Dielectric, X-ray, and piezoelectric studies of highly-resistive Li-doped (1-x)PbFe(1/2)Nb(1/2)O(3)-(x)PbTiO(3) (PFN-xPT) ceramics from the 0 ≤ x ≤ 0.2 range fabricated by solid-state synthesis and usual sintering have been carried out. Distinct anomalies of dielectric and piezoelectric parameters, corresponding to the transition between rhombohedral (monoclinic) and tetragonal ferroelectric phases, have been observed in pure PFN and PFN-xPT compositions with PbTiO(3) content up to 8 mol.%. As applications of nanoparticles in medical imaging and biomedicine rapidly expand, the interactions of nanoparticles with living cells have become an area of active interest. For example, intracellular accumulation of nanoparticles-an important part of cell-nanoparticle interaction-has been well studied using plasmonic nanoparticles and optical or optics-based techniques due to the change in optical properties of the nanoparticle aggregates. However, magnetic nanoparticles, despite their wide range of clinical applications, do not exhibit plasmonic-resonant properties and therefore their intracellular aggregation cannot be detected by optics-based imaging techniques. In this study, we investigated the feasibility of a novel imaging technique-pulsed magneto-motive ultrasound (pMMUS)-to identify intracellular accumulation of endocytosed magnetic nanoparticles. In pMMUS imaging a focused, high intensity, pulsed magnetic field is used to excite the cells labeled with magnetic nanoparticles, and ultrasound imaging is then used to monitor the mechanical response of the tissue. We demonstrated previously that clusters of magnetic nanoparticles amplify the pMMUS signal in comparison to the signal from individual nanoparticles. Here we further demonstrate that pMMUS imaging can identify interaction between magnetic nanoparticles and living cells, i.e.?intracellular accumulation of nanoparticles within the cells. The results of our study suggest that pMMUS imaging can not only detect the presence of magnetic nanoparticles but also provides information about their intracellular accumulation non-invasively and in real-time. 相似文献

Wet adhesive systems of insects strongly rely for their function on the formation of capillary bridges with the substrate. Studies on the chemical composition and evaporation dynamics of tarsal secretions strongly suggest a difference in chemistry of secretion in beetles and flies, both possessing hairy attachment devices. This difference is assumed to influence the viscosity of the secretion. Here, we applied a microrheological technique, based on the immersion of nanometric beads in the collected tarsal footprints, to estimate secretion viscosity in a beetle (Coccinella septempunctata) and a fly (Calliphora vicina). Both species studied possess distinct differences in viscosity, the median of which was calculated as 21.8 and 10.9 mPa s, respectively. We further present an approximate theoretical model to calculate the contact formation time of spatula-like terminal contact elements using the viscosity data of the covering fluid. The estimated contact formation time is proportional to the tarsal secretion viscosity and to the square of the contact radius of the contact element. 相似文献

Characterization of the stimulated release of neuropeptides from brain slices and individual cultured neurons requires efficient collection of the releasate from relatively large volumes of physiological saline. Here, several collection approaches are optimized using particle-embedded monolithic capillaries (PEMCs) with poly(stearyl methacrylate-co-ethylene glycol dimethacrylate) monolith acting as a "glue". Two distinct extraction particles, with either pyrrolidone (PY) or ethylenediamine (EDA) as the functional group on polystyrene backbone, have been embedded into capillaries having an inner diameter of 250 μm. The capillaries act as collection devices for sampling neuropeptide release; the collection protocols are described, and the extraction efficiency of the probes are characterized. Specifically, the binding of angiotensin II from a peptide mixture onto the PY and EDA columns was 16 and 28 pmol, respectively, in a volume of 20 μL of saline. The peptides released from these columns have been characterized via matrix-assisted laser desorption/ionization time-of-flight mass spectrometry with low femtomole detection limits. When the PEMC columns were positioned in close proximity to individual neurons and 50 mM KCl was used as the secretagogue, peptides released from individual identified cultured neurons isolated from Aplysia californica were collected and characterized. 相似文献

