1. The aim of this study was to determine whether different signal transduction mechanisms underlie the Ca2+ sensitizing effects of guanosine 5'-O-(3-thiotriphosphate) (GTP(gamma)S) and receptor agonists on beta-escin-skinned smooth muscle of rabbit mesenteric artery. 2. In the homogenate of the beta-escin-skinned arterial strip, C3 exoenzyme of Clostridium botulinum catalyzed the [32P]-ADP-ribosylation of only one protein that had the same molecular mass as the protein detected in Western blots with anti-rho p21 antibody. Pretreatment of preparations with C3 resulted in great inhibition of GTP(gamma)S-induced Ca2+ sensitization, although the effect of GTP(gamma)S at higher concentrations (> or = 30 microM) was not completely blocked by this treatment. In contrast, the enhancement by phenylephrine and histamine, in the presence of guanosine 5'-triphosphate, of the Ca2+-induced contraction was not affected by C3 pretreatment. 3. The protein kinase C (PKC) inhibitors calphostin C and staurosporine completely eliminated the enhancement by phorbol ester 12,13-dibutyrate of the Ca2+-induced contraction. However, these PKC inhibitors had no effect on GTP(gamma)S- and receptor agonist-induced Ca2+ sensitization. 4. The tyrosine kinase inhibitors genistein and tyrphostin 25 caused an irreversible and complete block of the enhancement by GTP(gamma)S of the Ca2+-induced contraction without affecting this Ca2+ contraction. The inactive genistein analogue daidzein did not modify the effect of GTP(gamma)S. The Ca2+ sensitizing effects of phenylephrine and histamine were also blocked by these tyrosine kinase inhibitors. 5. These results suggest that rho p21 predominantly mediates GTP(gamma)S-induced Ca2+ sensitization of beta-escin-skinned smooth muscle of rabbit mesenteric artery, while the Ca2+ sensitizing actions of heterotrimeric G protein-coupled receptor agonists do not involve this small G protein. However, it seems that tyrosine phosphorylation, but not PKC activation, plays an important role in both of the rho p21 protein- and heterotrimeric G protein-mediated Ca2+ sensitization mechanisms.  相似文献

A mathematically simple spatial difference method has been applied for analysing three-dimensionally, and for illustrating graphically, the process of the development of a flame after the propane-air stoichiometric mixture is ignited. The calculated results show that the mathematical simulation can well express the process of mixture explosion in cylindrical vessels, and can evaluate the effects of laminar swirl flow on the flame development. It is concluded that the swirl motion deforms the flame front and accelerates the flame enlargement. The statistical properties of decaying compressible turbulence are investigated by direct numerical simulations of flow in a periodic cube. Starting with fully developed turbulence for various microscale Reynolds numbersR , rms Mach numbersM, and small- and large-scale compressive ratiosr CS andr CL , we find that the kinetic energy exhibits an exponential decay in time. Interactions between the compressive and rotational components of kinetic energy are weaker than self-interactions of the respective components. The energy spectrum of rotational component obeys the same Kolmogorov similarity law as for incompressible turbulence and forced compressible turbulence. The form of the energy spectrum of the compressive component, on the other hand, depends strongly onM. As the turbulence decays freely,R andM decrease in time butr CS andr CL tend to some universal values.  相似文献

Feed rate characteristics of a blow tank solids conveyor in which the conveying pipe exits at the upper part of the tank are experimentally examined by the use of polyethylene pellets having a large minimum fluidizing velocity. The flow pattern of solid particles in the tank resembles suction flow at the pipe inlet and bears a strong resemblance to that of gravity discharge of solids from an orifice, and there exists a dead solids region on the porous support through which compressed air is blown. Within this region pipe inlet height from the porous support and tank size have no effect on the feed rate. For a given tank, the feed rate increases with an increase in pipe diameter. An empirical equation is proposed to relate these variables.  相似文献

