We present a novel algorithm for joint state-parameter estimation using sequential three dimensional variational data assimilation (3D Var) and demonstrate its application in the context of morphodynamic modelling using an idealised two parameter 1D sediment transport model. The new scheme combines a static representation of the state background error covariances with a flow dependent approximation of the state-parameter cross-covariances. For the case presented here, this involves calculating a local finite difference approximation of the gradient of the model with respect to the parameters. The new method is easy to implement and computationally inexpensive to run. Experimental results are positive with the scheme able to recover the model parameters to a high level of accuracy. We expect that there is potential for successful application of this new methodology to larger, more realistic models with more complex parameterisations.  相似文献

The Hybrid Sulfur Process, as well as similar sulfur cycles for the production of nuclear hydrogen, requires the decomposition of sulfuric acid into sulfur dioxide, oxygen, and water at temperatures above 800 °C and at pressures up to 9 MPa. The design of a reactor for this process presents numerous challenges in terms of maintaining small pressure differentials and utilizing currently available materials of construction. This paper focuses on design calculations for a composite reactor that preheats, concentrates, and decomposes sulfuric acid for use in the production of hydrogen. A pharmacological and genetic blockade of the dopamine D3 receptor (D3R) has shown to be neuroprotective in models of Parkinson’s disease (PD). The anxiolytic drug buspirone, a serotonin receptor 1A agonist, also functions as a potent D3R antagonist. To test if buspirone elicited neuroprotective activities, C57BL/6 mice were subjected to rotenone treatment (10mg/kg i.p for 21 days) to induce PD-like pathology and were co-treated with increasing dosages of buspirone (1, 3, or 10 mg/kg i.p.) to determine if the drug could prevent rotenone-induced damage to the central nervous system (CNS). We found that high dosages of buspirone prevented the behavioural deficits caused by rotenone in the open field test. Molecular and histological analyses confirmed that 10 mg/kg of buspirone prevented the degeneration of TH-positive neurons. Buspirone attenuated the induction of interleukin-1β and interleukin-6 expression by rotenone, and this was paralleled by the upregulation of arginase-1, brain-derived neurotrophic factor (BDNF), and activity-dependent neuroprotective protein (ADNP) in the midbrain, striatum, prefrontal cortex, amygdala, and hippocampus. Buspirone treatment also improved mitochondrial function and antioxidant activities. Lastly, the drug prevented the disruptions in the expression of two neuroprotective peptides, pituitary adenylate cyclase-activating polypeptide (PACAP) and vasoactive intestinal peptide (VIP). These results pinpoint the neuroprotective efficacy of buspirone against rotenone toxicity, suggesting its potential use as a therapeutic agent in neurodegenerative and neuroinflammatory diseases, such as PD.  相似文献

Pathogenic mutations in USH2A are a leading cause of visual loss secondary to non-syndromic or Usher syndrome-associated retinitis pigmentosa (RP). With an increasing number of RP-targeted clinical trials in progress, we sought to evaluate the photoreceptor topography underlying patterns of loss observed on clinical retinal imaging to guide surrogate endpoint selection in USH2A retinopathy. In this prospective cross-sectional study, twenty-five patients with molecularly confirmed USH2A-RP underwent fundus autofluorescence (FAF), spectral-domain optical coherence tomography (SD-OCT) and adaptive optics scanning laser ophthalmoscopy (AOSLO) retinal imaging. Analysis comprised measurement of FAF horizontal inner (IR) and outer (OR) hyperautofluorescent ring diameter; SD-OCT ellipsoid zone (EZ) and external limiting membrane (ELM) width, normalised EZ reflectance; AOSLO foveal cone density and intact macular photoreceptor mosaic (IMPM) diameter. Thirty-two eyes from 16 patients (mean age ± SD, 36.0 ± 14.2 years) with USH2A-associated Usher syndrome type 2 (n = 14) or non-syndromic RP (n = 2) met the inclusion criteria. Spatial alignment was observed between IR-EZ and OR-ELM diameters/widths (p < 0.001). The IMPM border occurred just lateral to EZ loss (p < 0.001), although sparser intact photoreceptor inner segments were detected until ELM disruption. EZ width and IR diameter displayed a biphasic relationship with cone density whereby slow cone loss occurred until retinal degeneration reached ~1350 μm from the fovea, beyond which greater reduction in cone density followed. Normalised EZ reflectance and cone density were significantly associated (p < 0.001). As the strongest correlate of cone density (p < 0.001) and best-corrected visual acuity (p < 0.001), EZ width is the most sensitive biomarker of structural and functional decline in USH2A retinopathy, rendering it a promising trial endpoint.  相似文献

Antimicrobial peptides (AMPs) are small proteins produced by epithelial surfaces and inflammatory cells, which have broad-spectrum antimicrobial and immunomodulatory activities. They are known to be important in a number of infectious and inflammatory conditions and have been shown to be present in a number of sites throughout the female reproductive tract. Inflammation and infection are associated with a number of complications of pregnancy including preterm labor, and AMPs may play a key role in maintaining and protecting pregnancy. Weanling rats were fed fat free diets supplemented with 10% added fatty acids so that dietary effects on bone marrow fatty acids could be determined. The addition or deletion of linoleic acid from the fatty acid supplement resulted in alterations of the fatty acid patterns of bone marrow lipids but to a lesser degree than in erythrocyte lipids. With myristic acid supplementation, increased amounts of stearic acid were found in the lipid fractions, the difference between the bone marrow and erythrocyte lipids being less marked than when linoleic acid was fed. The activities of the bone marrow lipases varied with the dietary treatment. When linoleic acid was fed, higher rates of hydrolysis were observed with saturated fatty acid substrates. The reverse occurred when saturated fatty acids were fed.  相似文献

Although the relevance problem in recommender systems, which typically refers to the similarity between the preference of the user and the items the system recommends, has been well studied, the issue of making recommendations in right orders has barely been mentioned before. The order defines the way in which items should be better consumed in relation to each other. The analysis of real-world data sets demonstrates strong item consumption patterns in the degree of order. In this paper, we propose a novel method to tackle the right-order recommendation problem based on a graph structure that incorporates the item consumption orders. The proposed method can combine relevance and order effects in recommendations. We attempt to recommend the relevant items in the consecutive steps within a user session so that the user’s selected items in the consecutive steps can be stringed together in a right order. The experimental evaluation conducted on three real-world data sets shows that the recommendation accuracy is improved by considering item consumption orders compared with the baseline recommendation methods. In addition, the study on right-order recommendation contributes to the exploration on recommendation appropriateness besides relevance.  相似文献
