Transendothelial electronic resistance (TEER) measurements are performed across a cell layer immobilized on a microfluidic device that also enables the cell layer to interact with a flowing stream of red blood cells (RBCs). A bipolar pulsed square wave potential is applied across a monolayer of bovine pulmonary artery endothelial cells, and the resulting current response is measured and integrated. The overall impedance of the cell layer provides an indicator of cell layer integrity. After cell seeding on the device, a decrease in TEER signal from 22.3 ± 1.6 μC to 3.5 ± 0.4 μC (corresponding to a resistance of 40.9 ± 2.9 Ω·cm(2) to 259.1 ± 27.4 Ω·cm(2)) was observed after 8 h of cell growth. Intracellular nitric oxide (NO) production by the immobilized endothelial cells that had reached confluence was 34% higher than those cells that had not reached confluence, as indicated by the integrated TEER system. Importantly, this NO production by the confluent endothelium was stimulated by ATP released from RBCs flowing under the endothelial cells. In this construct, the described microfluidic device enables both a TEER-based evaluation of cell layer integrity and molecularly communicated interactions of these cells with a flowing stream of blood components.  相似文献

Vitamins are essential to human health, and dietary supplements containing vitamins are widely used by individuals hoping to ensure they have adequate intake of these important nutrients. Measurement of vitamins in nutritional formulations is necessary to monitor regulatory compliance and in studies examining the nutrient intake of specific populations. Liquid chromatographic methods, primarily with UV absorbance detection, are well established for both fat- and water-soluble measurements, but they do have limitations for certain analytes and may suffer from a lack of specificity in complex matrices. Liquid chromatography-mass spectrometry (LC-MS) provides both sensitivity and specificity for the determination of vitamins in these matrices, and simultaneous analysis of multiple vitamins in a single analysis is often possible. In this work, LC-MS methods were developed for both fat- and water-soluble vitamins and applied to the measurement of these analytes in two NIST Standard Reference Materials. When possible, stable isotope labeled internal standards were employed for quantification.  相似文献

Dust is found throughout the universe and plays an important role for a wide range of astrophysical phenomena. In recent years, new IR facilities have provided powerful new data for understanding these phenomena. However, interpretation of these data is often complicated by a lack of complementary information about the optical properties of astronomically relevant materials. The Optical Properties of Astronomical Silicates with Infrared Techniques (OPASI-T) program at NASA's Goddard Space Flight Center is designed to provide new high-quality laboratory data from which we can derive the optical properties of astrophysical dust analogues. This program makes use of multiple instruments, including new equipment designed and built specifically for this purpose. The suite of instruments allows us to derive optical properties over a wide wavelength range, from the near-IR through the millimeter, also providing the capability for exploring how these properties depend upon the temperature of the sample. In this paper, we discuss the overall structure of the research program, describe the new instruments that have been developed to meet the science goals, and demonstrate the efficacy of these tools.  相似文献

We report the application of interferometry to the form measurement of a family of highly astigmatic optical surfaces. These measurements are based on a null test that employs a double-pass off-axis test arrangement with a tilted test surface and a reference sphere. This arrangement provides a perfect null test for an ellipsoid of revolution, or prolate spheroid. Its application is illustrated in detail in the presentation of results for the measurement of a specific family of eight differing surfaces that are incorporated into the K-Band Multi-Object Spectrometer Integral Field Unit. All surfaces measured here are sufficiently close to a prolate spheroid to justify its practical application. We discuss the application of the technique as a flexible low-cost approach for the generation of null interferograms in the measurement of a variety of complex surfaces.  相似文献

The concept of retardance is critically analyzed for ray paths through optical systems described by a three-by-three polarization ray-tracing matrix. Algorithms are presented to separate the effects of retardance from geometric transformations. The geometric transformation described by a "parallel transport matrix" characterizes nonpolarizing propagation through an optical system, and also provides a proper relationship between sets of local coordinates along the ray path. The proper retardance is calculated by removing this geometric transformation from the three-by-three polarization ray-tracing matrix. Two rays with different ray paths through an optical system can have the same polarization ray-tracing matrix but different retardances. The retardance and diattenuation of an aluminum-coated three fold-mirror system are analyzed as an example.  相似文献

A full-bandwidth k-domain linearization method for spectral-domain optical coherence tomography (SD-OCT) is demonstrated. The method uses information of the wavenumber-pixel-position provided by a translating-slit-based wavelength filter. For calibration purposes, the filter is placed either after a broadband source or at the end of the sample path, and the filtered spectrum with a narrowed line width (~0.5?nm) is incident on a line-scan camera in the detection path. The wavelength-swept spectra are co-registered with the pixel positions according to their central wavelengths, which can be automatically measured with an optical spectrum analyzer. For imaging, the method does not require a filter or a software recalibration algorithm; it simply resamples the OCT signal from the detector array without employing rescaling or interpolation methods. The accuracy of k-linearization is maximized by increasing the k-linearization order, which is known to be a crucial parameter for maintaining a narrow point-spread function (PSF) width at increasing depths. The broadening effect is studied by changing the k-linearization order by undersampling to search for the optimal value. The system provides more position information, surpassing the optimum without compromising the imaging speed. The proposed full-range k-domain linearization method can be applied to SD-OCT systems to simplify their hardware/software, increase their speed, and improve the axial image resolution. The experimentally measured width of PSF in air has an FWHM of 8?μm at the edge of the axial measurement range. At an imaging depth of 2.5?mm, the sensitivity of the full-range calibration case drops less than 10?dB compared with the uncompensated case.  相似文献

