The design, synthesis, and bioevaluation of fluorescence- and biotin-labeled CXCR4 antagonists are described. The modification of D-Lys8 at an epsilon-amino group in the peptide antagonist Ac-TZ14011 derived from polyphemusin II had no significant influence on the potent binding of the peptide to the CXCR4 receptor. The application of the labeled peptides in flow cytometry and confocal microscopy studies demonstrated the selectivity of their binding to the CXCR4 receptor, but not to CXCR7, which was recently reported to be another receptor for stromal cell-derived factor 1 (SDF-1)/CXCL12.  相似文献

Conformationally constrained peptides hold promise as molecular tools in chemical biology and as a new modality in drug discovery. The construction and screening of a target-focused library could be a promising approach for the generation of de novo ligands or inhibitors against target proteins. Here, we have prepared a protein kinase-focused library by chemically modifying helix-loop-helix (HLH) peptides displayed on phage and subsequently tethered to adenosine. The library was screened against aurora kinase A (AurA). The selected HLH peptide Bip - 3 retained the α-helical structure and bound to AurA with a KD value of 13.7 μM. Bip - 3 and the adenosine-tethered peptide Bip - 3 - Adc provided IC50 values of 103 μM and 7.7 μM, respectively, suggesting that Bip - 3 - Adc bivalently inhibited AurA. In addition, the selectivity of Bip - 3 - Adc to several protein kinases was tested, and was highest against AurA. These results demonstrate that chemical modification can enable the construction of a kinase-focused library of phage-displayed HLH peptides.  相似文献

Phylogenetic analyses were carried out on a total of 118 Listeria monocytogenes isolates from foods or food processing environments, and 7 isolates from listeriosis patients in Japan to evaluate the genetic variation in the pathogen in this country. Isolates of serotypes 1/2a, 1/2b and 4b were mainly examined to assess the risk of exposure of humans to L. monocytogenes from foods in Japan. The nucleotide sequences of the part of the iap gene that contains the region encoding the threonine-asparagine repeat units were determined in order to construct phylogenetic trees of the isolates investigated. A phylogram showed high genetic diversity among lineage 2 isolates, while the lineage 1 isolates showed clonal characteristics. The results of the genetic analyses suggested the presence of rare putative lineage 3 isolates and epidemic clone I (ECI) isolates in foods in Japan. The results showed that ECI was also isolated from listeriosis patients. The genetic variation in L. monocytogenes in Japan reported here suggests the necessity of monitoring the pathogen in foods and environments in addition to surveillance of listeriosis patients.  相似文献

This study examines the performance of “string-of-beads” liquid flow on vertical wires as a novel gas-liquid contact device for gas absorption using a chemically reacting liquid absorbent. The string-of-beads flow is a distinct on-wire liquid-flow pattern consisting of annular thin liquid films sheathing a wire and teardrop-shaped liquid beads alternately aligned on the wire at regular intervals. We have performed experiments of CO2 absorption by an aqueous monoethanolamine solution in string-of-beads flow on a single wire and have developed an analytic gas-absorption model relevant to the experiments. The model well simulates the CO2-absorption characteristics observed in the experiments, thereby suggesting the utility of the model in predicting the chemical gas-absorption performance of multiple-wire gas-liquid contactors.  相似文献

In 2007, samples of treated effluent were collected at point of discharge to the environment from 39 wastewater treatment plants (WWTPs) located across Victoria, Australia grouped by treatment type. Sample genotoxicity was assessed with a high-throughput luminescent umu test method using Salmonella typhimurium TL210 strain, with and without addition of a commercially available metabolic activation system. Samples were also screened using a gas chromatographic-mass spectrometric mass-structure database recognition method. Amorphous matrices made up of sugar molecules, are frequently used in food and pharmaceutical industries. A drawback to their use is that they are susceptible to collapse, as a result of water uptake and an increase in temperature and subsequently crystallize. Herein, the crystallization characteristics of amorphous sugar (sucrose and α-lactose) preparations were analyzed, with the purpose of obtaining knowledge that could lead to the prediction of how long the amorphous state is retained under various conditions. The impact of compression, physical aging and freezing rate on the induction period (tind) for crystallization were examined. Freeze-dried sugar samples were compressed at 74 or 443 MPa (5 min) and then rehumidified at specified RHs. Some freeze-dried sucrose samples were physically aged, and alternatively freeze-drying was conducted under different conditions. The isothermal crystallization of the prepared samples at different temperatures (T), the glass transition and the crystallization temperature (Tcry) were measured, using differential scanning calorimetry. The compression markedly decreased the tind, while significantly lowered the hygroscopicity. Physical aging and slower-freezing also shortened the tind. The tind was found to be correlated exclusively with (TcryT), regardless of rehumidification, compression, sugar type, physical aging and freezing rate in the freeze-drying process.  相似文献

A micro‐grooved evaporator is composed of µm‐wide grooves on a heat transfer plate in which the inter‐line regions at the liquid–vapor meniscus of coolant become identifiable. The high‐heat performance of the evaporator is realized by this inter‐line region (ILR) where the liquid thin film reduces the thermal resistance on the heat transfer surface. In this report, we propose a numerical simulation model of heat and mass transfer in a single groove to predict its capillary force and heat flux. The capillary force performance (capillary‐rise length in a groove) of a single groove was measured for samples of varying width, superheat, and inclination. The performance was found to be a maximum at a specific groove width of 200–400 µm, which is in good agreement with the predicted results calculated by the proposed model. For a better prediction of capillary‐rise length, the effective capillary force and the effective flow resistance were considered. © 2009 Wiley Periodicals, Inc. The structure development in the continuous laser-heated drawing process of poly(ethylene 2,6-naphthalene dicarboxylate) (PEN) fiber was analyzed by in-situ X-ray diffraction measurement. Because of the rapid and uniform laser heating, and the resultant steady-state nature of the necking-drawing, the structure development after the on-set of necking could be measured in the time resolution of several hundred microseconds. We found for the first time the temporal appearance of meridional (001′) diffraction at several milliseconds after the on-set of necking indicating that the mesophase structure similar to the one reported for poly(ethylene terephthalate) was also formed in the initial stage of fiber structure development of PEN. The d-spacing of the (001′) diffraction 1.230 ± 0.003 nm was shorter than the c-axis lengths of both α and β crystals.  相似文献

The effect of the addition of certain low molecular weight compounds to highly crosslinked epoxy resins was investigated. The behavior which is known as antiplasticization, such as increases in modulus and yield strength and the elimination of β-transition, was observed in the highly crosslinked system as well as in the reported cases of thermoplastics—poly(vinyl chloride), bisphenol A polycarbonate, and polysulfone. Chlorinated biphenyl was found to be one of the most effective antiplasticizers examined in the present paper. However, the remarkable reduction in compressive strain at break and impact strength was not recognized for the highly antiplasticized samples. These phenomena have been interpreted on the basis of free volume concept.  相似文献
