Novel Very Long-Chain α-Methoxylated Δ5,9 Fatty Acids from the Sponge Asteropus niger Are Effective Inhibitors of Topoisomerases IB 下载免费PDF全文

The novel fatty acids (2R,5Z,9Z)‐2‐methoxy‐25‐methyl‐5,9‐hexacosadienoic acid ( 1a ) and (2R,5Z,9Z)‐2‐methoxy‐24‐methyl‐5,9‐hexacosadienoic acid ( 1b ) were isolated in 80 % purity from the Caribbean sponge Asteropus niger by chloroform/methanol extraction followed by solvent partitioning and silica gel column chromatography. The compounds were characterized by utilizing a combination of gas chromatography‐mass spectrometry, nuclear magnetic resonance, and circular dichroism. Acids 1a and 1b were not detected in the phospholipids (PtdCho and PtdIns) of the sponge, but rather as free FA and possibly in glycosylceramides. The mixtures of 1a and 1b displayed cytotoxicity towards THP‐1 and HepG2 cells with EC50's between 41 and 35 μg/mL. Apoptosis was not the preferred mode of cell death induced by 1a – 1b in the THP‐1 cells. This implies other types of cytotoxicity mechanisms, such as membrane disruption and/or the inhibition (EC50 = 1.8 μg/mL) of the human topoisomerase IB enzyme (hTopIB), with a mechanism of inhibition different from the one displayed by camptothecin (CPT). In a separate experiment, the mixture of 1a and 1b also displayed cytotoxicity towards ex vivo mouse splenocytes infected with Leishmania infantum amastigotes (IC50 = 0.17 mg/mL) and free living promastigotes (IC50 = 0.34 mg/mL). It was also found that the FA were inhibitory of the Leishmania topoisomerase IB (LTopIB) with an EC50 = 5.1 μg/mL. Taken together, 1a and 1b represent a new class of FA with potential as TopIB inhibitors that preferentially inhibit hTopIB over LTopIB. 相似文献

The electrochemical characteristics of sintered CuAg4 at. pct alloy in different stages of synthesis and thermomechanical treatment were examined by cyclic voltammetry in a NaOH medium. On the voltammograms recorded at the sintered alloy, six current waves can be noticed at the anodic part, and six corresponding current waves are observed at the cathodic part. A possible electrochemical reaction is attributed to each wave. The effect of the potential reversing limit was used to distinguish the correlation between the anodic and cathodic peaks. After the deformation with 60 pct deformation degree, the alloy becomes more corrosion resistant, whereas the corrosion mechanism remains the same as before mechanical treatment. Subsequent anneal of the deformed alloy at 533 K (260 °C) for 150 minutes leads to the hardening effect. At the same time, the alloy remains corrosion stable, and some of the current waves present at the other voltammograms are hardly noticeable here. The relaxation processes of hybrid organic-inorganic polymer nanosystems (OIS) synthesized by joint polymerization of organic and inorganic components were studied using methods of differential scanning calorimetry (DSC), dynamic mechanical thermal analysis (DMTA), and broadband dielectric relaxation spectroscopy (DRS). The organic component was a mixture of two products: high-molecular-weight macrodiisocyanate (MDI) with low reactivity and low-molecular-weight isocyanate-containing modifier poly(isocyanate) (PIC) with high reactivity. Sodium silicate (SS) was used as inorganic component. The structures of the OIS obtained were in the form of hybrids with covalently connected building blocks and interpenetrating networks: weakly cross-linked network MDI/SS and highly cross-linked network PIC/SS. Depending on the MDI/PIC ratio, one of the networks was prevailing and created a continuous structure with domains of second network.

