This paper introduces a novel nonlinear extension of Fisher's classical linear discriminant analysis (FDA) known as high-order Fisher's discriminant analysis (HOFDA). The ability of the new method to capture nonlinear relationships stems from its use of an extended polynomial space constructed out of the original features. Furthermore, a genetic algorithm (GA) is used in order to incrementally generate an optimal subset of polynomial features out of an initial pool of minimal discriminants. This procedure yields surprisingly compact discriminants with state of the art recognition rates for the difficult UCI thyroid classification problem. 相似文献

This study compares the performance of 4 antimicrobial residue screening tests [brilliant black reduction test AiM (Analytik in Milch Produktions- und Vertriebs GmbH, München, Germany), Delvotest MCS (DSM Food Specialties, Delft, the Netherlands), Eclipse 100 test (ZEU-Inmunotec SL, Zaragoza, Spain), and Copan Milk Test (Copan Italia S.p.a., Brescia, Italy)] used to detect 20 antimicrobial agents (aminoglycosides, macrolides, tetracyclines, sulfonamides, and quinolones) in goat's milk, according to International Dairy Federation guidelines. Composite milk samples from 30 antibiotic-free goats were used to prepare spiked milk samples and 11,520 analytical determinations were carried out. According to a logistic regression model, agreement coefficients were greater than 98% for most of the antibiotics, with higher b values obtained for macrolides. Neither tetracyclines nor quinolones were detected at European Union maximum residue limits. Only the Copan Milk Test and the Delvotest MCS were able to detect 3 antimicrobials below their maximum residue limits (neomycin, tylosin, and sulfadimethoxine). Given that these tests are used in control programs for goat's milk, our results indicate their sensitivity would need to be improved to guarantee safety for consumers. 相似文献

The primary objective of this study was to determine whether the development of behavioral sensitization to the putative dopamine D3 receptor agonist 7-OH-DPAT could be prevented by either selective D1-type or D2-type dopamine receptor antagonists. In three experiments, male Wistar rats (250-350 g) were given seven to nine injections (at 48-h intervals) of 7-OH-DPAT (1.0 mg/kg, SC) or vehicle in combination with the D2-type dopamine antagonist eticlopride (0.3 mg/kg, SC), the D1-type dopamine antagonist SCH 23390 (0.1 or 0.2 mg/kg, SC), or vehicle. After the injections, the rats were tested for locomotor activity in photocell arenas for 2 h. In the first two experiments, after seven injections, all rats were tested for activity following vehicle injections to test for possible conditioning effects. In each experiment, after the last pre-exposure session, all rats were given a challenge injection of 7-OH-DPAT (1.0 mg/kg, SC) and tested for activity. Major findings were as follows: a) 7-OH-DPAT treatments produced a progressively greater increase in locomotor activity with repeated treatment; b) concurrent treatment with eticlopride or SCH 23390 (0.1 and 0.2 mg/kg) blocked the acute locomotor-activating effects of 7-OH-DPAT across days; c) eticlopride, but not SCH 23390, completely blocked the development of behavioral sensitization to 7-OH-DPAT. Although the low dose of SCH 23390 (0.1 mg/kg) produced a partial attenuation of sensitization, the higher dose (0.2 mg/kg) of SCH 23390 appeared to augment, rather than block, sensitization to 7-OH-DPAT; d) rats previously treated with SCH 23390 (0.2 mg/kg, but not 0.1 mg/kg) without 7-OH-DPAT displayed a hyperactive response to the 7-OH-DPAT challenge injection; and e) after vehicle injections, rats previously given 7-OH-DPAT, SCH 23390, or eticlopride either alone or in combination were more active than vehicle control rats. These findings suggest that the neurochemical mechanisms mediating the development of behavioral sensitization to 7-OH-DPAT may differ from those of other dopamine D2-type agonists such as quinpirole or bromocriptine. Moreover, these results demonstrate that hyperactivity responses following vehicle injections in drug-pretreated animals do not necessarily reflect conditioning mechanisms. 相似文献

Supercritical fluids have been extensively used for particle production of many natural and pharmaceutical substances providing useful alternatives for pharmaceutical and nutraceutical particulate system formulation. Among the different methods, the gas or supercritical antisolvent (GAS or SAS) process and its variants, have received a considerable interest due to the wide range of materials that can be micronized. Controlling particle formation in order to nucleate small particles is a key issue in GAS and SAS processes and this is directly related to mixing at all scales. In this work, we focus on numerical simulation of the process, emphasizing mixing modeling. Different mixing devices characterized by different nozzles are analyzed, to get an insight into mixing dynamics and its influence on the final particle size distribution. Results show that mixing is determinant in obtaining small particles, and that mixing at the microscale is a significant parameter to account for in the proper design of precipitators. © 2011 American Institute of Chemical Engineers AIChE J, 2012 相似文献

Tricalcium silicate does not undergo hydration at relative humidities (RH's) below 80%. But, the rate at which its hydration rate decreases as a function of the RH has not yet been elucidated. By invoking correspondence between RH and water activity (aH, unitless), both of which are related to the chemical potential of water, the reaction evolutions of triclinic tricalcium silicate (i.e., T1‐Ca3SiO5 or C3S) are tracked in water + isopropanol (IPA) mixtures, prepared across a wide range of water activities. Emphasis is placed on quantifying the: (a) rate of hydration as a function of aH, and (b) the critical (initial, aH0c or the achieved) water activity at which hydration effectively ceases, i.e., does not progress; here identified to be ≈ 0.70. The hydration of tricalcium silicate is arrested even when the system remains near saturated with a liquid phase, such that small, if any, capillary stresses develop. This suggests that changes in chemical potential induced via a vapor‐phase or liquid‐phase route both induce similar suppressions of C3S hydration. A phase boundary nucleation and growth (pBNG) model is fit to measured hydration rates from the onset of the acceleration period until well beyond the rate maximum when the water activity is altered. The simulations suggest that for a fixed hydrate nucleation density, any water activity reductions consistently suppress the growth of hydration products. Thermodynamic considerations of how water activity changes may influence reactions/hydrate evolutions are discussed. The outcomes improve our understanding of the chemical factors that influence the rate of Ca3SiO5 hydration. 相似文献

