The degradation of diuron and 3,4-dichloroaniline (DCA) has been studied at boron-doped diamond (BDD) anode. A three electrode impinging jet cell was used to improve the mass transfer of the reactant to the electrode surface. In the whole range of experimental conditions examined, the reactants were completely mineralised and passivation of the electrode surface was not observed. HPLC and ionic chromatography analyses revealed that the reaction involves the oxidative ring-cleavage and the formation of stoichiometric amounts of chloride and ammonium ions. Depending on the specific experimental conditions, short-chained organic acids were also identified as intermediate products. Values of global current efficiencies greater than 20% were obtained even operating with low reactant concentration (0.17 mM for diuron and 2 mM for dichloroaniline) provided that a suitable ratio between applied current and mass transfer rate was guaranteed. 相似文献

Phagocytosis involves interactions between cell-surface receptors and the actin-based cytoskeleton. Plasma membrane glycoproteins cosedimenting with detergent-insoluble cytoskeletons were postulated to be phagocytosis receptor candidates of the unicellular slime mold Dictyostelium discoideum. A 130-kDa glycoprotein (gp130) was associated with cytoskeletons of bacterially but not axenically grown cells, suggesting a cytoskeletal interaction that depended on nutrient conditions. Labeling of gp130 with a membrane-impermeant biotinylating reagent showed it was surface-exposed and provided a tag that was used to monitor gp130. Biotin-labeled gp130 was an integral protein and found to be a single species by two-dimensional gel electrophoresis. An antibody was raised against a synthetic octapeptide corresponding to internal amino acid sequence of biotin-labeled gp130 enriched through avidin affinity chromatography. The affinity-purified antibody was monospecific, reacting with both axenic and bacterial forms of gp130 on immunoblots. There was less gp130 in plasma membranes of bacterially grown cells than in plasma membranes of axenically grown cells, which was consistent with the idea that as a receptor, gp130 would be internalized during phagocytosis and fewer molecules would be on the cell surface of actively feeding cells. This suggestion was supported by the observation that there was a reduced amount of surface (biotin)-labeled gp130 on bacterially grown cells relative to axenically grown cells. gp130 also was implicated in phagocytosis through immunoblotting analyses that revealed smaller versions of gp130 in plasma membranes of phagocytosis mutant HV29 (Vogel et al.J. The role of autophagy is known to be highly complex and context-dependent, leading to both cancer suppression and progression in several tumors including melanoma, breast and prostate cancer. In the present review, recent advances in an understanding of the involvement of autophagy in prostate cancer treatment are described. The regulatory effects of androgens on prostate cancer cell autophagy are particularly discussed in order to highlight the effects of autophagy modulation during androgen deprivation. A critical evaluation of the studies examined in the present review suggests the attractive possibility of autophagy inhibition combined with hormonal therapy as a promising approach for prostate cancer treatment. 相似文献

IgE-mediated allergy to soybean may be connected to an incomplete protein digestion causing an inappropriate immune response in the gut. The aim of this study was to gain insight on the digestibility and immunoreactivity of β-conglycinin and its deglycosylated form. To achieve this goal both proteins were digested mimicking gastrointestinal physiological conditions. Digests were characterised by SDS–PAGE, MALDI-TOF-MS and immunological assays. Data on IgG binding western blot provided key information related to the composition of the digests indicating that whereas deglycosylated β-conglycinin was completely removed by gastrointestinal digestion, α and β subunits of β-conglycinin glycoprotein partially survive the process of digestion. Such differential digestive behaviour might be influenced by glycan moieties, at least to some extent, and should be an additional factor to be considered on its potential allergenicity. Results also support the existence of linear epitopes in the three subunits (α, α′, and β) of both protein forms. 相似文献

