Objective: Quantification of the cerebrospinal fluid (CSF) flow through the aqueduct of Sylvius by means of magnetic resonance imaging (MRI) is subject to interobserver variability due to the region of interest (ROI) selection. Our objective is to develop a semiautomatic measurement method to achieve reproducible quantitative analysis of CSF flow rate and stroke volume. Material and methods: MR examinations were performed using a 1.5 T scanner with a phase contrast sequence (velocity encoding [V
enc] of 20 cm/s, FOV = 160, 3 mm slice thickness, image matrix size = 256×256, TR = 53 ms, TE = 11 ms, NSA = 2, flip angle = 15° and 23 frames per cardiac cycle with peripheral retrospective pulse gating). Our method was developed using MATLAB R7. Errors introduced by background offset and possible aliased pixels were automatically detected and corrected if necessary in order to calculate the flow parameters that characterize CSF dynamics. The semiautomatic seed method reproducibility was evaluated and compared with the radius method by two observers analysing 21 healthy subjects. Results: The measurements using the semiautomatic seed method reduced the interobservers variability (intra-class correlation [ICC] = 1.0 for stroke volume and for volumetric flow rate) versus the radius method (ICC = 0.46 for stroke volume and 0.65 for flow rate). Normal stroke volume (39.19 ± 20.13 μl/cycle), flow rate (3.81 ± 2.81 ml/min), maximal mean systolic velocity (5.27 ± 1.3 cm/s) and maximal mean diastolic velocity (4.20 ± 1.4 cm/s) were calculated with the half moon and aliasing corrected seed method. Conclusions: Semiautomatic measurements (seed method with half moon background and aliasing correction) allow a generalization of the calculus of flow parameters with great consistency and independency of the operator. 相似文献

Feline immunodeficiency virus (FIV) is a naturally occurring pathogen that causes an AIDS-like syndrome in domestic cats and is a valuable model system by which criteria for antiviral vaccines and drugs development can be tested. The cell-entry step of the lentivirus life cycle is regarded as a promising target for the development of new generation inhibitors. We have previously described potent in vitro anti-FIV activity associated with a synthetic octapeptide, termed C8 (Ac-Trp-Glu-Asp-Trp-Val-Gly-Trp-Ile-NH2), containing the Trp-rich motif of FIV transmembrane glycoprotein, which shares a common structural framework with the corresponding molecule of HIV and appears to play a similar role in cell entry. In this report, in an attempt to develop simpler potential fusion inhibitors to be tested in vivo, we describe further studies focused on synthetic peptide analogues of C8. Since C8 inhibitory activity is dependent upon the Trp motif, we systematically replaced these residues with bulky and/or aromatic natural and unnatural amino acids, in order to develop a rational structure-activity relationship. Furthermore, the amino acids located between the Trp residues, which are not crucial for inhibitory activity, were replaced by simple alkyl spacers of appropriate length. Design, NMR structural analysis, in vitro anti-FIV activity in lymphoid cell cultures, and serum stability of these new analogues are reported. The final results indicate that a simpler hexapeptide (Ac-Nal2-Ape-Nal2-Ape-Nal2-Ile-NH2; Nal2 = 3-naphthalen-2-yl-L-alanine, Ape = 5-aminopentanoic acid), almost entirely made up of unnatural amino acid residues, has markedly increased enzymatic stability, while maintaining strong antiviral potency in vitro. 相似文献

Herein, we report the synthesis of novel stable analogues of geranylgeranyl diphosphate (GGPP), in which the "natural" all-trans geranylgeranyl portion has been replaced by a (Z,E,E)-geranylgeranyl chain. The change in configuration and consequent change in the relative position of the polar portion with the lipophilic side chain did not improve the properties of the E,E,E analogues in their inhibition of geranylgeranyl protein transferase I (GGTase I). However, a significant level of GGTase I inhibition and selectivity for GGTase I over farnesyl transferase (FTase) was maintained the unsubstituted phosphonoacetamidoxy derivative 4 a. This has shed light on the relative importance of the configuration at the C2=C3 double bond among GGPP derivatives. Moreover, the biological activities of all the compounds reported herein, in particular the preferential FTase inhibitory activity shown by compound 6, were in good agreement with the results of docking analysis. 相似文献

A FEM model of a conventional hot dipping fluidized bed (FB) coating process on a low carbon steel rod using a PPA thermoplastic powder was developed. Two computational domains, the former for the metal substrate and the growing coating and the latter for fluidized bed emulsion nearby the metal substrate were set. A novel recursive procedure was employed to solve the ‘coupled’ thermal problem. Besides, realistic values of polymer thermal properties according to temperature as well as the whole curve of polymer enthalpy were input to the model so as to increase its reliability.
As a result, consistent trend of coating thickness and growth rate according to dipping time with experimental data and theoretical considerations were calculated. Moreover, the calibration procedure of FEM model took to realistic values of heat transfer coefficient according to preheating temperature of substrate as the comparisons with available experimental data confirm. At last, dimensionless analysis stated once more the FEM model reliability by comparing the numerical findings with those of pioneer models available in literature. 相似文献

Sustainability is a key concept for our future and the role of thermodynamics in its assessment is fundamental. The use of energy and matter must be considered not only from a microscopic viewpoint (the use of a single fuel or material, or the presence of a single pollutant) but also by means of holistic approaches able to synthesize all the characteristics of a single process. Exergy is a suitable function for this purpose. The exergy concept can also be applied to natural systems and to systems at the interface between natural and artificial ones. In this context also emergy can express very helpful indications. Four different efficiency indices are here examined to better understand different aspects of the sustainability of processes and systems. The effect of thermal processing and the berry cultivar has been the subject of several studies, to serve as a basis for the least possible loss of bioactive compounds and antioxidant capacity. In Brazil, there are numerous blackberry cultivars under cultivation, and in this context, the aim of this work was to study how the bioactive compound contents and antioxidant capacity of blackberry jelly were influenced by the cultivars (Guarani, Brazos, Comanche, Tupy, Cherokee, Caingangue and Choctaw). Analysis was made of 7 fresh blackberry cultivars and 7 blackberry jelly formulations. Analysis of antioxidant activity, total phenolics, total anthocyanins and ascorbic acid was performed. It was verified that the contribution of the heat sensitive antioxidants compounds is considerable in all blackberry cultivars, and the degradation of these compounds was significantly different between the different blackberry cultivars. The Brazos and Caingangue cultivars stand out as undergoing the lowest losses in processing, resulting in jellies richer in bioactive compounds and higher antioxidant capacity. 相似文献
