Purpose : Zilongjin, a complementary Chinese herbal medicine, has been used to alleviate the adverse effects of chemotherapeutic drugs in cancer therapy. However, the mechanisms of anti‐cancer activity of Zilongjin are still largely unkonwn. Experimental design : First, the proteomic approach of combined 2‐DE and ESI‐MS/MS was used to investigate the effect of Zilongjin on the protein expression in MCF‐7 cells. Then, the differential expression of some proteins was confirmed by Western blot, cytoimmunofluoresecnce, and quantitative real‐time RT‐PCR analysis. Results : The identified proteins with differential expression, involved in such events as protein translation, cellular signal transduction, cytoskeleton formation and transportation, include seven downregulating proteins, such as Eukaryotic translation initiation factor 3 subunit I, Eukaryotic translation initiation factor 1A Y‐chromosomal, Ran‐specific GTPase‐activating protein, Ubiquitin‐conjugating enzyme E2 N, Tropomodulin‐3, Macrophage‐capping protein, and Tumor protein D52, as well as two upregulating proteins, HSP β‐1 and keratin18. Moreover, the differential expression of three proteins was confirmed. Conclusions and clinical relevance : (i) These results provide a new insight into the molecular mechanisms of Zilongjin on therapy for breast cancer. (ii) The application of the proteomic approaches will result in the more extended appreciation of Chinese medicine than those known at present.  相似文献

This paper illustrates a hierarchical generative model for representing and recognizing compositional object categories with large intra-category variance. In this model, objects are broken into their constituent parts and the variability of configurations and relationships between these parts are modeled by stochastic attribute graph grammars, which are embedded in an And-Or graph for each compositional object category. It combines the power of a stochastic context free grammar (SCFG) to express the variability of part configurations, and a Markov random field (MRF) to represent the pictorial spatial relationships between these parts. As a generative model, different object instances of a category can be realized as a traversal through the And-Or graph to arrive at a valid configuration (like a valid sentence in language, by analogy). The inference/recognition procedure is intimately tied to the structure of the model and follows a probabilistic formulation consisting of bottom-up detection steps for the parts, which in turn recursively activate the grammar rules for top-down verification and searches for missing parts. We present experiments comparing our results to state of art methods and demonstrate the potential of our proposed framework on compositional objects with cluttered backgrounds using training and testing data from the public Lotus Hill and Caltech datasets.  相似文献

Image guided computational fluid dynamics is attracting increasing attention as a tool for refining in vivo flow measurements or predicting the outcome of different surgical scenarios. Sharp interface Cartesian/Immersed-Boundary methods constitute an attractive option for handling complex in vivo geometries but their capability to carry out fine-mesh simulations in the branching, multi-vessel configurations typically encountered in cardiovascular anatomies or pulmonary airways has yet to be demonstrated. A major computational challenge stems from the fact that when such a complex geometry is immersed in a rectangular Cartesian box the excessively large number of grid nodes in the exterior of the flow domain imposes an unnecessary burden on both memory and computational overhead of the Cartesian solver without enhancing the numerical resolution in the region of interest. For many anatomies, this added burden could be large enough to render comprehensive mesh refinement studies impossible. To remedy this situation, we recast the original structured Cartesian formulation of Gilmanov and Sotiropoulos [Gilmanov A, Sotiropoulos F. A hybrid Cartesian/immersed boundary method for simulating flows with 3D, geometrically complex, moving bodies. J Comput Phys 2005;207(2):457–92] into an unstructured Cartesian grid layout. This simple yet powerful approach retains the simplicity and computational efficiency of a Cartesian grid solver, while drastically reducing its memory footprint. The method is applied to carry out systematic mesh refinement studies for several internal flow problems ranging in complexity from flow in a 90° pipe bend to flow in an actual, patient-specific anatomy reconstructed from magnetic resonance images. Finally, we tackle the challenging clinical scenario of a single-ventricle patient with severe arterio-venous malformations, seeking to provide a fluid dynamics prospective on a clinical problem and suggestions for procedure improvements. Results from these simulations demonstrate very complex cardiovascular flow dynamics and underscore the need for high-resolution simulations prior to drawing any clinical recommendations.  相似文献

