Chronic neuropathic pain emerges from either central or peripheral lesions inducing spontaneous or amplified responses to non-noxious stimuli. Despite different pharmacological approaches to treat such a chronic disease, neuropathic pain still represents an unmet clinical need, due to long-term therapeutic regimens and severe side effects that limit application of currently available drugs. A critical phenomenon involved in central sensitization is the exchange of signalling molecules and cytokines, between glia and neurons, driving the chronicization process. Herein, using a chronic constriction injury (CCI) model of neuropathic pain, we evaluated the efficacy of the mu (M-) and delta (D-) opioid receptor (-OR) targeting agent LP2 in modulating connexin-based heterocellular coupling and cytokine levels. We found that long-term efficacy of LP2 is consequent to MOR-DOR targeting resulting in the reduction of CCI-induced astrocyte-to-microglia heterocellular coupling mediated by connexin 43. We also found that single targeting of DOR reduces TNF and IL-6 levels in the chronic phase of the disease, but the peripheral and central discharge as the primary source of excitotoxic stimulation in the spinal cord requires a simultaneous MOR-DOR targeting to reduce CCI-induced neuropathic pain. 相似文献

Acute lung injury (ALI) is a group of lung illnesses characterized by severe inflammation, with no treatment. The fatty acid amide hydrolase (FAAH) enzyme is an integral membrane protein responsible for the hydrolysis of the main endocannabinoids, such as anandamide (AEA). In pre-clinical pain and inflammation models, increasing the endogenous levels of AEA and other bioactive fatty acid amides (FAAs) via genetic deletion or the pharmacological inhibition of FAAH produces many analgesic benefits in several different experimental models. To date, nobody has investigated the role of FAAH inhibition on an ALI mouse model. Mice were subjected to a carrageenan injection and treated orally 1 h after with the FAAH inhibitor URB878 dissolved in a vehicle consisting of 10% PEG-400, 10% Tween-80 and 80% saline at different doses: The inhibition of FAAH activity was able to counteract not only the CAR-induced histological alteration, but also the cascade of related inflammatory events. URB878 clears the way for further studies based on FAAH inhibition in acute lung pathologies. 相似文献

Fibromyalgia is a syndrome characterized by chronic and widespread musculoskeletal pain, often accompanied by other symptoms, such as fatigue, intestinal disorders and alterations in sleep and mood. It is estimated that two to eight percent of the world population is affected by fibromyalgia. From a medical point of view, this pathology still presents inexplicable aspects. It is known that fibromyalgia is caused by a central sensitization phenomenon characterized by the dysfunction of neuro-circuits, which involves the perception, transmission and processing of afferent nociceptive stimuli, with the prevalent manifestation of pain at the level of the locomotor system. In recent years, the pathogenesis of fibromyalgia has also been linked to other factors, such as inflammatory, immune, endocrine, genetic and psychosocial factors. A rheumatologist typically makes a diagnosis of fibromyalgia when the patient describes a history of pain spreading in all quadrants of the body for at least three months and when pain is caused by digital pressure in at least 11 out of 18 allogenic points, called tender points. Fibromyalgia does not involve organic damage, and several diagnostic approaches have been developed in recent years, including the analysis of genetic, epigenetic and serological biomarkers. Symptoms often begin after physical or emotional trauma, but in many cases, there appears to be no obvious trigger. Women are more prone to developing the disease than men. Unfortunately, the conventional medical therapies that target this pathology produce limited benefits. They remain largely pharmacological in nature and tend to treat the symptomatic aspects of various disorders reported by the patient. The statistics, however, highlight the fact that 90% of people with fibromyalgia also turn to complementary medicine to manage their symptoms. 相似文献

Chitin and its oligosaccharides are involved in the plant defense response against fungal pathogens. In most studies, these molecules come from crustacean shell, and there are scarce studies on the use of fungal chitin. Usually the extraction of chitin is by alkaline treatments, which affect the acetylation degree (DA), and the obtaining of oligosaccharides of low molecular weight; so the use of enzymes is proposed as an alternative treatment to obtain chitin oligosaccharides from fungi. The objective of this work was to characterize the chitin fragments of Alternaria alternata extracted by alkaline and enzymatic treatments. The chitin extracted was ultrasonicated and ultrafiltrated to produce chitin fragments. Enzymatic treatments decreased the protein content to 29%, and the bands corresponding to β-glucans decreased when β-1,3-glucanase was applied. Fragments smaller than 1 kDa, DA of 76.7%, and 35.4 μg glucosamine/mg dry weight were obtained by enzymatic treatments. The enzymatic treatments show promising results to extract chitin fragments from A. alternata without severely affect the DA of the molecule. © 2018 Wiley Periodicals, Inc. J. Appl. Polym. A robustifying methodology for switched systems with matched and unmatched uncertainties/perturbations and autonomous location transitions is presented. We avail of such methodology to design a control strategy based on integral sliding modes, thereby ensuring theoretically exact compensation of the matched uncertainties/perturbations and the minimization of the effects of the unmatched ones. An output integral sliding‐mode control technique, based on a switched algebraic hierarchical observer is also proposed. This approach allows the theoretically exact compensation of the matched uncertainties/perturbations right after the first moment. The proposed approach is illustrated via numerical simulations. 相似文献

