Mitochondrial Complex II is composed of four core subunits and mutations to any of the subunits result in lowered Complex II activity. Surprisingly, although mutations in any of the subunits can yield similar clinical outcomes, there are distinct differences in the patterns of clinical disease most commonly associated with mutations in different subunits. Thus, mutations to the SdhA subunit most often result in mitochondrial disease phenotypes, whilst mutations to the other subunits SdhB-D more commonly result in tumour formation. The reason the clinical outcomes are so different is unknown. Here, we individually antisense-inhibited three of the Complex II subunits, SdhA, SdhB or SdhC, in the simple model organism Dictyostelium discoideum. Whilst SdhB and SdhC knockdown resulted in growth defects on bacterial lawns, antisense inhibition of SdhA expression resulted in a different pattern of phenotypic defects, including impairments of growth in liquid medium, enhanced intracellular proliferation of the bacterial pathogen Legionella pneumophila and phagocytosis. Knockdown of the individual subunits also produced different abnormalities in mitochondrial function with only SdhA knockdown resulting in broad mitochondrial dysfunction. Furthermore, these defects were shown to be mediated by the chronic activation of the cellular energy sensor AMP-activated protein kinase. Our results are in agreement with a role for loss of function of SdhA but not the other Complex II subunits in impairing mitochondrial oxidative phosphorylation and they suggest a role for AMP-activated protein kinase in mediating the cytopathological outcomes. 相似文献

We study the problem of minimizing data aggregation time in wireless sensor networks (WSNs) under the practical duty-cycle scenario where nodes switch between active states and dormant states periodically for energy efficiency. Under the protocol interference model, we show that the problem is NP-hard and present a lower bound of delay for any data aggregation scheme. To solve the problem efficiently, we then construct a routing tree based on connected dominator set and propose two aggregation scheduling algorithms, which are the centralized Greedy Aggregation Scheduling (GAS) and the distributed Partitioned Aggregation Scheduling (PAS), so as to generate collision-free transmission schedules for data aggregation in duty-cycled WSNs. To minimize the total delay, GAS tries to achieve maximal concurrent transmissions in each time-slot during each frame by using global information, while PAS leverages a network partition based strategy and local information to ensure the largest degree of channel reuse across space and time domains. Theoretical analysis indicates that each algorithm consumes at most \(O(R+\varDelta)\) frames and achieves nearly constant factor approximation on the optimal delay. Here R and \(\varDelta\) are the network radius and the maximum node degree, respectively. Sorption-based atmospheric water generation (SAWG) is a promising strategy to alleviate the drinkable water scarcity of arid regions. However, the high-water production efficiency remains challenging due to the sluggish sorption/desorption kinetics. Herein, a composite sorbent@biomimetic fibrous membrane (PPy-COF@Trilayer-LiCl) is reported by mimicking nature's Murray networks, which exhibits outstanding water uptake performance of 0.77–2.56 g g−1 at a wide range of relative humidity of 30%–80% within 50 min and fast water release capacity of over 95% adsorbed water that can be released within 10 min under one sun irradiation. The superior sorption–desorption kinetics of PPy-COF@Trilayer-LiCl are enabled by the novel hierarchically porous structure, which is also the critical factor to lead a directional rapid water transport and vapor diffusion. Moreover, as a proof-of-concept demonstration, a wearable SAWG device is established, which can operate 10 sorption–desorption cycles per day in the outdoor condition and produce a high yield of clean water reaching up to 3.91 kg m−2 day−1.
