Induction of apoptosis in human microvascular endothelial cells by divalent cobalt ions. Evidence for integrin-mediated signaling via the cytoskeleton

Wound healing following implantation is characterized by an acute inflammatory reaction and a subsequent reorganizing phase in which angiogenesis is involved. Endothelial cells (EC) participate in both inflammation and angiogenesis. Thus, the effects on functions of EC exerted by implanted materials could affect the progression of wound healing. The corrosion of metallic implants can cause high concentrations of heavy metal ions in the peri-implant tissues. The purpose of the present study was to test the effects of possible corrosion products on the function and viability of human EC in vitro. Long-term exposure of EC to CoCl2 and NiCl2 (3 days, 0.7 mM) leads to a decrease of cell number and changes in cellular morphology. However, the morphological changes between CoCl2- and NiCl2-treated cells differ significantly. The changed morphology of CoCl2-treated EC and the fragmented DNA pattern indicates apoptosis. Nickel-treated cells demonstrated necrosis. The activity of integrins was tested by an assay of cellular adhesion on collagen-coated surfaces. It was shown that the number of adherent cells significantly decreased upon exposure to CoCl2. Our studies suggest that induction of cell death in EC upon exposure to CoCl2 could be attributed to impaired integrin signaling, which leads to a damaged cytoskeleton and culminates in apoptosis.     

Polyionic fusion peptides function as specific dimerization motifs

The de novo design of a molecular adapter for directed associationand covalent linkage of two polypeptides is presented. Usingpeptides containing charged amino acid residues and an additionalcysteine residue (AlaCysLys₈ and AlaCysGlu₈) we demonstratethat the electrostatic interaction promotes the associationof two synthetic peptides and, subsequently, disulfide bondformation. The reaction depends on both the redox potentialand on the ionic strength of the buffer. Varying the redox potential,the interaction of the peptides was quantified by a G^0' of 6.6± 0.2 kcal/mol. Heterodimerization of the peptides ishighly specific, a competition of association by other cysteinecontaining compounds could not be observed. Two proteins comprisingcysteine-containing polyionic fusion peptides, a modified Fabfragment and an -glucosidase fusion, could be specifically conjugatedby directed association and subsequent disulfide bond formation.Both proteins retain their functional characteristics withinthe bifunctional conjugate: enzymatic activity of the glucosidaseand antigen-binding capacity of the Fab fragment are equivalentto the non-conjugated components.     

Internet vs. paper and pencil administration of questionnaires commonly used in panic/agoraphobia research

The aim of this study was to investigate the psychometric properties of Internet administered questionnaires used in panic research. Included were 494 people who had registered for an Internet-based treatment program for panic disorder (PD). Participants were randomly assigned to fill in the questionnaires either on the Internet or the paper-and-pencil versions, and then to fill in the same questionnaires again the next day using the other format. Results showed largely equivalent psychometric properties for the two administration formats (Cronbach’s α between 0.79 and 0.95). The results also showed high and significant correlations between the Internet and the paper-and-pencil versions. Analyses of order effects showed an interaction effect for the BSQ and the MI (subscale Accompanied), a main effect was identified for ACQ, MI-Alone, BAI and BDI II. However, in contrast to previous research, the Internet version did not consistently generate higher scores and effect sizes for the differences were generally low. Given the presence of an interaction effect, we recommend that the administration format should be stable in research across measurement points. Finally, the findings suggest that Internet versions of questionnaires used in PD research can be used with confidence.     

Business owners' action planning and its relationship to business success in three African countries.