Circular splicing has been introduced to model a specific recombinant behaviour of circular DNA, continuing the investigation initiated with
linear splicing. In this paper we focus on the relationship between regular circular languages and languages generated by finite circular splicing systems. We survey the known results towards a characterization of the intersection between these two classes and provide new contributions
on the open problem of finding this characterization. First, we exhibit a non-regular circular language generated by a circular simple system thus disproving a known result in this area. Then we give new results related to a restrictive class of circular splicing
systems, the marked systems. Precisely, we review in a graph theoretical setting the recently obtained characterization of marked systems generating
regular circular languages. In particular, we define a slight variant of marked systems, that is the g-marked systems, and we introduce the graph associated with a g-marked system. We show that a g-marked system generates a regular circular
language if and only if its associated graph is a cograph. Furthermore, we prove that the class of g-marked systems generating regular circular languages is closed under a complement

The interaction between tyrosine-decarboxylase and proteolytic activities of a Lactobacillus curvatus and Staphylococcus xylosus, respectively, on biogenic amine production during the ripening and the storage of dry fermented sausages was investigated. Water content, pH, proteolysis parameters, microbial counts, and biogenic amine contents were monitored in spontaneously and starter fermented sausages. The use of proteolytic staphylococci as starter resulted in a higher content of non-protein nitrogen and total free amino acids. Tyramine was the main amine produced in all batches. However, tyrosine-decarboxylase activity of the L. curvatus starter strain was weak and yielded lower amounts of tyramine than those produced by the wild mioroflora in the control batch. Association between tyramine production and proteolysis could only be established in a defectively dried batch. Putrescine and cadaverine accumulation was efficiently reduced in the starter-mediated fermentation, in agreement with the lower development of enterobacteria. Phenylethylamine and tryptamine were only detected in the spontaneously fermented sausages, while histamine, spermine and spermidine did not vary during the ripening. Biogenic amine levels and related parameters showed significant changes during the storage of dry sausages depending on the temperature and the batch. As a general rule, changes in the pH, proteolysis, microbial counts, and biogenic amine contents were stronger at 19 degrees C than at 4 degrees C. The results suggest that refrigeration would be advisable for preventing further accumulation of biogenic amines during the storage of dry fermented sausages. 相似文献

The effects of selected variables, i.e. temperature, water activity and yeast inoculation level, on the lipolytic pattern and volatile production by Yarrowia lipolytica Y16A (chosen on the basis of a previous screening) were assessed. The variables were varied according to a central composite design and the models obtained enabled evaluation and weighting of the effects of the independent variables on the free fatty acids (FFAs) and volatile profiles in pork fat based medium. The polynomial models showed the levels temperature, water activity of the pork fat based system and yeast strain inoculation were able to maximize the release of specific FFAs or molecules of sensory importance. 相似文献

Manganese-doped zinc oxide thin films (ZnO:Mn) were grown on silicon and corning glass using double beam pulsed laser deposition. In this configuration, two synchronized pulsed-laser beams were employed to ablate independently ZnO and Mn targets. The presence of the zinc blende phase was investigated by means of X-ray diffraction, pulsed laser photoacoustic analysis and the calculation of the lattice parameter a. The crystallography plane (110) of the cubic zinc blende was found in all the films. Energy dispersive X-ray spectroscopy and different statistical analysis were employed to analyze the effect of the relative delay between plasma plumes on the average incorporation of Manganese. The minimum content of Mn—0.176 at%—was found for a relative delay of 10 µs, this result suggest that this delay is the inflection point to be considered in relation to a significant decrease in the incorporation of the dopant element. A significant positive Correlation analysis—r (4)?=?0.98, p?<?0.05—between the thickness and the average Mn incorporation was found, this means that as the percentage of manganese in the structure increases the thickness also increases. 相似文献

Abstract Polycyclic aromatic hydrocarbons (PAH) are typical indicators of incomplete combustion during solid waste incineration. The PAH emissions caused by waste incineration vary according to waste composition and operating parameters such as furnace temperature, after-combustion conditions, excess air, carbon monoxide levels. In this work, the content of PAH in the fly ashes (sampled upstream the flue gas treatment system) and bottom ashes produced by the combustion of a refuse derived fuel (RDF) in a rotary kiln incinerator equipped with an after-combustion chamber is determined. The emission levels of PAH are correlated with different operating conditions of the incinerator to evaluate the destruction efficiency of the after-combustion chamber for these compounds. 相似文献
