The conceptual design of the 1 MW X-band transmit system for the Goldstone Solar System Radar represents the next step in the quest to expand planetary radar exploration. The radar transmitter requirements are summarized. The characteristics and expected performance of the major elements are discussed, including the klystrons, power amplifiers, microwave transmission lines, feed systems, beam power supply, heat exchanger, phase stable exciter, and the monitor/control system. An assessment of the technology development needed to meet the system requirements is given, and possible areas of difficulty are outlined  相似文献

C-terminal alpha-amidation is a post-translational modification necessary for the biological activity of many regulatory peptides produced in the respiratory tract. This modification is a two-step process catalyzed by two separate enzyme activities, both derived from the peptidyl-glycine alpha-amidating mono-oxygenase (PAM) precursor. The distribution of these two enzymes, peptidyl-glycine alpha-hydroxylating monoxygenase (PHM) and peptidyl-alpha-hydroxyglycine a amidating lyase (PAL), was studied in the normal lung and in lung tumors using immunocytochemical methods and in situ hybridization. In normal lung the enzymes were located in some cells of the airway epithelium and glands, the endothelium of blood vessels, some chondrocytes of the bronchial cartilage, the alveolar macrophages, smooth muscle cells, neurons of the intrinsic ganglia, and in myelinated nerves. A total of 24 lung tumors of seven different histological types were studied. All cases contained PAM-immunoreactive cells with various patterns of distribution. All immunoreactive cells were positive for the PHM antiserum but only some of them for the PAL antiserum. The distribution of PAM co-localizes with some other previously described amidated peptides, suggesting that amidation is an important physiological process taking place in the normal and malignant human lung tissue.  相似文献

This paper describes fabrication and characterization results of piezoelectric micromachined ultrasonic transducers (pMUTs) based on 2-microm-thick Pb(Zr0.53Ti0.47O3) (PZT) thin films. The applied structures are circular plates held at four bridges, thus partially unclamped. A simple analytical model for the fully clamped structure is used as a reference to optimize design parameters such as thickness relations and electrodes, and to provide approximate predictions for coupling coefficients related to previously determined thin film properties. The best coupling coefficient was achieved with a 270-microm plate and amounted to kappa2 = 5.3%. This value compares well with the calculated value based on measured small signal dielectric (epsilon = 1050) and piezoelectric (e3l,f = 15 Cm(-2)) properties of the PZT thin film at 100 kV/cm dc bias. The resonances show relatively large Q-factors, which can be partially explained by the small diameters as compared to the sound wavelength in air and in the test liquid (Fluorinert 77). A transmit-receive experiment with two quasi-identical pMUTs was performed showing significant signal transmission up to a distance of 20 cm in air and 2 cm in the test liquid.  相似文献

We have engineered a latent mouse interferon beta (mIFNbeta) using the latency associated peptide (LAP) of transforming growth factor beta1 (TGF-beta1) to protect the cytokine and avoid its interaction with its receptors. This approach improves the pharmacokinetic properties and reduces the pleiotropic effects limiting the current therapeutic use of cytokines. IFNbeta was fused to the LAP using two flexible linkers flanking a matrix metalloproteinase (MMP) cleavage site for the specific release of IFNbeta at disease sites. In order to improve the hydrolysis rate of the cleavage site, 15 different cleavable linkers were introduced in the LAP-mIFNbeta construct. The kinetic parameters relative to the linker cleavage by MMP-1 and MMP-3 were measured by an ELISA method. Among the modifications done, one of the constructs bearing the activation site of pro-MMPs was the best substrate for both enzymes. The introduction of a hydrophilic sequence derived from the furin cleavage site of the anthrax toxin protective antigen increased the sensitivity to MMP-3 to up to 29-fold. These data suggest that this strategy could be useful for improving the effectiveness of the delivery and targeting of protein therapeutics.  相似文献

A novel pipeline architecture for CMOS static RAMs (SRAMs) that allows operation at very high clock rates is described. Basic requirements for achieving high speed are the implementation of a hierarchical architecture and a memory cell with separate READ and WRITE data lines. The access speed of hierarchically organized memory blocks was between 2.5 and 3.5 ns. The maximum operating frequency of a 16 K pipelined hierarchical SRAM (PHSRAM) is in the range of 300 MHz. The hierarchical architecture and a seven-transistor memory cell provide a circuit using digital swings all over. Key advantages of the full-swing static logic circuitry are robustness with respect to fabrication tolerances and a high-noise immunity. Moreover, the circuit can be reduced to finer structure sizes without any redesign, since there are no critical analog circuit parts  相似文献

