One of the pathological hallmarks of Alzheimer’s disease (AD) associated with its progression that contributes to β-amyloid (Aβ) generation is oxidative stress (OS). Clinical data suggest that melatonin is a potent antioxidant that might be effective in the adjunctive therapy of this neurodegenerative disease. The present study aimed to explore the role of melatonin on behavioral changes and markers of OS in three rat models, namely, pinealectomy (pin) model of melatonin deficit, intracerebroventricular (icv)Aβ1-42 model of AD, and combination of both pin and Aβ1-42 model (pin+icvAβ1-42). The chronic injection with vehicle/melatonin (50 mg/kg, i.p. for 40 days) started on the same day of sham/pin and icv vehicle/Aβ1-42 infusion procedures. Anxiety in the open field and the elevated plus-maze test and cognitive responses in the object recognition test were tested between the 30th–35th day after the surgical procedures. Markers of OS in the frontal cortex (FC) and hippocampus were detected by the ELISA method. Melatonin treatment corrected the exacerbated anxiety response only in the pin+icvAβ1-42 model while it alleviated the cognitive impairment in the three models. Pinealectomy disturbed the antioxidant system via enhanced SOD activity and decreased GSH levels both in the FC and hippocampus. The Aβ1-42 model decreased the SOD activity in the FC and elevated the MDA level in the two brain structures. The pin+icvAβ1-42 model impaired the antioxidant system and elevated lipid peroxidation. Melatonin supplementation restored only the elevated MDA level of icvAβ1-42 and pin+icvAβ1-42 model in the hippocampus. In conclusion, our study reveals that the pin+icvAβ1-42 rat model triggers more pronounced anxiety and alterations in markers of OS that may be associated with melatonin deficit concomitant to icvAβ1-42-induced AD pathology.  相似文献

Chitosan coating of polyethylene (PE) was proposed as a new procedure to improve its biocompatibility and surface properties. The functionalization of the PE film surface by covalent bonding of chitosan coating and its effect on the surface mechanical properties, as surface elasticity, stiffness, and adhesion (that are important in different biological processes) were investigated by nano‐indentation, scratch, and atomic force microscopy. It has been established that chitosan grafting onto corona functionalized PE surface using various coupling agents significantly improves the surface hardness and elastic modulus although they decrease in depth of the layer. Compared to the neat PE substrate, the chitosan coated samples show significant improved friction properties and tear resistance. The surface roughness features correlate with the micro‐mechanical parameters. Therefore, the covalent immobilization of the chitosan onto PE leads to a stable coating with better mechanical performance being recommended as a promising material for medical applications and food packaging. © 2015 Wiley Periodicals, Inc. The study compared the effects of swelling and dissolution of a matrix polymer by food simulants on the release of graphene nanoplates (GNPs) and multiwall carbon nanotubes (MWCNTs) from poly(lactic) acid (PLA) and polypropylene (PP) composite films. The total migration was determined gravimetrically in the ethanol and acetic acid food simulants at different time and temperature conditions, while migrants were detected by laser diffraction analysis and transmission electron microscopy. Swelling, thermal analysis, and scanning electron microscopy were applied to characterize the degradation of polymer films at the migration conditions. The release of nanoparticles was found in a high‐temperature migration test of 4 h at 90 °C. The hydrolytic dissolution of the PLA polymer in the food simulants caused a migration of GNPs (>100 nm) from the PLA/GNP/MWCNT films into the simulant solvents, while the entangled MWCNTs formed a network on the film surface, preventing their migration from the PLA composite films. In contrast, the PP polymer slightly swells in ethanol solvents, allowing some short carbon nanotubes to be released from the surface and cut edges of the PP/MWCNT film into food simulants. Mathematical modeling of diffusion was applied that accounts for type of polymer, time–temperature conditions, and solvent concentration; model parameters were validated with experimental results. © 2017 Wiley Periodicals, Inc. Poly(vinyl acetate-co-vinyl chloroformate) ( 1 ) was synthesized via phosgenation of poly(vinyl acetate-co-vinyl alcohol). It was shown that ( 1 ) is capable of initiating the polymerization of 2-phenyl-2-oxazoline and 2-methyl-2-oxazoline, when the counter ion is exchanged using potassium iodide. Polyvinyl acetate-graft-poly-2-phenyl-2-oxazoline and polyvinyl acetate-graft-poly-2-methyl-2-oxazoline are obtained in the grafting reaction.  相似文献

The pineal gland regulates the aging process via the hormone melatonin. The present report aims to evaluate the effect of pinealectomy (pin) on behavioral and oxidative stress-induced alterations in cholesterol and sphingomyelin (SM) levels in young adult, mature and aging rats. Sham and pin rats aged 3, 14 and 18 months were tested in behavioral tests for motor activity, anxiety, and depression. The ELISA test explored oxidative stress parameters and SM in the hippocampus, while total cholesterol was measured in serum via a commercial autoanalyzer. Mature and aged sham rats showed low motor activity and increased anxiety compared to the youngest rats. Pinealectomy affected emotional responses, induced depressive-like behavior, and elevated cholesterol levels in the youngest rats. However, removal of the pineal gland enhanced oxidative stress by diminishing antioxidant capacity and increasing the MDA level, and decreased SM level in the hippocampus of 14-month-old rats. Our findings suggest that young adult rats are vulnerable to emotional disturbance and changes in cholesterol levels resulting from melatonin deficiency. In contrast, mature rats with pinealectomy are exposed to an oxidative stress-induced decrease in SM levels in the hippocampus.  相似文献

BACKGROUND: The polymerization of hexanelactam (HL) is complicated by numerous side reactions: chain transfer to monomer, termination of the polymer chain, exchange as transamidation and formation of β‐ketoimides and β‐ketoamides. The generation of by‐products and their effect on the anionic polymerization of HL in the bulk in the presence of functionalized tetrahydrofuran oligomers as polymeric activators (PACs) were investigated. RESULTS: In order to consider how side reactions influence the process of HL polymerization, qualitative and quantitative determination of the by‐products obtained and their exhaustion were carried out. An investigation of the polymerization kinetics and the formation of low molecular weight by‐products through the polymerization was performed. The products are further involved in the polymerization process and consumed until an equilibrium level is reached. The mechanism of their generation is discussed. These processes were followed during the first polymerization stage by means of gel permeation chromatography. A mechanism of by‐product formation and consumption is proposed. CONCLUSION: The investigation of the polymerization kinetics and the formation of low molecular weight by‐products through the polymerization has shown that the side reaction yields oligomeric HL‐based products. The products are further involved in the polymerization process and consumed until an equilibrium level is reached. The equilibrium values depend on the HL/PACs ratio. Using ICP-MS and ICP-AES platinum group elements (Pt, Pd, Rh, Ru and Ir) and Ce, La, Nd, Pb and Zr have been determined in street dust, Taraxacum officinale (dandelion), Plantago lanceolata (plantain), Lolium multiflorum (annual ryegrass), Rhytidiadelphus squarrosus (moss) and Vascellum pratense (mushrooms) collected along highways and streets in Germany during 1999. Among the plants Taraxacum officinale (dandelion) reflects most adequately the pollution with the investigated elements matching the results from street dust. A strong positive correlation between all elements determined in the plants is established. Transfer factor for Pt between soil and plants has been determined in an agricultural experiment ranging between 0.004 and 0.008 for two types of soils.  相似文献