Inverse gamma correction must be performed before displaying the received video signal because alternating current plasma display panel (AC PDP) has a linear output luminance response to a digital-valued input. At the same time contrast ratio enhancement is necessary for improving the image quality of display devices. The histogram equalization (HE) is an important contrast ratio enhancement method. But sometimes HE can produce unrealistic effects in images. In this paper, a new method of combining dynamic contrast ratio enhancement and inverse gamma correction for AC PDP is proposed. The dynamic contrast ratio enhancement and the inverse gamma correction are realized simultaneously in the proposed method. Furthermore the over-enhancement caused by the traditional HE can be avoided. A real-time image processor with the proposed method was designed and implemented. Simulations and experimental results on a 50-in. AC PDP show that the image quality of AC PDP can be improved obviously.  相似文献

In this paper, we formulate a stereo matching algorithm with careful handling of disparity, discontinuity, and occlusion. The algorithm works with a global matching stereo model based on an energy-minimization framework. The global energy contains two terms, the data term and the smoothness term. The data term is first approximated by a color-weighted correlation, then refined in occluded and low-texture areas in a repeated application of a hierarchical loopy belief propagation algorithm. The experimental results are evaluated on the Middlebury data sets, showing that our algorithm is the top performer among all the algorithms listed there.  相似文献

Single multiplicative neuron model is a novel neural network model introduced recently, which has been used for time series prediction and function approximation. The model is based on a polynomial architecture that is the product of linear functions in different dimensions of the space. Particle swarm optimization (PSO), a global optimization method, is proposed to train the single neuron model in this paper. An improved version of the original PSO, cooperative random learning particle swarm optimization (CRPSO), is put forward to enhance the performance of the conventional PSO. The proposed CRPSO, PSO, back-propagation algorithm and genetic algorithm are employed to train the model for three well-known time series prediction problems. The experimental results demonstrate the superiority of CRPSO-based neuron model in efficiency and robustness over the other three algorithms.  相似文献

More and more complicated conceptual design of ship’s engine room (CDSER) heavily depends on designers’ engineering knowledge and existing ship data. To achieve intelligent design at the initial ship design stage, many researchers have made much significant progress in this field, however, most of them only focused on how to find the similar constructed ships. At present, how to utilize these existing data remains an untouched topic. In order to make good use of the existing data and reduce the dependence on designers’ experience, a novel system named Expert System for Aided Conceptual Design of Ship’s Engine Room Automation (ESACD), is elaborated in this study. With the support of the constructed Ship Data Warehouse System, two core subsystems Configuration Selection Assistant (CSA) and Design Scheme Decision Assistant (DSDA) are included in ESACD. A promising approach integrating Fuzzy c-means algorithm (FCM) and Rough Sets Theory (RST) to extract configuration rules from the stored data is adopted in CSA. According to engineers’ proposals, RST is utilized to reason knowledge in incomplete scheme information systems for getting design scheme rules in DSDA, which are useful suggestions for engineers to get better schemes at this stage. Finally, the validity and necessity of this interactive expert system are demonstrated through the CDSER of a new 50,000 DWT Handymax bulk carrier. It is proved that ESACD can efficiently facilitate rapid and intelligent design in CDSER, and reduce the cost of a new ship design.  相似文献

In this paper, effects of the implant thread pitch on the maximum Von Mises stresses were evaluated in jaw bones and implant–abutment complex by a finite element method. The thread pitch ranged from 0.5 mm to 1.6 mm. Results suggested that under axial load, the maximum equivalent stresses in cortical bone, cancellous bone and implant–abutment complex decreased by 6.7%, 55.2% and 22.3%, respectively with the variation of thread pitch; and under buccolingual load, 2.7%, 22.4% and 13.0%, respectively. When thread pitch exceeded 0.8 mm, minimum stresses were obtained. Data indicated that cancellous bone was more sensitive to thread pitch than cortical bone did; thread pitch played a great role in protecting dental implant under axial load than under buccolingual load; and thread pitch exceeding 0.8 mm were optimal selection for a screwed implant by biomechanical consideration.  相似文献