Soybean germ oil (SGO) could take an important place among nutraceuticals if the finding were confirmed that a daily intake of SGO helps to maintain a normal level of serum cholesterol. Adapting the rice milling technology to the soybean refining process, we achieved a rapid and efficient separation of soybean bran and cotyledon from the hypocotyle. Our soybean germ was no less than 95% hypocotyle. SGO obtained from it by typical hexane extraction was very rich in tocopherols (total content 4.35 g/kg) and phytosterols (β‐sitosterol 57.4%, Δ‐7‐stigmastenol 20.2%, Δ‐7‐avenasterol 6.8%, stigmasterol 6.2%, campesterol 5.4%, Δ‐7‐campesterol 1.2%, Δ‐5‐avenasterol 0.95%, etc.; total sterols 44.3 g/kg). The most prominent fatty acids were linoleic (56.2%), linolenic (15.5%) and oleic acid (10.6%). Although isoflavones abound in soybean germ (17.7 g/kg), only traces of them passed into the hexane extract (7 mg/kg). In murine 3T3 fibroblasts, SGO was found to reduce the incorporation of [14C]acetate into sterols, by inhibiting oxidosqualene cyclase. 相似文献

Four Lactobacillus helveticus strains were studied for proteolytic capacity and general aminopeptidase (AP) and X-Pro dipeptidyl aminopeptidase (DAP) activity. The rate of hydrolysis and the activity against synthetic substrates with N-terminal residues of Arg, Lys, Leu, Glu or Pro, varied markedly among the strains. The X-Pro DAP activity was consistently high. The crude cell-wall and cytoplasm extracts from strain Lb. helveticus ISLC59 were analysed thoroughly for their proteolysis ability by using four synthetic peptide substrates, including alpha(s)1-CN(f1-23). Peptides formed during in vitro hydrolysis of the synthetic substrates by cell wall and cytoplasm preparations were identified by LC-ESI/MS. In doing so, it was possible to infer a prevalent endopeptidase activity splitting Lys7-His8 and Gln13-Glu14 bonds in the cytoplasm, and to deduce a secondary activity, which hydrolysed Glu14-Val15, Leu16-Asn17, Glu18-Asn19 and Lys3-His4 bonds lacking in the cell-wall. The presence of exopeptidases, as mainly AP, DAP, and carboxypeptidase (CPase) was deduced from the formation of several N- and C-terminally truncated peptides sets. The AP activity was higher in the cell-wall layer, where CPase activity was absent. The in vitro assays with cell extracts of the Lb. helveticus ISLC59 strain revealed extensive exopeptidase and endopeptidase activities. In several cases, the hydrolytic system of Lb. helveticus that splits in vitro alpha(s)1-CN(f1-23) peptide bonds was similar to that of Lactococcus lactis. Permeation tubes are convenient analyte sources for generating standard gas mixtures (containing, in particular, volatile organic compounds) used in the calibration of analytical instruments. For small permeation rates, corresponding to trace levels of analytes, the calibration of permeation tubes is time-consuming. The use of thermogravimetry as a means to measure rapidly the weight loss at constant temperature was investigated. An attempt to apply this technique to calibrating a permeation tube filled with benzene is described. In the 20 ng/min range of permeation rate, day-to-day variations of <5% were observed. The continuous weight loss monitoring of permeation tubes by thermogravimetry allows their rapid characterization. 相似文献

In the behavioral despair (forced swimming) test and in the tail-suspension test, long-term (30–32 days) castration significantly increased the duration of immobility in mice. Testosterone propionate (1 or 10 mg/kg/day–2 sc for 4 days), although not affecting the duration of immobility in sham-operated mice, reduced the duration of immobility in castrated mice to within normal limits. Desipramine (20 mg/kg ip) decreased the duration of immobility both in sham-operated and in castrated animals. These results indicate that castration favors an inactive behavior and that testosterone, although having no "antidepressant" effect per se, is necessary for the male animal to cope normally with adverse environmental situations. Among the many metrics used to compute the colour rendition of a light source which have been investigated in recent years, most consider only the spectral power distribution of the light source being tested. To highlight aspects not usually considered in the development of colour rendition metrics, and to propose future directions for alternative approaches consistent with human visual perception, an experiment was conducted to examine colour appearance variations on certain spatial visual content configurations, in other words the effects of illusions under different light sources. Results indicate that stronger simultaneous contrast configurations have a more unpredictable variation in appearance. Therefore, it is proposed that colour spatial distribution should be considered for the future generation of rendering metrics. 相似文献
