This study was undertaken to search for potential use of crude bacterial pectinase enzyme produced from Bacillus subtilis grown on hazelnut shell hydrolysate in clarification of carrot juice and to optimize the enzyme load, pH and time using the Box–Behnken response surface methodology (RSM). The carrot juice was treated with the crude pectinase enzyme (5.60 U mL?1) at different concentrations (0.1–0.5%), pH (4–7), and time (2–6 h). The obtained enzyme was also compared with commercial fungal pectinase at identical conditions. RSM provided optimal clarification conditions of 0.5% (w/v) enzyme load, 7.0 pH, and 6 h of time estimating 100% clarity, whose experimental counterpart was 94.47 ± 0.01%. High values of coefficient of determination (R2 = 0.9631), predicted R2 (0.8989) and insignificant lack‐of‐fit (0.12) also showed that the model was successful in predicting % clarity for various combinations. This study also indicated that crude bacterial pectinase providing about 95% clarity is superior to commercial fungal pectinase, which gave 78% clarity under tested conditions, in terms of clarification ability for carrot juice.  相似文献

This paper presents an integrated approach to solve the buffer allocation problem in unreliable production lines so as to maximize the throughput rate of the line with minimum total buffer size. The proposed integrated approach has two control loops; the inner loop and the outer loop. While the inner loop control includes an adaptive tabu search algorithm proposed by Demir et al. [8], binary search and tabu search are proposed for the outer loop. These nested loops aim at minimizing the total buffer size to achieve the desired throughput level. To improve the efficiency of the proposed tabu search, alternative neighborhood generation mechanisms are developed. The performances of the proposed algorithms are evaluated by extensive computational experimentation, and the results are reported.  相似文献

Glioblastoma (GBM) is a barely treatable disease due to its profound chemoresistance. A distinct inter- and intratumoral heterogeneity reflected by specialized microenvironmental niches and different tumor cell subpopulations allows GBMs to evade therapy regimens. Thus, there is an urgent need to develop alternative treatment strategies. A promising candidate for the treatment of GBMs is AT101, the R(-) enantiomer of gossypol. The present study evaluates the effects of AT101, alone or in combination with temozolomide (TMZ), in a microenvironmental glioma stem cell niche model of two GBM cell lines (U251MG and U87MG). AT101 was found to induce strong cytotoxic effects on U251MG and U87MG stem-like cells in comparison to the respective native cells. Moreover, a higher sensitivity against treatment with AT101 was observed upon incubation of native cells with a stem-like cell-conditioned medium. This higher sensitivity was reflected by a specific inhibitory influence on the p-p42/44 signaling pathway. Further, the expression of CXCR7 and the interleukin-6 receptor was significantly regulated upon these stimulatory conditions. Since tumor stem-like cells are known to mediate the development of tumor recurrences and were observed to strongly respond to the AT101 treatment, this might represent a promising approach to prevent the development of GBM recurrences.  相似文献

The performance analysis of double shield TBMs in difficult ground conditions in the Gerede tunnel is presented in this study. The strength of the encountered formations along the tunnel route varied from medium strength (sandstone, limestone) to high strength (basalt). The total length of tunnels is 31.6 km, which was excavated by three double shield TBMs having diameter of 5.57 m. Literature studies are first carried out in order to review the difficult ground conditions and their impacts on mechanized tunnelling. Later, the project, geology, and the characteristics of the TBMs are given in detail. Then the factors affecting the performance of the TBMs, machine utilization, and operational parameters (torque, thrust) are discussed in detail. In the light of these facts, the main objective of this study is to describe the possible improvement methods to reduce the effect of these difficulties on TBM performance.  相似文献

The objective of this research paper is to assess the biosorption potential of almond shell residues for methyl orange dye. The pseudo-second-order kinetic model described the dye biosorption process with a good fitting. The relationship between the pseudo-second-order model constants and the biosorption performance was also evaluated. The equilibrium data fit well with the Langmuir isotherm model presenting that the biosorption was the monolayer coverage of dye on the biosorbent and the homogeneity of active sites for dye binding on the biosorbent surface. Based on the Langmuir model, a single-stage batch biosorber was also designed to predict the biosorbent mass for certain percentage dye removal. Besides, the standard Gibbs free energy change was also calculated to define the nature of biosorption process. These results revealed that the utilization of almond shell residues as dye biosorbent could be an interesting option from both environmental and economic point of view.  相似文献

In this study we consider the operational fixed job scheduling problem under working time limitations. The problem has several practical implications in both production and service operations; however the relevant research is scarce. We analyse pre-emptive and non pre-emptive versions of the problem and its special cases. We provide polynomial-time algorithms for some special cases. We show that the non pre-emptive jobs problem is strongly NP-hard, and propose a branch-and-bound algorithm that employs efficient bounding procedures and dominance properties. We conduct a numerical experiment to observe the effects of parameters on the quality of the solution. The results of our computational tests for the branch-and-bound algorithm reveal that our algorithm can solve the instances with up to 100 jobs in reasonable times. To the best of our knowledge our branch-and-bound algorithm is the first optimisation attempt to solve the problem.  相似文献