Cathodic material for sodium-ion rechargeable batteries based on NaxMnO2 were synthesized by glycine nitrate method and subsequent annealing at high temperatures. Different crystal structures with different morphologies were obtained depending on the annealing temperature: hexagonal layeredα-Na0.7MnO2.05 nanoplates were obtained at 850 °C, while 3-D tunnel structured Na0·4MnO2 and Na0·44MnO2, both with rod-like morphology, were obtained at 800 °C and 900 °C, respectively. The investigations of the electrochemical behavior of obtained cathodic materials in aqueous NaNO3 solution have shown that Na0·44MnO2 obtained at 900 °C has shown the best battery performance. Its initial discharge capacities are 123.5 mA h/g, 113.2 mA h/g, and 102.0 mA h/g at the high current densities of 1000, 2000 and 5000 mA/g, respectively. 相似文献

In the search for new and effective treatments of breast and prostate cancer, a series of hybrid compounds based on tamoxifen, estrogens, and artemisinin were successfully synthesized and analyzed for their in vitro activities against human prostate (PC-3) and breast cancer (MCF-7) cell lines. Most of the hybrid compounds exhibit a strong anticancer activity against both cancer cell lines – for example, EC50 (PC-3) down to 1.07 μM, and EC50 (MCF-7) down to 2.08 μM – thus showing higher activities than their parent compounds 4-hydroxytamoxifen (afimoxifene, 7 ; EC50=75.1 (PC-3) and 19.3 μM (MCF-7)), dihydroartemisinin ( 2 ; EC50=263.6 (PC-3) and 49.3 μM (MCF-7)), and artesunic acid ( 3 ; EC50=195.1 (PC-3) and 32.0 μM (MCF-7)). The most potent compounds were the estrogen-artemisinin hybrids 27 and 28 (EC50=1.18 and 1.07 μM, respectively) against prostate cancer, and hybrid 23 (EC50=2.08 μM) against breast cancer. These findings demonstrate the high potential of hybridization of artemisinin and estrogens to further improve their anticancer activities and to produce synergistic effects between linked pharmacophores. 相似文献

The formation, morphology and transport properties of a new multilayer pervaporation membrane have been studied. This membrane consists of a microporous support based on a poly(amideimide), an intermediate poly[2-(N,N-dimethylamino)ethyl methacrylate] layer, and a top diffusion layer of ladder polysiloxane. The membrane has low permeability for weakly polar liquids (e.g. cyclohexane); it exhibits dehydrating properties for waterisopropanol mixtures and has high productivity (up to 14.5 kg h-1 m-2) and high separation factor (up to 27430) when methanol is isolated from mixtures with cyclohexane. The transport properties of the membrane are discussed with respect to the Henis-Tripodi resistance model, taking into account the hydrophobic-hydrophilic nature of the polymers used. 相似文献

The objective of the present study was to review the existing data on the association between Zn status and characteristics of gut microbiota in various organisms and the potential role of Zn-induced microbiota in modulating systemic effects. The existing data demonstrate a tight relationship between Zn metabolism and gut microbiota as demonstrated in Zn deficiency, supplementation, and toxicity studies. Generally, Zn was found to be a significant factor for gut bacteria biodiversity. The effects of physiological and nutritional Zn doses also result in improved gut wall integrity, thus contributing to reduced translocation of bacteria and gut microbiome metabolites into the systemic circulation. In contrast, Zn overexposure induced substantial alterations in gut microbiota. In parallel with intestinal effects, systemic effects of Zn-induced gut microbiota modulation may include systemic inflammation and acute pancreatitis, autism spectrum disorder and attention deficit hyperactivity disorder, as well as fetal alcohol syndrome and obesity. In view of both Zn and gut microbiota, as well as their interaction in the regulation of the physiological functions of the host organism, addressing these targets through the use of Zn-enriched probiotics may be considered an effective strategy for health management. 相似文献

In this paper, we consider the problem of scheduling a set of jobs on two parallel machines with set-up times. The set-up has to be performed by a single server. The objective is to minimise the forced idle time. The problem of minimising the forced idle time (interference problem) is known to be unary NP-hard for the case of two machines and equal set-up and arbitrary processing times. We propose a mixed integer linear programming model, which describes a special class of schedules where the jobs from a list are scheduled alternatively on the machines, and a heuristic algorithm is tested on instances with up to 100,000 jobs. The computational results indicate that the algorithm has an excellent performance even for very large instances, where mostly an optimal solution is obtained within a very small computational time. 相似文献
