple repair processes then deal with a 2-state alternating policy resulting from the superposition of failure and restoration, both showing a continuous aging with time. Availability can be expressed through linear differential equations or by means of integral equations; the approaches are equivalent. A computer code is then described which calculates (i) availability, and (ii) transition densities for any continuous time dependence of failure and restoration rates.
