Bacteriophage 63D, previously isolated from sewage, is associated with alpha-2,8-linked polysialic acid degrading activity. We cloned a DNA fragment containing the sialidase gene from a 63D phage genomic library and the enzyme was functionally expressed in Escherichia coli. Determination of the nucleotide sequence of the fragment revealed that it contained one open reading frame (ORF) coding for a 108-kDa polypeptide consisting of 984 amino acid residues. The fragment had promoter sequences similar to the E. coli consensus promoters for sigma70. The deduced amino acid sequence of the central region of the ORF showed homology to those of phages K1F (51.6% identity) and PK1E (51.7% identity) endosialidases. Two Asp-box motifs that are widely found in sialidases were conserved. Purification of the soluble enzyme from lysed culture broth of infected E. coli yielded a 90-kDa protein upon SDS polyacrylamide gel electrophoresis, suggesting that the primary translational product is processed to the mature 90-kDa protein. The molecular mass of the enzyme was determined as 360 kDa by gel filtration, indicating that the native enzyme was probably a tetramer of identical 90-kDa subunits.  相似文献

Cationic polyelectrolytes (chitosan, poly-L-lysine (PLL), polyethyleneimine (PEI) and trimethylammonium glycol chitosan iodide (TGCI)) were used to entrap anoxygenic phototrophic bacteria in order to prevent the inhibitory effect of NH4+ on hydrogen production. When combined with agar gel, chitosan and PLL demonstrated no obvious repressive effect on hydrogen production by Rhodobacter sphaeroides under light-anaerobic conditions with lactate and glutamate as the carbon and nitrogen sources, respectively. On the other hand, both PEI and TGCI exerted a detrimental effect on hydrogen production under these conditions. Hydrogen production in the presence of NH4+ by the bacteria entrapped in the complex gel containing chitosan and agar improved considerably compared to that in the control containing only agar. Evidence shows that chitosan improves the hydrogen production via various effects. Diffusion tests demonstrated that the addition of chitosan increased to some extent the resistance to the diffusion of positively charged NH4+, but had no effect on negatively charged lactate. A buffer effect in the complex gels was also revealed.  相似文献

From the temperature dependence of the hole concentration in unirradiated lightly Al-doped 4H-SiC epilayers, an Al acceptor with EV + 0.2 eV, which is an Al atom (AlSi) at a Si sublattice site, and an unknown deep acceptor with EV + 0.35 eV are found, where EV is the top of the valence band. Both the densities are similar. With irradiation of 0.2 MeV electrons the Al acceptor density is reduced, while the unknown deep acceptor density is increased. Judging from the minimum electron energy required to displace a substitutional C atom (Cs) or the AlSi, the bond between the AlSi and its nearest neighbor Cs is broken due to the displacement of the Cs by this irradiation. Moreover, the displacement of the Cs results in the creation of a complex (AlSi-VC) of AlSi and a carbon vacancy (VC), indicating that the possible origin of the deep acceptor with EV + 0.35 eV is AlSi-VC.  相似文献

There have been a number of contradictory views concerning the efficacy of mass screening for neuroblastoma. Three Japanese and one cohort study from Quebec, and three Japanese cross-sectional studies were reviewed. The four cohort studies revealed that mass screening using high performance liquid chromatography (HPLC) reduced the incidence of this disease by about half in children aged from 1 to 4 years while little reduction was noted in screening utilizing non-HPLC methods. There is a large difference in the efficacy of mass screening depending on the method used. In the three cross-sectional studies, there was no discrimination between screened and unscreened cases, or between subjects screened by HPLC and non-HPLC methods. The percentage of children in the cross-sectional studies screened by HPLC was low. Cross-sectional studies seem inappropriate in assessing the effectiveness of current HPLC mass screening.  相似文献

The evaporation, carburization and diffusion behavior of Nb-coated Mo and of Nb-coated Ti were investigated in order to estimate the surface characteristics of the coated systems. Nb layers were formed on Mo by both chemical and physical vapor deposition and on Ti by just physical vapor deposition. From the diffusion behavior, interdiffusion coefficients were obtained at 1860°C, 1980°C and 2080°C for the chemically vapor-deposited Nb/Mo system. From evaporation experiments it was found that simultaneous evaporation of Nb and Mo occured at about 2300°C for thick films of Nb on Mo. For thin films of Nb on Mo, the evaporation of both Nb and Mo was confirmed at 1900°C by ion microprobe analysis of the condensate. For Nb/Ti, rapid diffusion of Nb into Ti and evaporation of Ti similar to the case of Nb-Ti alloys were observed. From the carburization study, it was found that an Nb coating appreciably retards the carburization of Ti and Mo.  相似文献

Ground water contaminated with trichloroethylene (TCE) used in electronic, electric, dry cleaning and the like industries is often treated by air-stripping. In this treatment process, TCE in its vapor form is stripped from ground water by air stream and sometimes emitted into the atmosphere without any additional treatments. Activated carbon adsorption is one of the practical and useful processes for recovering the TCE vapor from the exhaust air stream. However, adsorption of the TCE vapor from the stripping air stream onto activated carbons is not so simple as that from dry air, since in the exhaust air stream the TCE vapor coexists with water vapor with relatively high concentrations. The understanding of the adsorption characteristics of the TCE vapor to be adsorbed on activated carbon in the water vapor-coexisting system is essential for successfully designing and operating the TCE recovery process. In this work, the adsorption equilibrium relations of the TCE vapor adsorption on activated carbons were elucidated as a function of various relative humidity. Activated carbon fibers (ACFs) were used as model activated carbon. The adsorption equilibrium relations were studied by the column adsorption method. The adsorption isotherms of TCE vapor adsorbed on sample ACFs were successfully correlated by the Dubinin-Radushkevich equation for both cases with and without coexistent water vapor. No effects of coexistent water vapor were found on the limiting adsorption volume. However, the adsorption characteristic energy was significantly reduced by the coexistence of water vapor and its reduction was successfully correlated with the equilibrium amount of water vapor adsorbed under the dynamic condition.  相似文献

We present conventional detection of 2,4-dinitrophenol (DNP) for using the competitive reaction between DNP and DNP-conjugated albumin onto DNP antibody immobilized quartz crystal microbalance (QCM). This QCM method allows detection of DNP concentration in the range of 0.01 to 100 ng/ml; linear correlation obtains DNP concentration from 1 to 100 ng/ml.  相似文献