Ultraviolet (UV) lasers have the capability to precisely remove tissue via ablation; however, due to strong absorption of the applicable portion the UV spectrum, their surgical use is currently limited to extraocular applications at the air/tissue boundary. Here we report the development and characterization of a fiber-optic laser delivery system capable of outputting high-fluence UV laser pulses to internal tissue surfaces. The system has been developed with a view to intraocular surgical applications and has been demonstrated to ablate ocular tissue at the fluid/tissue boundary. The fifth (213?nm) and fourth(266?nm) harmonics of a Nd:YAG laser were launched into optical fibers using a hollow glass taper to concentrate the beam. Standard and modified silica/silica optical fibers were used, all commercially available. The available energy and fluence as a function of optical fiber length was evaluated and maximized. The maximum fluence available to ablate tissue was affected by the wavelength dependence of the fiber transmission; this maximum fluence was greater for 266?nm pulses (8.4?J/cm2) than for 213?nm pulses (1.4?J/cm2). The type of silica/silica optical fiber used did not affect the transmission efficiency of 266?nm pulses, but transmission of 213?nm pulses was significantly greater through modified silica/silica optical fiber. The optical fiber transmission efficiency of 213?nm pulses decreased as a function of number of pulses transmitted, whereas the transmission efficiency of 266?nm radiation was unchanged. Single pulses have been used to ablate fresh porcine ocular tissue. In summary, we report a method for delivering the fifth (213?nm) and fourth (266?nm) harmonics of a Nd:YAG laser to the surface of immersed tissue, the reliability and stability of the system has been characterized, and proof of concept via tissue ablation of porcine ocular tissue demonstrates the potential for the intraocular surgical application of this technique.  相似文献

A method for fabricating an achromatic, athermalized quarter-wave retarder is presented that involves monitoring retardance during polishing. A design specified by thicknesses alone is unlikely to meet specification due to uncertainties in birefringence. This method facilitates successful fabrication to a retardance specification despite these uncertainties. A retarder made from sapphire, MgF(2), and quartz was designed, fabricated, and its performance validated for the 0.470 to 0.865 μm wavelength region. Its specifications are as follows: at wavebands centered at 0.470, 0.660, and 0.865 μm, the band-averaged retardance should be 90°±10° for all fields and retardance should change less than 0.1° for a 1° change in temperature. Retarder fabrication accommodated birefringence and thickness uncertainties via the following steps. The first plate was polished to a target thickness. The retardance spectrum of the first plate was then measured and used to determine a retardance target for the second plate. The retardance spectrum of the combined first and second plates was then used to specify a retardance target for the third plate. The retardance spectrum of the three plates in combination was then used to determine when the final thickness of the third plate was reached.  相似文献

Polymer films of varying thicknesses were deposited onto cotton and polyester fabric samples by dip-coating from solution. Scanning electron microscopy (SEM) images of the coated fabric samples were used to evaluate the quality of the polymer coating. The samples were analyzed by infrared diffuse reflection spectroscopy to determine the relationship between film thickness and the effect of the coating on the spectroscopy of the two fabrics. Effects observed in four limiting cases are examined: (Case I) weak coating absorption on a fabric with weak absorption at the same frequency; (Case II) strong coating absorption in a spectral region of weak fabric absorption; (Case III) weak coating absorption in a spectral region of strong fabric absorption; and (Case IV) strong coating absorption in a spectral region of strong fabric absorption. In the first case, effects were dominated by reduced scattering as the coating is added. In the second case, the strong coating absorption that was observed at low coverages plateaus at higher coverage due to depth of penetration effects. In the third and fourth cases, reduced Fresnel diffuse reflection is measured as the coating is added, consistent with the reduction of scattering observed in the first case.  相似文献

Emissions from Portland cement manufacturing facilities may increase health risks in nearby populations and are thus subject to stringent regulations. Direct testing of pollutant concentrations in exhaust gases provides the best basis for assessing the extent of these risks. However, these tests (i) are often conducted under stressed, rather than typical, operating conditions, (ii) may be limited in number and duration, and (iii) may be influenced by specific fuel-types and attributes of individual kilns. We report here on the results of more than 150 emissions-tests conducted of two kilns at a Portland cement manufacturing plant in Portugal. The tests measured various regulated metals and polychlorinated dibenzo(p)dioxins and furans (PCDD/Fs). Stack-gas concentrations of pollutants were found to be highly variable, with standard deviations on the order of mean values. Emission rates of many pollutants were higher when coal was used as the main kiln fuel (instead of petroleum coke). Use of various supplemental fuels, however, had little effect on stack emissions, and few statistically significant differences were observed when hazardous waste was included in the fuel mix. Significant differences in emissions for some pollutants were observed between the two kilns despite their similar designs and uses of similar fuels. All measured values were found to be within applicable regulatory limits.  相似文献
