Alveolar Type II cells in the rat respond to severe, acute ozone injury (3 ppm ozone for eight hours) by increasing their intracellular pool of surfactant; however, the newly stored surfactant is abnormal in composition. Lamellar bodies isolated between 24 and 96 hours after ozone exposure contained significantly more cholesterol in relation to phosphatidylcholine than did controls. By contrast, the cholesterol content of surfactant isolated from alveolar lavage remained unchanged throughout an 8-day post-ozone period. The total protein content of lamellar bodies in relation to phosphatidylcholine was significantly decreased at 24 and 48 hours post-ozone. Analysis of lamellar body proteins by sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed that the amount of a 14 kDa proteolipid was greatly reduced at the end of the eight-hour ozone exposure and remained low for at least 48 hours. This proteolipid appeared to be a specific lamellar body component since it was not detected in extracellular surfactant. The findings indicate that oxidative alveolar stress initiates characteristic alterations in both lipid and protein constituents of stored surfactant, without perturbation in the composition of extracellular surfactant.  相似文献

Glycol diesters and mixtures of mono- and diesters have been prepared from methyl esters of partially hydrogenated soybean oil fatty acids and diethylene, dipropylene, neopentyl and triethylene glycols. The catalyst used in these reactions was a mixture of calcium acetate/barium acetate (3∶1, w/w). The reactions were carried out under nitrogen with 0.5% catalyst at temperatures in the range of 190–275°C. Borated esters of mixed mono- and diesters were prepared with 0.33 equivalent of boric acid per 1.0 equivalent hydroxyl group on the ester. Refractive indices, viscosities, and flash and fire points were determined for diesters, mixed mono- and diesters, and mixed diesters and borated esters. The viscosities, flash points and fire points indicate that these esters can be used as a component of lubricating oils. Wear-prevention characteristics of mixed diesters and borated esters indicated that they can be used as antifriction additives in lubricating oils. Morphology and fracture mechanisms in two nanoclay‐filled epoxy systems were investigated using both microscopy and spectroscopy tools. Clay exfoliation was achieved using a series of sample preparation steps, and confirmed using wide angle X‐ray diffraction (XRD) and transmission electron microscopy (TEM) techniques. Significant improvement in modulus was obtained when clay exfoliation was achieved. Incorporation of core‐shell rubber (CSR) in both caly‐filled epoxy systems leads to greatly enhanced fracture toughness. Optical microscopy and TEM observations of the CSR‐modified nanocomposites suggest that CSR cavitation. shear yielding of the matrix, clay layer delamination. CSR bridging, crack bifurcation. and crack deflection are among the operative toughening mechanisms observed, depending on the nature of the epoxy matrix utilized.  相似文献

We give matching upper and lower bounds of \(\varTheta(\min(\frac{\log m}{\log \log m},\, n))\) for the individual step complexity of a wait-free m-valued adopt-commit object implemented using multi-writer registers for n anonymous processes. While the upper bound is deterministic, the lower bound holds for randomized adopt-commit objects as well. Our results are based on showing that adopt-commit objects are equivalent, up to small additive constants, to a simpler class of objects that we call conflict detectors. Our anonymous lower bound also applies to the individual step complexity of m-valued wait-free anonymous consensus, even for randomized algorithms with global coins against an oblivious adversary. The upper bound can be used to slightly improve the cost of randomized consensus against an oblivious adversary. For deterministic non-anonymous implementations of adopt-commit objects, we show a lower bound of \(\varOmega(\min(\frac{\log m}{\log \log m}, \frac{\sqrt{\log n}}{\log \log n}))\) and an upper bound of \(O(\min(\frac{\log m}{\log \log m},\, \log n))\) on the worst-case individual step complexity. For randomized non-anonymous implementations, we show that any execution contains at least one process whose steps exceed the deterministic lower bound.  相似文献

Two different combinatorial mutagenesis experiments on the light-harvestingII (LH2) protein of Rhodobacter capsulatus indicate that heuristicrules relating sequence directly to phenotype are dependenton which sets or groups of residues are mutated simultaneously.Previously reported combinatorial mutagenesis of this chromogenicprotein (based on both phylogenetic and structural models) showedthat substituting amino acids with large molar volumes at Glyß31caused the mutated protein to have a spectrum characteristicof light-harvesting I (LH1). The six residues that underwentcombinatorial mutagenesis were modeled to lie on one side ofa transmembrane -helix that binds bacteriochlorophyll. In asecond experiment described here, we have not used structuralmodels or phylogeny in choosing mutagenesis sites. Instead,a set of six contiguous residues was selected for combinatorialmutagenesis. In this latter experiment, the residue substitutedat Glyß31 was not a determining factor in whetherLH2 or LH1 spectra were obtained; therefore, we conclude thatthe heuristic rules for phenotype prediction are context dependent.While phenotype prediction is context dependent, the abilityto identify elements of primary structure causing phenotypediversity appears not to be. This strengthens the argument forperforming combinatorial mutagenesis with an arbitrary groupingof residues if structural models are unavailable.  相似文献

Enthusiasm for using beaver dam analogues (BDAs) to restore incised channels and riparian corridors has been increasing. BDAs are expected to create a similar channel response to natural beaver dams by causing channel bed aggradation and overbank flow, which subsequently raise water tables and support vegetation growth. However, lack of funding for monitoring projects post‐restoration has limited research on whether BDAs actually cause expected channel change in the Front Range and elsewhere. Geomorphic and hydrologic response to BDAs was monitored in two watersheds 1 year post‐restoration. BDAs were studied at Fish Creek, a steep mountainous catchment, and Campbell Creek, a lower gradient piedmont catchment from May to October 2018. At each restoration site, the upstream‐ and downstream‐most BDAs were chosen for intensive study in comparison with unrestored reference reaches. Monitoring focused on quantifying sediment volumes in BDA ponds and recording changes to stream stage and riparian groundwater. Despite differences in physical basin characteristics, BDA pools at both sites stored similar volumes of sediment and stored more sediment than reference pools. Sediment storage is positively correlated to BDA height and pool surface area. However, BDAs did not have a significant influence on shallow groundwater. The lack of groundwater response proximal to BDAs could indicate that local watershed factors have a stronger influence on groundwater response than restoration design 1 year post‐restoration. Systematic, long‐term studies of channel and floodplain response to BDAs are needed to better understand how BDAs will influence geomorphology and hydrology.  相似文献

The purpose statement of Deakin University Library is ‘We help people learn.’ This applies equally to on or off‐campus students, which has meant that the Library has strived to provide innovative resources and services to break down the barriers of geography and time experienced by our remote students. With the advances in information and communication technologies, Deakin University Library has been able to capitalise on these developments to deliver electronic services to both on and off‐campus students, thus complementing the valuable services provided by the Library team. While the services provided through the virtual or digital library are a great leap forward, providing access to a wide range of resources and services 24 hours per day x 7 days per week, these services must be seen in conjunction with the professional assistance provided by librarians.
