Oral droplet formulations of clonazepam (CZ) were developed to examine their potentials as an alternative to i.v. administration for the treatment of acute epileptic seizures. Propylene glycol containing 2.5% (wt/wt) CZ with or without 5.0% (wt/wt) oleic acid (OA) was prepared as a solution by heating at 90 degrees C and subsequently lowering the temperature to 30 degrees C. The droplet (20 microL) was administered to the oral cavity between the lower gum and bottom lip before CZ precipitation started. With a droplet of propylene glycol loaded with 2.5% (wt/wt) CZ and 5.0% (wt/wt) OA, the plasma concentration reached 20 ng/mL (minimal effective concentration) within 10 min and was maintained between 20 and 60 ng/mL, less than a toxic level, for a period of 60 min. For a droplet of propylene glycol loaded only with CZ at 2.5% (wt/wt), it took more than 15 min for the plasma concentration to reach 20 ng/mL. It is suggested that a droplet of CZ/OA/propylene glycol (2.5:5.0:92.5, wt/wt) might be useful as an alternative to i.v. injection of CZ for the treatment of acute epileptic seizures.  相似文献

Ferrimagnetic materials can be expected to be useful as thermo seeds for hyperthermic treatment of cancer, especially where the cancer is located in deep parts of body, as they can generate heat by magnetic hysteretic loss when they are placed in an alternating magnetic field. Recently, it was reported that ferrimagnetic maghemite (γ-Fe2O3) microspheres 20–30 μm in diameter prepared in aqueous solution can show excellent heat generating ability. However, these microspheres have many cracks on their surfaces. In this study, the preparation conditions for the microspheres was further optimized in order to obtain crack-free ferrimagnetic microspheres, and the in vitro heat generation of the obtained microspheres was measured in an agar phantom under an alternating magnetic field. Crack-free γ-Fe2O3 microspheres 20–30 μm in diameter were obtained successfully. Their saturation magnetization and coercive force were 68 emu g−1 and 198 Oe, respectively. Their heat generation under an alternating magnetic field of 300 Oe at 100 kHz was estimated to be 42 W g−1. The microspheres showed in vitro heat generation when they were dispersed in an agar phantom and placed under an alternating magnetic field. It is believed that these microspheres may be useful for the in situ hyperthermic treatment of cancer.  相似文献

We demonstrate a comprehensive study of self-assembled molecular nanowire, including molecular design, one-dimensional crystal growth, resistivity measurement of individual wire, and application to a field-effect transistor. Appropriate molecular design and control of interfacial interactions lead to single crystalline wire growth with an extensive pi-stacking motif. Resistivity measurements of an individual molecular wire indicate that these structural features are advantageous for electrical transport. Finally, field-effect transistors with single- and double-wire channels were fabricated to give some indication of the potential application of the molecular wires.  相似文献

River flood damages, worldwide, have increased dynamically in the last few decades, so that it is necessary to interpret this change. River flooding is a complex phenomenon which can be affected by changes coupled to terrestrial, socio-economic and climate systems. The climate track in the observed changes is likely, even if human encroaching into the harm’s way and increase in the damage potential in floodplains can be the dominating factors in many river basins. Increase in intense precipitation has already been observed, with consequences to increasing risk of rain-induced flooding. Projections for the future, based on climate model simulations, indicate increase of flood risks in many areas, globally. Over large areas, a 100-year flood in the control period is projected to become much more frequent in the future time horizon. Despite the fact that the degree of uncertainty in model-based projections is considerable and difficult to quantify, the change in design flood frequency has obvious relevance to flood risk management practice. The number of flood-affected people is projected to increase with the amount of warming. For a 4°C warming the number of flood-affected people is over 2.5 times higher than for a 2°C warming. The present contribution addresses the climate track in an integrated way, tackling issues related to multiple factors, change detection, projections, and adaptation to floods.  相似文献

Contact damage in materials is critical in engineering applications because it influences mechanical resistance, such as wear, erosion, and impact failure. Indentation tests were performed using a tungsten carbide ball indenter (Hertzian contact) on the surfaces of glass–ceramics containing hexagonal CaAl2Si2O8 or mica crystals (fluorophlogopite), both of which have a layered structure. The stress–strain relation and the permanent deformation on the surface, as well as the observation of the microcrack zone by X-ray computed tomography using synchrotron radiation, revealed that the glass–ceramic with hexagonal CaAl2Si2O8 showed ductility similar to the quasi-plastic behavior previously observed in the mica glass–ceramic. The yield stresses of the glass–ceramics were estimated from the stress deviating from the stress–strain relation assuming complete elastic response between the ball and the sample. The ratio of the yield stress to Young modulus (Y/E) of the glass–ceramic with hexagonal CaAl2Si2O8 was determined to be higher than that of the mica glass–ceramic.  相似文献
