A novel extracellular esterase was separated from culture supernatants of the basidiomycete Coprinopsis cinerea by means of foam fractionation. The parameters pH value, gas flow rate, addition of detergents, and the column design were varied to optimise the transport of the active enzyme into the foam phase. On the 70-mL scale, a recovery of activity of 79% with an enrichment factor of 10.5 was obtained at pH 7 and 20 mL air min−1 within 15 min. The enriched enzyme was characterized biochemically by semi-native SDS-PAGE and IEF electrophoresis with activity staining. Peptide sequencing was performed after tryptic digestion by electrospray tandem mass spectrometry. Homology searches revealed significant similarities to cutinases of various ascomycetes and to C. cinerea genome data recently annotated as cutinases.  相似文献

Queso Fresco has a limited shelf life and has been shown to support the rapid growth of Listeria monocytogenes during refrigerated storage. In addition to improving quality and extending shelf life, modified atmosphere packaging (MAP) has been used to control the growth of pathogenic microorganisms in foods. The objectives of this study were to determine the effects of MAP conditions on the survival and growth of spoilage microorganisms and L. monocytogenes during storage of Queso Fresco manufactured without starter cultures. For L. monocytogenes experiments, cheeses were surface inoculated at ~4 log10 cfu/g before packaging. Inoculated and uninoculated (shelf life experiments) cheeses were placed in 75-µm high-barrier pouches, packaged under 1 of 7 conditions including air, vacuum, or combinations of N2 and CO2 [100% N2 (MAP1), 30% CO2:70% N2 (MAP2), 50% CO2:50% N2 (MAP3), or 70% CO2:30% N2 (MAP4), 100% CO2 (MAP5)], and stored at 7°C. Samples were removed weekly through 35 d of storage. Listeria monocytogenes counts were determined for inoculated samples. Uninoculated samples were assayed for mesophilic and psychrotolerant counts, lactic acid bacteria, coliforms, and yeast and mold. In general, cheeses packaged under conditions consisting of higher contents of CO2 had lower pH levels during storage compared with those stored in conditions with lower levels or no CO2 at all. Similarly, the antimicrobial efficacy of MAP in controlling spoilage microorganisms increased with increasing CO2 content, whereas conditions consisting of 100% N2, vacuum, or air were less effective. Mean L. monocytogenes counts remained near inoculation levels for all treatments at d 1 but increased ~2 log10 cfu/g on cheeses packaged in air, vacuum, and 100% N2 (MAP1) conditions at d 7 and an additional ~1.5 log10 cfu/g at d 14 where they remained through 35 d. In contrast, treatments consisting of 70% CO2 (MAP4) and 100% CO2 (MAP5) limited increases in mean L. monocytogenes counts to <1 log10 cfu/g through 14 d and ~1.5 log10 cfu/g by d 21. Mean L. monocytogenes counts increased to levels significantly higher than inoculation (d 0) on cheeses stored in MAP2 and MAP3 on d 21, on d 28 for MAP4, and on d 35 for cheeses stored under MAP5 conditions. Overall, significant treatment × time interactions were observed between air, vacuum, and MAP1 when each was compared with MAP2, MAP3, MAP4, and MAP5. These data demonstrate that packaging fresh cheese under modified atmospheres containing CO2 may be a promising approach to extend shelf life while limiting L. monocytogenes growth during cold storage.  相似文献

Photosynthesis and nutrient status measurements were compared in size fractioned water samples collected in southern Lake Malawi in three different seasons. The size fractions analyzed were 0–2.0 μm, 0–20 μm, and 0–200 μm. Total chlorophyll concentrations were relatively invariant at about 1 μg L?1 in all seasons (wet stratified, deep mixing, dry stratified). Over 90% of total chlorophyll was < 20 μm and 40–50% < 2 μm. Stoichiometric ratios of carbon (C):nitrogen (N) and C:phosphorus (P) for the < 2-μm seston were similar to < 20-μm and < 200-μm seston samples and indicated that all size fractions were moderately N and P deficient in all seasons. N and P uptake experiments demonstrated that when the < 2-μm picoplankton were isolated from the larger sizes, they showed high N and P nutrient debts, apparently in response to the removal of nano- and micrograzers. This effect was strongest for N debt in the dry stratified season, but was apparent for P debt in all seasons. The chlorophyll-normalized light saturated rate of photosynthesis, Pbm, was always higher in whole water samples compared to the rates of the isolated picophytoplankton which were lowest in the dry season. We infer that nano- and micrograzers were an especially important source of regenerated nutrients when the lake was stratified although their removal as a source of nutrient to the picophytoplankton may affect Pbm in all seasons. Nitrogen regeneration was especially critical to the picophytoplankton in the dry stratified season but less important in the windy deep mixing season.  相似文献

