The enzyme glucose oxidase (GOD) was immobilized on the nonwoven fabrics, which have excellent properties in diffusivity of substrates, mechanical strength, and handling, with Bombyx mori silk fibroin gel. The nonwoven fabrics of silk fibroin, viscose rayon, poly-ethyleneterephthalate, 6-nylon, and polypropylene with activated surface by fluoline treatment were used. The stabilities of GOD to heat or pH changes were much improved by the immobilization as well as the case of the GOD immobilized in the silk fibroin membrane. Among nonwoven fabrics, silk fibroin was the most excellent support material for the immobilization of GOD although all nonwoven fabrics used here are able to be used as the support materials. The increase of the sensitivity was observed when the glucose sensor was made with the GOD immobilized on nonwoven silk fabrics as four times compared with the case of the GOD immobilized in the silk fibroin membrane.  相似文献

According to the investigations of Cameron and his coworkers1–5 into the chain-matching phenomena between normal paraffin as a solvent and straight-chain fatty acid as an additive, the optimum condition in boundary lubrication is attained when the chain lengths of the additive and solvent are equal. A series of experiments has been conducted over the past ten years on the role of the chain-matching effect, not only on boundary lubrication but also widely on relevant interfacial phenomena. Reproducible amounts of lactic acid accumulate in minced kitchen refuse under open conditions with intermittent pH neutralization [Sakai et al., Food Sci. Technol. Res., 6, 140 (2000)]. Here, we showed that such pH-controlled open fermentation of kitchen refuse reproducibly resulted a selective proliferation of a major lactic acid bacterial (LAB) species. In one experiment, the predominant microorganisms isolated during the early phase (6 h) were Gammaproteobacteria. In contrast, those that predominated during the late phase (48 h) were always Lactobacillus plantarum in three independent experiments. To further quantify the microbial community within open lactic acid fermentation, we performed fluorescent in situ hybridization (FISH) analysis targeting 16S (23S) rRNA. We designed two new group-specific DNA probes: LAC722(L) was active for most LAB including the genera Lactobacillus, Pediococcus, Leuconostoc and Weisella, whereas Lplan477 was specific for L. plantarum and its related species. We then optimized sample preparation using lysozyme and hybridization conditions including temperature, as well as the formamide concentration and the salt concentration in the washing buffer. We succeeded in quantification of microorganisms in semi-solid, complex biological materials such as minced kitchen refuse by taking color microphotographs in modified RGB balance on pre-coated slides. FISH analysis of the fermentation of kitchen refuse indicated that control of the pH swing leads to domination by the LAB population in minced kitchen refuse under open conditions. We also confirmed that L. plantarum, which generates lactic acid in high quantities but with low optical activity, became the dominant microorganism in kitchen refuse during the late phase of open fermentation.  相似文献

Acetate ester synthesis was studied in vitro with the ethyl acetate-producing yeast Candida utilis. The level of enzyme activity observed for the NAD+-dependent hemiacetal dehydrogenase acting on hemiacetal, which was produced non-enzymatically from an alcohol and an aldehyde, was much greater than that for the other enzyme involved in ester synthesis, alcohol acetyltransferase. The level of ethyl acetate synthesis in vivo approximately paralleled the hemiacetal dehydrogenase (HADH) activity. The results suggest that the main pathway for ethyl acetate synthesis in C. utilis involves a novel hemiacetal dehydrogenase activity.  相似文献

Direct modulation of an InGaAsP/InP double heterostructure laser was investigated experimentally. Sinusoidal modulation up to 2.5 GHz was achieved with almost constant modulation efficiency. Pulse responses showed that the damped relaxation oscillation in light output was well suppressed.  相似文献

Room-temperature c.w. operation of InGaAsP/InP heterostructure lasers grown by liquid-phase epitaxy was achieved at 1.56 ?m. An active InGaAsP layer was essentially sandwiched by InP, though a thin InGaAsP buffer layer was deposited to prevent the melt-back of the active layer. Threshold current was typically 300 mA for a 17 ?m wide oxide-defined stripe laser.  相似文献

The evolution mechanisms of new high-angle boundaries as well as ultrafine grains at large strains were studied by means of multidirectional forging (MDF) of pure copper at low temperature and aluminum alloy at high temperature, where dynamic recovery operates as a main restoration process. The structural changes can be characterized by the evolution of deformation bands such as microshear or kink bands at moderate strains. Multidirectional forging accelerates the evolution of many mutually crossing microshear or kink bands developed in various directions. The misorientations between (sub)grains increased gradually with increasing cumulative strain, finally leading to the development of a new fine-grained structure. The dynamic grain formation can be resulted from in situ or continuous dynamic recrystallization which is discussed in detail.  相似文献

To clarify the effect of the suppression of natural heat transfer, the local heat transfer coefficients on a vertical cooled flat plate with circular grooves were measured by a multi‐type thermocouple method. Two flat plates with and without periodic circular grooves were tested in this experiment. The characteristics of heat transfer along the plate for both plates were compared. The local heat transfer coefficients on the periodic grooved plate became smaller than that of the flat plate. The flow pattern was changed when it passed over the grooves, and circulation was generated in the grooves in the downstream. As a result, the thickness of the thermal boundary layer on the grooved plate was more developed than the normal flat plate. © 2010 Wiley Periodicals, Inc.