Gingival overgrowth is a common health problem caused by genetic and environmental risk factors. Animal models for quantitative histological studies are needed to uncover genetic predisposition and dose-response data that might put individuals at increased risk for gingival disease. Gingival height, thickness, inflammation, and the degree of encroachment of gingiva over the tooth, are clinical measures of overgrowth; most of these parameters can be measured histologically, but in order to quantify gingival coverage of the tooth, the image of the crown must be present. Tooth and bone typically require decalcification for histology; thus, the tooth crown, a critical landmark, is lost. We describe a method for imaging the crown histologically, using impression materials applied to dissected mouse mandibles. Four dental alginates, three polyvinyl siloxanes, and one polyether and gelatin were used. The impression-material/mandibular tissue blocks were processed routinely. Polyvinyl siloxanes were incompatible with embedding resin; alginates, polyether and gelatin could be fixed, decalcified, embedded, and sectioned. Alginates and gelatin could be stained. Success in imaging the tooth crown varied with the preparation, but the alginates, polyether, and gelatin permitted a useful degree of measurement of exposed crown and enamel thickness, along with other morphometric parameters such as thickness of the dentin, lateral mandibular ramus, rete pegs, height of the gingiva, and volume density of vessels and inflammatory cells in the lamina propria. In conclusion, this new application for impression materials allows gingival coverage of tooth crown, as well as numerous other parameters to be measured for comparison with clinical data.  相似文献

In this article, mid-infrared Fourier transform (Mid-FT-IR) and carbon thirteen nuclear magnetic resonance (13C NMR) spectroscopy have been used to determine possible interactions between sucrose and various alkali or alkaline earth metals in aqueous solution. In the presence of these metals, significant shifts in the absorption bands of sucrose were noted by mid-FT-IR coupled with principal component analysis (PCA). These shifts were explained on the basis of weakening of the H-bond network between sucrose and water and possible interactions between sucrose and the metal ion. Factorial maps were established and the spectral patterns obtained show that these interactions vary according to the nature of the metal ion. 13C NMR analysis showed that the carbon atoms of sucrose undergo shielding or deshielding in the presence of metal ions in aqueous solutions. Two factors were invoked to account for the variation of chemical shifts: the rupture of hydrogen bonds due to hydration of the metal ion and the possible coordination of the metal ion to the oxygen atoms of sucrose.  相似文献

Photoreactive analogues of substance P (biotin sulfone-spacer (amino pentanoic or Gly(3))-Arg-Pro-Lys-Pro-(pBzl)Phe-Gln-Phe-Phe-Gly-Leu-Met(O(2))NH(2)) with or without isotope (deuterium) labeling have been synthesized. Deuteriums were present on (d)-biotin or epibiotin sulfone (D(3)), on the Gly(3) spacer linker (D(6)), or on the Gly in position 9 of SP (D(2)). Therefore, peptide analogues could be either unlabeled or tri-, penta-, or hexadeuterated. Results obtained with the use of these peptide analogues show that (d)-biotin sulfone and epibiotin sulfone are not recognized with the same affinity by streptavidin, with (d)-biotin sulfone displaying better affinity for the protein. Photolabeling of the human NK-1 receptor with a 1:1 molar ratio of nondeuterated and deuterated photoreactive substance P (SP) analogues in position 5, followed by combined digestions, purification, and MALDI-TOF mass spectrometry analysis, made the identification of the domain of the receptor covalently linked by the photoreactive SP analogue easier. Indeed, doublets in mass spectra were specific for the covalent complex whereas single peaks could be attributed to contaminating species. This method is particularly suitable when minute amounts of complex have to be analyzed, as in the case of highly hydrophobic G-protein coupled receptors.  相似文献

The technological and economical pressure in production engineering requires an optimisation of existing production processes. The finite element method (FEM) is an appropriate tool to gain the necessary knowledge of these processes. This paper will present some examples of the application of FEM in different branches of manufacturing technology: cutting with a geometrically defined and an undefined cutting edge; electro-erosion technology; metal forming processes; tribology. After the presentation of the problem, the simulation set-up for each example will be discussed and the results will be presented. Thus, this paper will provide an overview of the current possibilities for the application of FEM in manufacturing technology.  相似文献

A simple nucleus for a single-processor computer is considered. The entire nucleus program is written in the high-level programming language Modula-21 without assembly coding. The concepts of the nucleus and some simple applications are described. A complete listing of the nucleus is included. We conclude that programming of a nucleus in an appropriate high-level language is simpler than in an assembly language. The resulting nucleus has a comparable performance with other nuclei written in assembly code.  相似文献
