Matrix-dominated mechanical behavior of a graphite/epoxy composite has been found to be affected by sub-Tg annealing. Postcured (±45°)4S specimens of Thornel 300 graphite/Narmco 5208 epoxy were quenched from above Tg and given a sub-Tg annealing at 140°C for times up to 105 min. The ultimate tensile strength, strain-to-break, and toughness of the composite material were found to decrease as functions of sub-Tg annealing time. No weight loss was observed during the sub-Tg annealing. The time-dependent change in mechanical behavior is explained on the basis of free-volume changes that are related to the physical aging of the nonequilibrium glassy network-epoxy. The results imply possible changes in composite properties with service time.  相似文献

Hypertension is a major public health concern and poses a significant risk for sudden cardiac death (SCD). However, the characterisation of human tissues tends to be macroscopic, with little appreciation for the quantification of the pathological remodelling responsible for the advancement of the disease. While the components of hypertensive remodelling are well established, the timeline and comparative quantification of pathological changes in hypertension have not been shown before. Here, we sought to identify the phasing of cardiac remodelling with hypertension using post-mortem tissue from SCD patients with early and advanced hypertensive heart disease (HHD). In order to study and quantify the progression of phenotypic changes, human specimens were contrasted to a well-described angiotensin-II-mediated hypertensive mouse model. While cardiomyocyte hypertrophy is an early adaptive response in the mouse that stabilises in established hypertension and declines as the disease progresses, this finding did not translate to the human setting. In contrast, optimising fibrosis quantification methods and applying them to each setting identified perivascular fibrosis as the prevailing possible cause for overall disease progression. Indeed, assessing myocardial inflammation highlights CD45+ inflammatory cell infiltration that precedes fibrosis and is an early-phase event in response to elevated arterial pressures that may underscore perivascular remodelling. Along with aetiology insight, we highlight cross-species comparison for quantification of cardiac remodelling in human hypertension. As such, this platform could assist with the development of therapies specific to the disease phase rather than targeting global components of hypertension, such as blood pressure lowering.  相似文献

Immunosuppressants and biologicals are widely used therapeutics for various chronic inflammatory diseases (CID). To gain more detailed insight into their downstream effects, we examined their impact on serum immunoglobulin G (IgG) glycosylation. We analyzed IgG subclass-specific fragment crystallizable (Fc) N-glycosylation in patients suffering from various CID using the LC-MS approach. Firstly, we compared IgG Fc N-glycosylation between 128 CID patients and 204 healthy controls. Our results replicated previously observed CID-related decrease in IgG Fc galactosylation (adjusted p-value range 1.70 × 10−2–5.95 × 10−22) and sialylation (adjusted p-value range 1.85 × 10−2–1.71 × 10−18). Secondly, to assess changes in IgG Fc N-glycosylation associated with therapy and remission status, we compared 139 CID patients receiving either azathioprine, infliximab, or vedolizumab therapy. We observed an increase in IgG Fc galactosylation (adjusted p-value range 1.98 × 10−2–1.30 × 10−15) and sialylation (adjusted p-value range 3.28 × 10−6–4.34 × 10−18) during the treatment. Furthermore, patients who reached remission displayed increased Fc galactosylation levels (p-value range 2.25 × 10−2–5.44 × 10−3) in comparison to patients with active disease. In conclusion, the alterations in IgG Fc glycosylation and the fact these changes are even more pronounced in patients who achieved remission, suggest modulation of IgG inflammatory potential associated with CID therapy.  相似文献

Clusters of mobile (LEO) satellites, flying at a non-geostationary orbit, have been recently proposed, designed and made operational for achieving a global coverage of roaming users. An example of these is Teledesic. The satellites of these clusters are equipped with a radio frequency switch and connected via intersatellite links to form a specific topology. Uncoordinated packet transmission in these systems may result in collisions. Collided packets must be retransmitted, with an obvious degradation in performance, in terms of both bandwidth usage and delay. This performance degradation can be overcome by a proper scheduling of the packets. In this paper, we consider optimal (namely minimum length) packet scheduling in LEO satellite clusters. We present a preprocessing algorithm which, together with the optimal, polynomial time, packet scheduling algorithm for isolated satellites, produces an optimal schedule in tree-connected clusters. The overall algorithm inherit the time complexity of the optimal one for isolated systems, and is O(N 4) for a cluster serving N roaming users.  相似文献

Oligonucleotide-directed mutagenesis has been used to replaceglycine residues by alanine in neutral protease from Bacillussubtilis. One Gly to Ala substitution (G147A) was located ina helical region of the protein, while the other (G189A) wasin a loop. The effects of mutational substitutions on the functional,conformational and stability properties of the enzyme have beeninvestigated using enzymatic assays and spectroscopic measurements.Single substitutions of both G1y147 and Gly189 with Ala residuesaffect the enzyme kinetic properties using synthetic peptidesas substrates. When Gly replacements were concurrently introducedat both positions, the kinetic characteristics of the doublemutant were roughly intermediate between those of the two singlemutants, and similar to those of the wild-type protease. Bothmutants G147A and G189A were found to be more stable towardsirreversible thermal inactivation/unfolding than the wild-typespecies. Moreover, the stabilizing effect of the Gly to Alasubstitution was roughly additive in the double mutant G147A/G189A,which shows a 3.2°C increase in Tm with respect to the wild-typeprotein. These findings indicate that the Gly to Ala substitutioncan be used as a strategy to stabilize globular proteins. Thepossible mechanisms of protein stabilization are also discussed.  相似文献

The present author comments on the study of S. J. Lederman et al (see record 1986-04159-001), which focuses on the strategies people use to make haptic judgments. It is argued that contextual and task conditions should not be regarded as irrelevant noise that leads to unfortunate perceptual distortions, but rather as important sources of information that elicit the strategies people use and that partly determine performance. People are not confined to a single heuristic even within a task. Some practical and theoretical implications of this view are discussed briefly. The Network of Tasks (NOT) model allows adaptive node programs written in a variety of parallel languages to be connected together in an almost acyclic task graph. The main difference between NOT and other task graphs is that it is designed to make the performance of the graph predictable from knowledge of the performance of the component node programs and the visible structure of the graph. It can therefore be regarded as a coordination language that is transparent about performance. For large-scale computations that are distributed to geographically-distributed compute servers, the NOT model helps programmers to plan, assemble, schedule, and distribute their problems.  相似文献

Nanoparticle based magnetic relaxation switch (MRSw) biosensors offer the opportunity to develop magnetic resonance based in vitro diagnostics. Critical attributes for point of care in vitro diagnostic products include simple instrumentation and ease of use. To this end, high-resolution biexponential analysis was used to permit measurement and assignment of two samples with a single radio frequency detection coil. This approach was used to calibrate and expand the dynamic range of MRSw biosensors in a single step. The potential for easy-to-use MRSw-based diagnostics was demonstrated by combining the method for single-step measurement of two samples with a disposable, plastic cartridge and dried MRSw reagents to obtain a calibrated reading after only two steps: mix and read. Taken together, our results suggest the feasibility of developing magnetic resonance based in vitro diagnostics that offer extreme ease of use and simple instrumentation.  相似文献
