Wnt signaling is a fundamental pathway that drives embryonic development and is essential for stem cell maintenance and tissue homeostasis. Dysregulation of Wnt signaling is linked to various diseases, and a constitutively active Wnt pathway drives tumorigenesis. Thus, disruption of the Wnt response is deemed a promising strategy for cancer drug discovery. However, only few clinical drug candidates that target Wnt signaling are available so far, and new small‐molecule modulators of Wnt‐related processes are in high demand. Here we describe the synthesis of small molecules inspired by withanolide natural products by using a pregnenolone‐derived β‐lactone as the key intermediate that was transformed into a δ‐lactone appended to the D‐ring of the steroidal scaffold. This natural‐product‐inspired compound library contained potent inhibitors of Wnt signaling that act upstream of the destruction complex to stabilize Axin in a tankyrase‐independent manner.  相似文献

The aim of this study is to investigate the process of friction stir welding (FSW) by using finite element method (FEM). Currently, the materials that are difficult to be joined with conventional fusion methods can now be easily joined with the method of friction stir welding. In this paper, the welding capability of many different materials with this method has been investigated by using analytical and numeric methods. In this study, a finite element (FE) model was developed for welding process with friction stir welding of AZ31 magnesium alloy. This model was performed by the software of DEFORM 3D finite element in 960, 1,964, and 2,880 rpm rotational speeds and in 10 and 20 mm?min?1 transverse speeds. The temperature values taken from experiments and the temperature values with FEM are compared, and according to these results, it can be stated that the FE model gives reasonable results with experimental results based on temperatures values. Hence, the FE model can be used to predict other parameters of FSW process in future studies.  相似文献

Autoignition characteristics of an alternative (non-petroleum) and two conventional jet fuels are investigated and compared using a heated rapid compression machine. The alternative jet fuel studied is known as “S-8”, which is a hydrocarbon mixture rich in C7-C18 linear and branched alkanes and is produced by Syntroleum via the Fischer-Tropsch process using synthesis gas derived from natural gas. Specifically, ignition delay times for S-8/oxidizer mixtures are measured at compressed charge pressures corresponding to 7, 15, and 30 bar, in the low-to-intermediate temperature region ranging from 615 to 933 K, and for equivalence ratios varying from 0.43 to 2.29. For the conditions investigated for S-8, two-stage ignition response is observed. The negative temperature coefficient (NTC) behavior of the ignition delay time, typical of higher order hydrocarbons, is also noted. Further, the dependences of both the first-stage and the overall ignition delays on parameters such as pressure, temperature, and mixture composition are reported. A comparison between the autoignition responses obtained using S-8 and two petroleum-derived jet fuels, Jet-A and JP-8, is also conducted to establish an understanding of the relative reactivity of the three jet fuels. It is found that under the same operating conditions, while the three jet fuels share the common features of two-stage ignition characteristics and a NTC trend for ignition delays over a similar temperature range, S-8 has the shortest overall ignition delay times, followed by Jet-A and JP-8. The difference in ignition propensity signifies the effect of fuel composition and structure on autoignition characteristics.  相似文献

The primo vascular system (PVS) is observed in different parts of the body under different physiological and disease conditions. Previously, the PVS was not observed in the vagina. The vaginal samples of this study were collected from the female genitalia of healthy New Zealand white rabbits from the animal house, Faculty of Medicine, Assiut University. The vaginal samples were fixed in Bouin's solution. The sections were stained with hematoxylin and eosin and Crossmon's trichrome. Additionally, the sections were immunohistochemically stained with neuron-specific enolase (NSE) and vascular endothelial growth factor (VEGF). A primo node was observed on the lymph vessel of the vagina and has several characteristics that resemble those of the previously discovered primo nodes. The primo node in this study was surrounded by mesothelial cells that provide positive immunoreactivity to NSE and VEGF. Sinuses of different sizes, floating cells, telocyte-like cell, and primo microcells were observed as the main constituents of the primo node. Additionally, migratory cells were detected, which passed from the primo node to the enclosing lymph vessel.  相似文献

This study examined the effects of samarium and erbium co-doping on the structural, optical, and electrical properties of ceria (CeO2). Ceramic (Ce0.8Sm0.2-xErxO2-δ; x?=?0, 0.05, 0.10, 0.15, 0.20) electrolytes were synthesized via sol-gel assisted citric acid–nitrate combustion and calcined at 850?°C for 5?h. The calcined electrolytes possessed a cubic fluorite crystal structure without impure phases. The direct band gap of the calcined electrolytes increased as the erbium content increased and the lowest band gap was obtained for Ce0.8Sm0.2O2-δ (SDC) electrolyte. The calcined electrolyte powders were subsequently pressed into cylindrical pellets by uniaxial die pressing, and the pellets were sintered at 1400?°C for 5?h. The sintered densities of the pellets were measured with Archimedes’ method. The relative density of Ce0.8Sm0.1Er0.1O2-δ co-doped ceria electrolyte was higher than those of singly doped ones, and these findings were further confirmed through field emission scanning electron microscopy. Electrochemical impedance spectroscopy indicated that the conductivity of erbium-doped ceria increased as the samarium content increased. The maximum total ionic conductivity was observed in Ce0.8Sm0.1Er0.1O2-δ co-doped electrolyte. However, the singly doped SDC electrolyte exhibited the highest ionic conductivity of 13.12 mS/cm and the lowest activation energy of 0.580?eV at 600?°C among all other Ce0.8Sm0.2-xErxO2-δ co-doped ceria electrolytes.  相似文献

This study was performed to investigate certain major and toxic metal concentrations in different tissues of three demersal fish species (Triglia lucerna, Lophius budegassa, Solea lascaris). Generally, skin and liver exhibited higher metal concentrations than did muscle. Sodium and arsenic concentrations were found at higher levels in muscle tissues than in livers and skin, while zinc and nickel in different organs were classified as follows: skin > liver > muscle tissues. The concentrations of trace metals in fish samples indicated that S. lascaris was more contaminated than were other fish species, followed by L. budegassa and T. lucerna. It may be concluded that consumption of these species from this region is not likely to pose a threat for human health. However, although the concentrations are below the limit values for fish muscles, a potential danger may emerge in the future, depending on domestic waste waters and industrial activities in the region. Therefore, further monitoring programmes should be conducted.  相似文献

The demand for high-performance engineering products made from natural resources is increasing because of the low-cost, low-density, biodegradability, renewable nature and lighter than synthetic fibers. With these characteristics, the tribological performance of natural fiber composite has become an important element to be considered in most industrial and manufacturing functions. This paper presents an overview of the factors that influence the tribological performance of natural fiber composites, which include applied load, sliding distance, sliding velocity and fiber orientation. Influences of chemical treatment is also reviewed and illustrated through scanning electron microscope (SEM) observations. This review will focus on kenaf fibers (KFs) and oil palm fibers (OPFs) which have been widely exploited over the past few years among the available natural resources. The results show that the operating parameter, fiber orientation and chemical treatment has significant effects on the tribological performance of natural composite. A clear understanding of the factors that affect the tribological performance is very essential in performance improvement on natural fibers reinforced polymer composite for potential applications.  相似文献
