CD22 (Siglec-2) is a B-cell surface inhibitory protein capable of selectively recognising sialylated glycans, thus dampening autoimmune responses against self-antigens. Here we have characterised the dynamic recognition of complex-type N-glycans by human CD22 by means of orthogonal approaches including NMR spectroscopy, computational methods and biophysical assays. We provide new molecular insights into the binding mode of sialoglycans in complex with h-CD22, highlighting the role of the sialic acid galactose moieties in the recognition process, elucidating the conformational behaviour of complex-type N-glycans bound to Siglec-2 and dissecting the formation of CD22 homo-oligomers on the B-cell surface. Our results could enable the development of additional therapeutics capable of modulating the activity of h-CD22 in autoimmune diseases and malignancies derived from B-cells.  相似文献

In this study, the behavior of carbon steel and galvanized steel in nontropical coastal marine environments was evaluated. Evaluation was carried out with specimens with dimensions of 10 cm × 10 cm × 0.3 cm. These specimens were exposed to four testing stations (Iquique, Mejillones, Los Vilos, and San Vicente), where racks were installed both at ground level (ground), as well as in the upper zone of electrical transmission towers (tower). In each station, 24 specimens of A36 carbon steel and galvanized steel were placed (12 each). The corrosivity of the environment was measured using the ISO 9223, 9225, and 9226 standards. The specimens were evaluated on-site, monthly, through visual inspection and photographic record. Once withdrawn, the corrosion rate was determined and the corrosion products were analyzed through Raman and Fourier-transform infrared. The results show that, in all cases, the corrosion rate is greater in the tower than on the ground. However, even though the Los Vilos station is located farther from the sea (3,500 vs. ≈500 m), the corrosion rate of steel in the tower is the highest. This is caused by the generation of HCl from the transformation of lepidocrocite into goethite, in the presence of low chloride content, which acidifies the steel/corrosion product interface. In the case of galvanized steel, the corrosion rate is a function of the chloride content in the atmosphere, obtaining an excellent correlation between both parameters.  相似文献

The authenticity and traceability of meat products are issues of primary importance to ensure food safety. Unfortunately, food adulteration (e.g. the addition of inexpensive cuts to minced meat products) and mislabelling (e.g. the inclusion of meat from species other than those declared) happens frequently worldwide. The aim of this study was to apply a droplet digital PCR assay for the detection and quantification (copies μL−1) of the beef, pork, horse, sheep, chicken and turkey in meat products. The analysis conducted on commercial meat showed the presence of traces of DNA from other animal species than those declared. We show that the method is highly sensitive, specific and accurate (accuracy = 100%). This method could be adopted by competent food safety authorities to verify compliance with the labelling of meat products and to ensure quality and safety throughout the meat supply chain, from primary production to consumption.  相似文献

Thermally sprayed coatings are often used to mitigate corrosion of offshore structures. They act as a physical barrier to the aggressive marine environment and as a sacrificial distributed anode for low carbon steel. In such environments, the severity of material degradation depends on many factors. The effect of temperature, exposure time or the presence of micro‐organisms are the focus of many studies, for example, however, the effect of the different ions present in seawater remains largely unexplored. The chemical composition of the water changes considerably depending on the location; industrial, glacial, estuarine, and so forth. In addition, when thermal spray aluminum (TSA) protects steel in seawater, calcareous matter precipitates as a result of the cathodic polarization and subsequent localized increase in pH. Therefore, understanding how ions such as magnesium (II), calcium (II), or carbonates alter the coating properties in the marine environment is important. This paper reports the experimental work carried out with TSA‐coated steel samples with defects to simulate mechanical damage or erosion of the coating. The combination of electrochemical tests and surface characterization provided evidence of the efficiency of the calcareous bilayer that forms on top of steel reducing the TSA degradation.  相似文献