A model of business success was developed with motivational resources (locus of control, self-efficacy, achievement motivation, and self-reported personal initiative) and cognitive resources (cognitive ability and human capital) as independent variables, business owners' elaborate and proactive planning as a mediator, and business size and growth as dependent variables. Three studies with a total of 408 African micro and small-scale business owners were conducted in South Africa, Zimbabwe, and Namibia. Structural equation analyses partially supported the hypotheses on the importance of psychological planning by the business owners. Elaborate and proactive planning was substantially related to business size and to an external evaluation of business success and was a (partial) mediator for the relationship between cognitive resources and business success. Proteins Marking the Sequence of Genotoxic Signaling from Irradiated Mesenchymal Stromal Cells to CD34+ Cells

Non-targeted effects (NTE) of ionizing radiation may initiate myeloid neoplasms (MN). Here, protein mediators (I) in irradiated human mesenchymal stromal cells (MSC) as the NTE source, (II) in MSC conditioned supernatant and (III) in human bone marrow CD34+ cells undergoing genotoxic NTE were investigated. Healthy sublethal irradiated MSC showed significantly increased levels of reactive oxygen species. These cells responded by increasing intracellular abundance of proteins involved in proteasomal degradation, protein translation, cytoskeleton dynamics, nucleocytoplasmic shuttling, and those with antioxidant activity. Among the increased proteins were THY1 and GNA11/14, which are signaling proteins with hitherto unknown functions in the radiation response and NTE. In the corresponding MSC conditioned medium, the three chaperones GRP78, CALR, and PDIA3 were increased. Together with GPI, these were the only four altered proteins, which were associated with the observed genotoxic NTE. Healthy CD34+ cells cultured in MSC conditioned medium suffered from more than a six-fold increase in γH2AX focal staining, indicative for DNA double-strand breaks, as well as numerical and structural chromosomal aberrations within three days. At this stage, five proteins were altered, among them IQGAP1, HMGB1, and PA2G4, which are involved in malign development. In summary, our data provide novel insights into three sequential steps of genotoxic signaling from irradiated MSC to CD34+ cells, implicating that induced NTE might initiate the development of MN.     

Comparing Google Scholar and ISI Web of Science for Earth Sciences

In order to measure the degree to which Google Scholar can compete with bibliographical databases, search results from this database is compared with Thomson’s ISI WoS (Institute for Scientific Information, Web of Science). For earth science literature 85% of documents indexed by ISI WoS were recalled by Google Scholar. The rank of records displayed in Google Scholar and ISI WoS, is compared by means of Spearman’s footrule. For impact measures the h-index is investigated. Similarities in measures were significant for the two sources.     

Determination of the antioxidant capacity: influence of the sample concentration on the measured values

The aim of the present analysis was to study the influence of the sample concentration on the measured antioxidant capacity, since such investigations are scarce but necessary to ensure the reproducibility of the results. Pure substances (ascorbic acid, gallic acid, Trolox^®, uric acid) and food extracts (strawberry nectar, tomato extract, white tea) were analysed using seven common antioxidant capacity assays (three versions of Trolox equivalent antioxidant capacity, TEAC; ferric reducing antioxidant power, FRAP; photochemiluminescence, PCL; oxygen radical absorbance capacity, ORAC and total phenolics assay). For all applied pure substances and in most of the assays effects of the sample concentration on the measured antioxidant capacity were observed. Since it remains speculative how sample concentration does affect the measured antioxidant capacity exactly, it is strongly recommended to use at least three sample concentrations for analysis to detect and to discuss concentration-dependent effects.     

Two-layer channel routing with vertical unit-length overlap

We show that anyn-net 2-terminal channel routing problem of densityd can be wired on a two-layer grid of widthw =d +O(d ^2/3) when vertical wire segments are allowed to overlap for a distance of length 1. This is a considerable asymptotic improvement over the best known, and optimal, channel width of 2d-1 for models in which no vertical overlap is allowed [RBM, PL]. Our result also improves the 3d/2+O(1) channel width achieved by a recent algorithm [G] for the same vertical overlap model. The algorithm presented in this paper produces at most 4 overlaps of unit length between any two nets, usesO(n) contacts, and can be implemented to run inO(nd ^2/3) time.
