Drug which shows extensive first pass effect is difficult task that, needs to be solved by formulators in the pharmaceutical science. The low oral bioavailability (49%) of flutamide may be due to poor wettability, low aqueous solubility and extensive first pass effect. The aim of present investigation was to prepare flutamide loaded microspheres and incorporate it into suppositories for rectal delivery to avoid first pass effect and enhance residence time. Flutamide loaded mucoadhesive microspheres of Ocimum Basilicum mucilage (OBM) were prepared using spray drying and characterized by percent production yield, encapsulation efficiency, particle size, zeta potential, polydispersity index, DSC, SEM, XRPD, in vitro drug release and stability studies. Moreover, ex vivo mucoadhesion was investigated using falling liquid film technique to determine the adhesion of microspheres to sheep rectal mucosa. The microspheres had nearly spherical shape and size about 2.53?μm. The encapsulation efficiency and mucoadhesion of optimized formulation MBF10 were found to be 69.6?±?2.3% and 89.01?±?2.18%, respectively. Percent CDR of optimized flutamide loaded mucoadhesive microspheres was found to be 88.7?±?1.3 at 7?h. In conclusion, OBM microparticles based suppository could be used to deliver drug through rectal delivery. 相似文献

Here we report a successful preparation of nanostructured calcium silicate by wet chemical approach. The synthesized sample was characterized by various physico-chemical methods. Thermal stability was investigated using thermo-gravimetric and differential thermal analysis (TG-DTA). Structural characterization of the sample was carried out by the X-ray diffraction technique (XRD) which confirmed its single phase hexagonal structure. Transmission electron microscopy (TEM) was used to study the nanostructure of the ceramics while homogeneous grain distribution was revealed by scanning electron microscopy studies (SEM). The elemental analysis data obtained from energy dispersive X-ray spectroscopy (EDAX) were in close agreement with the starting composition used for the synthesis. Superhydrophilic nature of CaSiO3 was investigated at room temperature by sessile drop technique. Effect of porous nanosized CaSiO3 on early adhesion and proliferation of human bone marrow mesenchymal stem cells (BMMSCs) and cord blood mesenchymal stem (CBMSCs) cells was measured in vitro. MTT cytotoxicity test and cell adhesion test showed that the material had good biocompatibility and promoted cell viability and cell proliferation. It has been stated that the cell viability and proliferation are significantly affected by time and concentration of CaSiO3. This paper presents a geocast technique for underwater sensor networks. The model named as Routing and Multicast Tree based Geocasting (RMTG) (TRACE'10, Bhopal, India, 2010; ICWET'10, Mumbai, India, 2010) has been designed for underwater sensor networks. In our previous work (TRACE'10, Bhopal, India, 2010; ICWET'10, Mumbai, India, 2010) we proposed only a theoretical model for underwater geocasting. The RMTG technique uses greedy forwarding and previous hop handshaking to route the packets towards the geocast region and further disseminates the data within the geocast region by creating a multicast shortest path tree. We also presented various link broken scenarios along with their solutions. The proposed model does not use the flooding technique to deliver the packets inside the geocast region that is used in most of the geocasting techniques. In this paper, we present the simulated RMTG model. The simulation results show that the proposed model provides an efficient distribution of data in the geocast region in terms of node mobility handling, packet delivery ratio and a better end‐to‐end latency. Most of the work on geocasting has been done for mobile and vehicular ad hoc networks; hence we present in this paper a novel simulated model for underwater environment. In Brazil, the National Council for Scientific and Technological Development (CNPq) distributes productivity fellowships in research (RS) as a recognition to individuals with outstanding productivity levels in their areas. The aim of this study is to evaluate the scientific production of the Brazilian Pharmacy area, one division of the Health Sciences Great Area of CNPq, considering the profile and productivity levels of RS fellows. The results showed that most of the 156 active RS fellows in 2015 were female, with doctorate completed in the Southeast region (mainly in University of São Paulo) and with research activities developed in the South and Southeast regions. Most of their work was published in journals classified as B1 and B2 Qualis in Pharmacy by the Coordination for the Improvement of Higher Education Personnel (CAPES), with high prevalence of publications in local journals and/or specialized on medicinal plants. Besides, they featured much dependence on advising and productivity indexes related to the category and level of RS fellowship. The evolution of such data must be continually evaluated to determine the influence of CNPq productivity fellowships on performance and stratification of researchers in the Pharmacy area in Brazil. 相似文献

Here we report fabrication and evaluation of novel surface modified polymer–lipid hybrid nanoparticles (PLN) as robust carriers for intranasal delivery of ropinirole hydrochloride (ROPI HCl). Sustained release, avoidance of hepatic first pass metabolism, and improved therapeutic efficacy are the major objectives of this experiment. PLN were fabricated by emulsification-solvent diffusion technique and evaluated for physicochemical parameters, in vitro mucoadhesion, in vitro diffusion, ex vivo permeation, mucosal toxicity and stability studies. Box-Behnken experimental design approach has been employed to assess the influence of two independent variables, viz. surfactant (Pluronic F-68) and charge modifier (stearylamine) concentration on particle size, ζ-potential and entrapment efficiency of prepared PLN. Numerical optimization techniques were used for selecting optimized formulation sample, further confirmed by three dimensional response surface plots and regression equations. Results of ANOVA demonstrated the significance of suggested models. DSC and SEM analysis revealed the encapsulation of amorphous form of drug into PLN system, and spherical shape. PLN formulation had shown good retention with no severe signs of damage on integrity of nasal mucosa. Release pattern of drug-loaded sample was best fitted to zero order kinetic model with non-Fickian super case II diffusion mechanism. In vivo pharmacodynamic studies were executed to compare therapeutic efficacy of prepared nasal PLN formulation against marketed oral formulation of same drug. In summary, the PLN could be potentially used as safe and stable carrier for intranasal delivery of ROPI HCl, especially in treatment of Parkinson’s disease. 相似文献

A model was developed to simulate oxygen accumulation in space and time within cylindrical dynamic accumulation chambers that are used to measure oxygen transmission rate (OTR) of materials. The model is based on Fick's law of diffusion and was validated against actual OTR measurements of polymer film samples. Measured OTR values and thicknesses were inputted into the model and oxygen concentrations outputted by the model. OTRs determined from the output of the model was in close agreement to within 0.3–3% of the measured OTR. Oxygen concentration versus time curves generated from model output oxygen concentrations and experimentally measured oxygen concentrations for three actual films were also in agreement. The model was then used to simulate results from three hypothetical test films at varying chamber lengths in order to evaluate effects of accumulation chamber dimensions relative to films on resulting OTR measurements. A typical design scenario was used, where the oxygen sensor is mounted on the chamber wall opposite the sample film. Results demonstrate that dynamic accumulation OTR instrument designers have considerable flexibility in choosing accumulation chamber dimensions because deviations in OTR are only expected to occur at impractically extreme chamber lengths (>10 m) for the entire envelope of OTRs expected for typical packaging films. There is growing interest in understanding how emotion regulation affects adaptation. The present study examined expressive suppression (which involves inhibiting the overt expression of emotion) and how it affects a critical domain of adaptation, social functioning. This investigation focused on the transition to college, a time that presents a variety of emotional and social challenges. Analyses focused on 2 components of suppression: a stable component, representing individual differences expressed both before and after the transition, and a dynamic component, representing variance specific to the new college context. Both components of suppression predicted lower social support, less closeness to others, and lower social satisfaction. These findings were robustly corroborated across weekly experience reports, self-reports, and peer reports and are consistent with a theoretical framework that defines emotion regulation as a dynamic process shaped by both stable person factors and environmental demands.
