Urinary tract infections caused by uropathogenic E. coli are among the most prevalent infectious diseases. The mannose‐specific lectin FimH mediates the adhesion of the bacteria to the urothelium, thus enabling host cell invasion and recurrent infections. An attractive alternative to antibiotic treatment is the development of FimH antagonists that mimic the physiological ligand. A large variety of candidate drugs have been developed and characterized by means of in vitro studies and animal models. Here we present the X‐ray co‐crystal structures of FimH with members of four antagonist classes. In three of these cases no structural data had previously been available. We used NMR spectroscopy to characterize FimH–antagonist interactions further by chemical shift perturbation. The analysis allowed a clear determination of the conformation of the tyrosine gate motif that is crucial for the interaction with aglycone moieties and was not obvious from X‐ray structural data alone. Finally, ITC experiments provided insight into the thermodynamics of antagonist binding. In conjunction with the structural information from X‐ray and NMR experiments the results provide a mechanism for the often‐observed enthalpy–entropy compensation of FimH antagonists that plays a role in fine‐tuning of the interaction.  相似文献

The use of multiblock copolymers for the compatibilization of immiscible polymer blends is controversially discussed in the literature. Investigations have been carried out to estimate the effect of multiblock copolymers containing segments of a liquid crystalline polyester (LCP) and polysulfone (PSU) segments in blends of the based homopolymers. One goal was to determine whether multiblock copolymers provide an opportunity for compatibilizing PSU/LCP blends. By using PSU/LCP multiblock copolymers with different molecular weights of the blocks in the appropriate binary, solution-casted blends, it was shown that the interpenetration of the polysulfone phase of the block copolymer and the PSU matrix leads to an improved miscibility of the blend. This effect is retained in ternary blends of PSU, LCP, and the multiblock copolymer, assuming a certain critical molecular weight of the multiblock copolymer segments. In addition, some mechanical characteristics of PSU/LCP melt blends such as the E-modulus and fracture strength are improved by adding long-segmented multiblock copolymers. © 1997 John Wiley & Sons, Inc. Thermogravimetric analysis of a coccolith-containing biogenic broth showed a three-step degradation process. According to this system behavior, the biogenic broth was heated to specific temperatures and characterized in terms of its morphology, surface chemistry, and crystallinity. The elemental and organic composition of the treated samples was also evaluated and compared to the reference material. The presented results were acquired in an effort to exploit pretreatment scenarios for such a biogenic system that would improve and support a separation process.  相似文献

Weakly lignified biomass, like brewers' spent grain and grass silage, is good feedstock for biorefineries. The biomass can be separated and converted into valuable products in different ways. In this study, brewers' spent grain is used to investigate solid state fermentations with Cellulomonas uda. Additionally, hydrothermal pretreatment and subsequent enzymatic saccharification, as well as formation of inhibitory compounds, is demonstrated. Hydrothermal pretreatment combined with simultaneous saccharification and fermentation is shown as an example for the utilization of grass silage.  相似文献

Fracture behavior of binary blends comprising of styrene-butadiene block copolymers having star and triblock architectures was studied via instrumented Charpy impact test. The toughness of the ductile blends was characterized by dynamic crack resistance curves (R-curves).This study represents a systematic investigation of crack resistance behavior of nanometer structured binary block copolymer blends and the development of a new material with a combination of high toughness and transparency, usually not observed in incompatible polymer blends. While the lamellar star block copolymer shows an elastic behavior (small-scale yielding and unstable crack growth), adding of 20 wt% of the triblock copolymer leads to a stable crack growth and at 60 wt% of the triblock copolymer the strong increase of toughness values indicate a tough/high-impact transition, demonstrating the existence of novel toughening concepts for polymers based on nanometer structured materials.  相似文献

Cuticular hydrocarbons were extracted from workers of 63 different nests of five species of Tetramorium ants (Hymenoptera: Formicidae) from Austria, Hungary, and Spain. The GC-MS data were classified (data mining) by self-organizing maps (SOM). SOM neurons derived from primary neuron separation were subjected to hierarchical SOM (HSOM) and were grouped to neuron areas on the basis of vicinity in the hexagonal output grid. While primary neuron separation and HSOM resulted in classifications on a level more sensitive than species differences, neuron areas resulted in chemical phenotypes apparently of the order of species. These chemical phenotypes have implications for systematics: while the chemical phenotypes for T. ferox and T. moravicum correspond to morphological determination, in T. caespitum and T. impurum a total of six chemical phenotypes is found. Three hypotheses are discussed to explain this disparity between morphological and chemical classifications, including in particular the possibility of hybridization and the existence of cryptic species. Overall, the GC-MS profiles classified by SOM prove to be a practical alternative to morphological determination (T. ferox, T. moravicum) and indicate the need to revisit systematics (T. caespitum, T. impurum).  相似文献

Analysis of samples taken at the Gentilly 2 (Québec) CANDU-PHW (CANadian Deuterium Uranium - Pressurized Heavy Water) plant after chlorination and demineralization revealed the presence of all four trihalomethanes (THMs) (CHCl3, CHBrCl2, CHBr2Cl and CHBr3) and other unidentified halogenated volatile compounds. Among the THMs, chloroform was the major contaminant. A study of its thermal stability in water at different temperatures confirmed the degradation of the CHCl3 molecule according to the equation CHCl3 + H2O → CO + 3 HCl. The reaction follow a first order kinetics and has an activation energy of 100 kJ/mol. The estimated half-life is six seconds at 260°C, the maximum temperature of the steam-condensate cycle.  相似文献

Gas chromatography – mass spectrometry analyses of the headspace volatiles of Scots pine (Pinus sylvestris) induced by egg deposition of the sawfly Diprion pini were conducted. The odor blend of systemically oviposition-induced pine twigs, attractive for the eulophid egg parasitoid Chrysonotomyia ruforum, was compared to volatiles released by damaged pine twigs (control) that are not attractive for the parasitoid. The mechanical damage inflicted to the control twigs mimicked the damage by a sawfly female prior to egg deposition. The odor blend released by oviposition-induced pine twigs consisted of numerous mono- and sesquiterpenes, which all were also present in the headspace of the artificially damaged control twigs. A quantitative comparison of the volatiles from oviposition-induced twigs and controls revealed that only the amounts of (E)--farnesene were significantly higher in the volatile blend of the oviposition-induced twigs. Volatiles from pine twigs treated with jasmonic acid (JA) also attract the egg parasitoid. No qualitative differences were detected when comparing the composition of the headspace of JA-treated pine twigs with the volatile blend of untreated control twigs. JA-treated pine twigs released significantly higher amounts of (E)--farnesene. However, the JA treatment induced a significant increase of the amount of further terpenoid components. The release of terpenoids by pine after wounding, egg deposition, and JA treatment is discussed with special respect to (E)--farnesene.  相似文献