Background: Treatment resistance of glioblastoma multiforme to chemo- and radiotherapy remains a challenge yet to overcome. In particular, the O6-methylguanine-DNA-methyltransferase (MGMT) promoter unmethylated patients have only little benefit from chemotherapy treatment using temozolomide since MGMT counteracts its therapeutic efficacy. Therefore, new treatment options in radiotherapy need to be developed to inhibit MGMT and increase radiotherapy response. Methods: Lomeguatrib, a highly specific MGMT inhibitor, was used to inactivate MGMT protein in vitro. Radiosensitivity of established human glioblastoma multiforme cell lines in combination with lomeguatrib was investigated using the clonogenic survival assay. Inhibition of MGMT was analyzed using Western Blot. Cell cycle distribution and apoptosis were investigated to determine the effects of lomeguatrib alone as well as in combination with ionizing radiation. Results: Lomeguatrib significantly decreased MGMT protein and reduced radiation-induced G2/M arrest. A radiosensitizing effect of lomeguatrib was observed when administered at 1 µM and increased radioresistance at 20 µM. Conclusion: Low concentrations of lomeguatrib elicit radiosensitization, while high concentrations mediate a radioprotective effect.  相似文献

One important category of non-ideal conditions for iris recognition is off-angle iris images. Practically it is very difficult for images to be captured with no offset. It then becomes necessary to account for off angle information in order to maintain robust performance. A biorthogonal wavelet based iris recognition system, previously designed at our lab, is modified and demonstrated to perform off-angle iris recognition. Biorthogonal wavelet network (BWN) are developed and trained for each class. The non-ideal factors are adjusted by repositioning the BWN. To test, along with the real data, synthetic iris images are generated by using affine and geometric transforms of 0°, 10° and 20° experimentally collected images. The tests were carried out on the experimentally collected off-angle data and synthetically generated data for angles from 0° to 60° with a resolution of 5°. This approach is shown to have less constraints than a transformation based iris recognition approach. Iris images off-angle by up to 42° for synthetic data and up to 45° for experimental data are successfully recognized.  相似文献

However, the justification for capital investments on resilience requires utilities to justify that the economic benefits outweigh the costs. This paper reviews the types of resilience measures being analyzed in cost-benefit analyses and addresses opportunities for improvement in characterizing the benefits for investments that enhance the resilience of electricity systems.  相似文献

Objective

Image post-processing corrects for cardiac and respiratory motion (MoCo) during cardiovascular magnetic resonance (CMR) stress perfusion. The study analyzed its influence on visual image evaluation.

Materials and methods

Sixty-two patients with (suspected) coronary artery disease underwent a standard CMR stress perfusion exam during free-breathing. Image post-processing was performed without (non-MoCo) and with MoCo (image intensity normalization; motion extraction with iterative non-rigid registration; motion warping with the combined displacement field). Images were evaluated regarding the perfusion pattern (perfusion deficit, dark rim artifact, uncertain signal loss, and normal perfusion), the general image quality (non-diagnostic, imperfect, good, and excellent), and the reader’s subjective confidence to assess the images (not confident, confident, very confident).

Results

Fifty-three (non-MoCo) and 52 (MoCo) myocardial segments were rated as ‘perfusion deficit’, 113 vs. 109 as ‘dark rim artifacts’, 9 vs. 7 as ‘uncertain signal loss’, and 817 vs. 824 as ‘normal’. Agreement between non-MoCo and MoCo was high with no diagnostic difference per-patient. The image quality of MoCo was rated more often as ‘good’ or ‘excellent’ (92 vs. 63%), and the diagnostic confidence more often as “very confident” (71 vs. 45%) compared to non-MoCo.

Conclusions

The comparison of perfusion images acquired during free-breathing and post-processed with and without motion correction demonstrated that both methods led to a consistent evaluation of the perfusion pattern, while the image quality and the reader’s subjective confidence to assess the images were rated more favorably for MoCo.

Neurons are especially susceptible to oxidative damage, which is increasingly implicated in neurodegenerative disease. Certain N‐acylethanolamines (NAEs) have been shown to protect neurons from oxidative stress. Since glaucoma may be considered a neurodegenerative disorder and the survival of retinal neurons could also be influenced by N‐acylethanolamines, our goal was to quantify changes in certain N‐acylethanolamine species and their oxylipin derivatives in the retina of a mouse model for glaucoma. We also sought to identify relationships between these and parameters of glaucoma disease development, specifically intraocular pressure, visual acuity, and contrast sensitivity. Five N‐acylethanolamine species and three NAE oxylipin derivatives were quantified in retina from young and aged DBA/2Crl mice. N‐Acylethanolamines and NAE‐oxylipins in retinal extracts were quantified against deuterated standards by isotope dilution gas chromatography–mass spectrometry. Levels (nmol/g dry weight) of N‐arachidonoylethanolamine (anandamide; NAE 20:4) were significantly (p = 0.008) decreased in aged (2.875 ± 0.6702) compared to young animals (5.175 ± 0.971). Conversely, the anandamide oxylipin, 15(S)‐HETE ethanolamide (15(S)‐HETE EA), was significantly (p = 0.042) increased in aged (0.063 ± 0.009) compared to young animals (0.039 ± 0.011). Enzymatic depletion of the anandamide pool by 15‐lipoxygenase and consequent accumulation of 15(S)‐HETE ethanolamine may contribute to decreased visual function in glaucomatous mice. Since N‐acylethanolamines effectively attenuate glaucoma pathogenesis and associated visual impairment, our data provides additional rationale and novel targets for glaucoma therapies.  相似文献