Plastic waste increases at a rate of several tons per year and the time required for their biodegradation is often unknown. The Phanerochaete chysosporium fungus oxidizes a wide variety of toxic organic compounds to nontoxic compounds or CO2 and H2O. The biodegradability of the poly(vinyl alcohol) (PVOH) caused by this fungus was analyzed by gel permeation chromatography (GPC) with differential refractive index (Δn) and ultraviolet (UV) detectors. During the GPC analyses, the PVOH is in contact with the components of the culture medium. These components can interact with the stationary phase and also with the polymer producing erroneous results in the molecular weight determination. Therefore, we compare results using differnt mobile phases. A phosphate-acetate buffer of pH 7.4 was found to be appropriate mobile phase to follow the degree of degradation of the PVOH by the change in the average molecular weight determined by GPC in this culture medium. UV detection enabled us to identify structural changes associated with transformations of the carbonyl groups, mainly to enolized β-diketones groups. Infrared absorption (FTIR) confirmed these changes and also showed important modifications in the C-O stretching region probably due-to apertures of cyclic ether groups or transformations of the OH groups by oxidation reactions. 相似文献

This study was conducted to compare the detection limits (DL) of several antibiotic residue screening tests with the maximum residue limits (MRL) authorized by the EU according to the guidance for the standardized evaluation of microbial inhibitor tests of the International Dairy Federation. Composite antibiotic-free milk samples from 30 primiparous Murciano-Granadina goats in good health condition were used to prepare test samples spiked with different concentrations of each antimicrobial. In total, 5,760 analytical determinations of 10 β-lactam antibiotics (penicillin-G, ampicillin, amoxicillin, cloxacillin, oxacillin, dicloxacillin, cefadroxyl, cefalexin, cefoperazone, and cefuroxime) were performed using 4 antibiotic residue screening tests: the brilliant black reduction test BRT AiM (AiM-Analytik in Milch Produktions-und Vertriebs GmbH, München, Germany), Delvotest MCS (DSM Food Specialties, Delft, the Netherlands), Eclipse 100 (ZEU-Inmunotec SL, Zaragoza, Spain), and the Copan Milk Test (CMT; Copan Italia SpA, Brescia, Italy). For each method, we estimated the detection limits of the antimicrobial agents using a logistic regression model. Using the CMT and Delvotest on samples spiked with the 8 antibiotics for which MRL were available, DL were at or below the MRL. The BRT test provided DL at or below the MRL for all of the agents except cefalexin, whereas the Eclipse 100 method failed to detect 4 antibiotics (ampicillin, amoxicillin, cloxacillin, and cefoperazone) at MRL or below. Logistic regression-determined levels of agreement were highest for the CMT method (98.6 to 100%) and lowest for Eclipse 100 (66.3 to 100%). In general, agreement levels indicated good correlation between observed results and those predicted by logistic regression. The lowest b values (closely related to test sensitivity) were recorded for the cephalosporins (0.074 to 0.430) and highest for penicillin G, ampicillin, and amoxicillin (11.270 to 11.504). Delvotest and CMT best fulfilled IDF criteria for the ideal test for detecting antibiotic residues in milk. 相似文献

Heat-resistant cast steels are highly sensitive to cracking as they are weld-repaired because of their very low ductility. To prevent weld-repair cracking of three different heat-resistant cast steels used for the manufacturing of superplastic forming (SPF) dies, the effect of various welding parameters, such as the choice of the filler material, the number of weld passes and the pre-heating temperature has been investigated. The choice of an appropriate filler metal and the pre-heating to 400 °C of the material prior to welding drastically lower the propensity to cracking, but remain unable to eliminate cracks entirely. To further reduce weld-repair cracking and hopefully prevent it completely, a buttering technique has been developed. Buttering of the base metal surface with nickel alloys before weld-repair has been shown to prevent cracking of the base metal, but results in some hot-cracking of the buttering layer itself. On the other hand, buttering with Ni–Fe alloys, less sensitive to hot-cracking, results in crack-free weld-repairs. 相似文献

Engineered luciferase-luciferin pairs have expanded the number of cellular targets that can be visualized in tandem. While light production relies on selective processing of synthetic luciferins by mutant luciferases, little is known about the origin of selectivity. The development of new and improved pairs requires a better understanding of the structure−function relationship of bioluminescent probes. In this work, we report a biochemical approach to assessing and optimizing two popular bioluminescent pairs: Cashew/d -luc and Pecan/4′-BrLuc. Single mutants derived from Cashew and Pecan revealed key residues for selectivity and thermal stability. Stability was further improved through a rational addition of beneficial residues. In addition to providing increased stability, the known stabilizing mutations surprisingly also improved selectivity. The resultant improved pair of luciferases are >100-fold selective for their respective substrates and highly thermally stable. Collectively, this work highlights the importance of mechanistic insight for improving bioluminescent pairs and provides significantly improved Cashew and Pecan enzymes which should be immediately suitable for multicomponent imaging applications. 相似文献