A poly(vinyl alcohol) (PVA) fiber support incorporating various aminoacetal functional groups has been developed for immobilizing enzymes. The aminated PVA fiber seems to adsorb enzymes with electrostatic force of attraction; thus the immobilization procedure is simple. By the use of this fiber having immobilized enzymes, the reaction between enzymes and substrates is nearly independent of the size of subtstrates. This newly developed type of fiber, which is formed by a mass superfine fibers (SFF), each measuring 1 μm or less in diameter, permits much more increased surface area than the conventional enzyme immobilization supports. Our studies of the properties of the fiber for immobilization of enzymes show the following results: (1) SFF has a greater ability for the immobilization of invertase than ordinary fibers; (2) dimethyl-aminated SFF has the best performance for the immobilization of invertase. From these results, it is concluded that the dimethyl-aminated SFF is an excellent support for the immobilization of invertase.  相似文献

Dynamic mechanical properties and adhesive strengths of power feed copolymer and random copolymer synthesized using styrene or methyl methacrylate and n-butyl acrylate were investigated. Although the two systems were synthesized from the same raw materials, power feed copolymer had a very broad transition compared with random copolymer. This fact was explained by the fact that the system synthesized through power feed method was an alloy of copolymers which are a continuous series from monomer A rich copolymers to monomer B rich copolymers. The dynamic mechanical behavior of film cast from solution was almost the same as that of emulsion film, which indicated more extensive application of power feed copolymer. In the P(nBA/St) system, power feed copolymer maintained its adhesive strengths over a wide temperature range compared with random copolymer. The absolute value, however, was not so high. This was due to the low cohesive strengths of the films.  相似文献

As previously reported, high surface area microporous carbons with long-range order can be synthesized by using zeolite Y as a template. In this work, an attempt is made to synthesize porous carbon using several other types of zeolites (zeolite β, ZSM-5, mordenite and zeolite L). Special attention is paid to whether the structural regularity of each zeolite can be transferred to the carbon structure as well as with the case of zeolite Y. The carbon filling method was then examined to see which gives the highest regularity to the carbon structure. It has been found that the optimum carbon filling method for zeolite Y is not an optimum one for the other zeolites and the degree of the regularity of long-range ordering in the carbons strongly depends on zeolite type. The order of the regularity in the resultant carbons is zeolite β>>zeolite L>mordenite>ZSM-5. The effect of zeolite type on the regularity is discussed in relation to the size and the shape of the zeolite channels.  相似文献

Power feed copolymers were synthesized using styrene and n-butyl acrylate through non-uniform feeding emulsion polymerization. Poly(vinyl alcohol) (PVA) was used as a protective colloid, onto which vinyl monomers were grafted. Power feed copolymer had a very broad glass transition temperature compared with random copolymer, even if grafting and/or crosslinking were introduced to the system. This tendency was almost the same as the non-grafted power feed copolymer where only low molecular weight surfactant was used.

Adhesive joint strengths of power feed copolymers were evaluated compared with random copolymers. In the case of usual linear power feed copolymer, the adhesive joint strengths were not higher than those of random copolymer, which was considered to be due to the lower film strengths of the power feed copolymer. Power feed copolymer having grafting showed slightly higher adhesive joint strengths over a wide range of temperatures than random copolymer. When crosslinking was introduced to the system, power feed copolymer showed much higher adhesive joint strengths over a wide temperature range.  相似文献

The polymers 3 having pyridine moieties in the main chain were synthesized by the reaction of the organocobalt polymers 1 having cobaltacyclopentadiene moieties in the main chain with various nitriles 2. When brown colored 1 and excess 2 were heated in tetrahydrofuran at 80 °C for 24 h and then at 150 °C for 12 h in a sealed tube, the polymers 3 were obtained by the precipitation with methanol in good yields. From the spectroscopic measurements, the resulting polymers 3 were found to contain 35 – 90% of the pyridine moieties depending on the structures of 1. Styrene was grafted onto cellulose acetate p-nitrobenzoate (CANB) by chain transfer reaction of growing polymer radicals to the pendant nitro groups of CANB. A copolymer with a branch for every 17.2 nitro groups was obtained. This result indicates that the pendant aromatic nitro group is more effective in obtaining a graft copolymer by radical mechanism than pendant double bond on the trunk polymer previously reported, where graft copolymers with a branch for several hundred of double bonds are produced.  相似文献