Capillary electrophoresis (CE) has been combined with matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) and radionuclide detection to assay mass-limited biological samples. Nanovial sampling techniques enable injections into the CE capillary from 50 to 150-nL volume samples; after the separation, nanoliter fraction collection combines the CE effluent with a MALDI matrix and minimizes sample spreading, thus allowing both MALDI MS and radionuclide detection on the CE fractions. MALDI MS complements the elution time information of CE by providing accurate molecular mass data, and radionuclide detection provides zeptomole limits of detection with quantitative information. While MALDI MS detects all fully processed peptides at sufficient concentration, culturing the neuron in media containing 35S-Met provides selective radionuclide detection of newly synthesized methionine-containing peptides. The analysis and detection of the expected neuropeptides and hormones in a single 40-microm bag cell neuron from Aplysia californica with CE/MALDI MS/radionuclide detection demonstrates the ability of this hyphenated approach to work with chemically complex mass-limited samples. 相似文献

Partition coefficients of phenol, salicylic acid, and several environmentally important chloro- and nitrophenols in a supercritical CO2-water system were measured using direct cocurrent extraction of aqueous solutions of the individual solutes with CO2. Partitioning data on the nitrophenols and salicylic acid were obtained for the first time. To bypass the troublesome and error-prone analysis of the CO2-rich phase, the present method employed only the solute concentrations in the aqueous phase before and after extraction to determine the partition coefficient. Unlike most previous engineering studies of phenol partitioning in a CO2-water system, the concentrations of phenolic solutes approached infinite dilution in both phases. This makes the results relevant to analytical-scale SFE of environmental water samples with CO2. Because of effective infinite dilution of the solutes, the partition coefficients provide a direct measure of relative CO2-philicity/hydrophilicity of the individual phenols. Compared to the octanol-water partition coefficients of substituted phenols, the CO2-water partition coefficients are more sensitive to substitution in the position neighboring the hydroxyl group. 相似文献

An improved variable-fidelity optimization algorithm for the simulation-driven design of microwave structures is presented. It exploits a set of electromagnetic-based models of increasing discretization density. These models are sequentially optimized with the optimum of the ‘coarser’ model being the initial design for the ‘finer’ one. The found optimum is further refined using a response surface approximation model constructed from the coarse-discretization simulation data. In this work, the computational efficiency of the variable-fidelity algorithm is enhanced by employing a novel algorithm for optimizing the coarse-discretization models. This allows reduction of the overall design time by up to 50% compared to the previous version. The presented technique is particularly suitable for problems where simulation-driven design is the only option, for example, ultra wideband and dielectric resonator antennas. Operation of the presented approach is demonstrated using two examples of antennas and a microstrip filter. In all cases, the optimal design is obtained at a low computational cost corresponding to a few high-fidelity simulations of the structure. 相似文献

Leafhoppers (Insecta, Hemiptera, Cicadellidae) actively coat their integuments with buckyball-shaped submicron proteinaceous secretory particles, called brochosomes. Here, we demonstrate that brochosomal coats, recently shown to be superhydrophobic, act as non-stick coatings and protect leafhoppers from contamination with their own sticky exudates—filtered plant sap. We exposed 137 wings of Alnetoidia alneti (Dahlbom), from half of which brochosomes were removed, to the rain of exudates under a colony of live A. alneti. One hundred and fifty-two droplets became stuck to the bared wings and only three to the intact wings. Inspection of the wings with a scanning electron microscope confirmed that the droplets that had hit the intact wings had rolled or bounced off the brochosomal coats. This is the first experimental study that tested a biological function of the brochosomal coats of leafhopper integuments. We argue that the production of brochosomes in leafhoppers and production of epidermal wax blooms in other sap-sucking hemipterans are alternative solutions, both serving to protect these insects from entrapment by their exudates. 相似文献
