Multiple-allergen testing for high throughput and high sensitivity requires the development of miniaturized immunoassays that allow for a large test area and require only a small volume of the test analyte, which is often available only in limited amounts. Developing such miniaturized biochips containing arrays of test allergens needs application of a technique able to deposit molecules at high resolution and speed while preserving its functionality. Lipid dip-pen nanolithography (L-DPN) is an ideal technique to create such biologically active surfaces, and it has already been successfully applied for the direct, nanoscale deposition of functional proteins, as well as for the fabrication of biochemical templates for selective adsorption. The work presented here shows the application of L-DPN for the generation of arrays of the ligand 2,4-dinitrophenyl[1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine-N-[6-[(2,4-dinitrophenyl)amino]hexanoyl] (DNP)] onto glass surfaces as a model system for detection of allergen-specific Immunoglobin E (IgE) antibodies and for mast cell activation profiling.  相似文献

Anthropogenic activities generate a high quantity of organic pollutants, which have an impact on human health and cause adverse environmental effects. Monitoring of many hazardous contaminations is subject to legal regulations, but some substances such as therapeutic agents, personal care products, hormones, and derivatives of common organic compounds are currently not included in these regulations. Classical methods of removal of organic pollutants involve economically challenging processes. In this regard, remediation with biological agents can be an alternative. For in situ decontamination, the plant-based approach called phytoremediation can be used. However, the main disadvantages of this method are the limited accumulation capacity of plants, sensitivity to the action of high concentrations of hazardous pollutants, and no possibility of using pollutants for growth. To overcome these drawbacks and additionally increase the efficiency of the process, an integrated technology of bacteria-assisted phytoremediation is being used recently. For the system to work, it is necessary to properly select partners, especially endophytes for specific plants, based on the knowledge of their metabolic abilities and plant colonization capacity. The best approach that allows broad recognition of all relationships occurring in a complex community of endophytic bacteria and its variability under the influence of various factors can be obtained using culture-independent techniques. However, for practical application, culture-based techniques have priority.  相似文献

The aim of this work was to determine modifying effects of lactic fermentation and extrusion processes on functionality of lupin proteins. Protein content, surface hydrophobicity, water absorption capacity (WAC), water solubility index (WSI) and emulsifying properties (EAI, ESI) of protein preparations obtained from lupin seeds (Lupinus luteus, Lupinus albus, Lupinus angustifolius), with various contents of hull, were analyzed. Changes of protein properties were affected by lupin cultivar, hull content and applied processing method. An increase of soluble protein content after controlled lactic fermentation of lupin seeds, and changes of surface protein hydrophobicity, WAC and WSI values after each treatment and significant worsening of protein emulsifying properties were observed. Correlations were found between parameters examined in this study.  相似文献

The use of III-V materials as the channel in future transistor devices is dependent on removing the deleterious native oxides from their surface before deposition of a gate dielectric. Trimethylaluminium has been found to achieve in situ 'clean-up' of the oxides of GaAs and InGaAs before atomic layer deposition (ALD) of alumina. Here we propose six reaction mechanisms for 'clean-up,' featuring exchange of ligands between surface atoms, reduction of arsenic oxide by methyl groups and desorption of various products. We use first principles Density Functional Theory (DFT) to determine which mechanistic path is thermodynamically favoured based on models of the bulk oxides and gas-phase products. We therefore predict that 'clean-up' of arsenic oxides mostly produces As4 gas. Most C is predicted to form C2H6 but with some C2H4, CH4 and H2O. An alternative pathway is non-redox ligand exchange, which allows non-reducible oxides to be cleaned-up.  相似文献

This article presents the results from research related to graphene functionality based on the production of spatial structures provided for the reversible storage of hydrogen. The functionality process was conducted during graphene synthesis onto a liquid metallic support, on a single level, using SiC nanoparticles. Within the scope of research it was proved that heterogenic growth of the domains of polycrystalline graphene onto the SiC nanoparticles is possible. These nanoparticles are in-built into the graphene structure constituting the pillars of the spatial structure. Material produced in such a way constitutes the foundation for creating a spatial 3D structure (through the rolling operation), called GraphRoll, for the reversible storage of hydrogen in order to conduct its sorption and de-sorption. So, independently of the theoretical configuration, deviations or a possible exception from the 2D configuration on the silicon carbide/graphene were discussed. These differences resulted from the difference between the crystallographic structures of the analyzed forms as well as the structure determined to decrease tensions within the structure.  相似文献

In this paper the interaction of lubricant additives with hydrogen at the frictional interface has been investigated. Three different states of the base AISI 52100 alloy steel have been tested: untreated, nitrided and sulphonitrided, with different combinations of PAO6 base oil and ZDDP/MoDTC (Zinc DialkylDithioPhosphate and Molybdenum Dialkyldithiocarbamate) additives. Experiments have been carried out on pin-on-plate reciprocating tester, immersed in the lubricant heated to 100 °C. In the boundary lubricated regime the results showed the best friction behaviour for treated surfaces tested in presence of PAO6 with additives suggesting some interaction at the frictional interface of nitrided and sulphonitrided surfaces with lubricant additives. The minimum recorded value of coefficient of friction was as low as 0.05 for the sulphonitrided sample with PAO6+MoDTC oil. In the case of the treated surfaces a characteristic “low friction phase” has been observed when tested with PAO6 with additives. After a given time, the coefficient of friction was increasing to a higher steady-state value and the duration of this low friction phase varied from sample to sample. This can be explained by the mechanism of hydrogen interaction in the boundary lubrication regime, which was postulated for the base oil case by some of the authors in their previous papers. To validate the hypothesis, an experiment has been carried out where the test was stopped at the end of the “low friction phase” and during the hold period the sample was re-saturated with hydrogen. After resuming the experiment the low friction regime was again observed. The effect of a potential synergistic mechanism between hydrogen and ZDDP or MoDTC lubricant additives on frictional behaviour of nitrided and sulphonitrided surfaces is discussed in this paper.  相似文献

The combined effects of injection, packaging (modified atmosphere packaging [MAP] with 70% O2/ 30% CO2 and vacuum packaging [VP]), storage temperature (‐1 °C and +4 °C), and storage time on the color, microbial and oxidative stability of beef and bison longissimus lumborum (LL) steaks were investigated. Beef LL steaks in MAP retained their bright red color longer than bison steaks. Bison steaks developed higher 2‐Thiobarbituric acid reactive substances (TBARS) during storage, and this might have influenced the resulting rapid loss of redness from the bloomed meat. Storage at ‐1 °C in MAP provided greater color stability and a longer storage life for both meat species studied. Injection of salt/phosphate had a beneficial effect on the color stability of steaks during retail display; however, this positive effect was more pronounced for bison steaks compared with those of beef. Steaks stored overnight under MAP before retail display maintained the highest a* values for up to 5 d compared with those stored under vacuum. MAP‐OV steaks generally maintained the highest OMB content for up to 5 d during retail display compared with those stored under vacuum. Nevertheless, OMB levels were significantly (P < 0.05) lower in bison steaks compared with those of beef irrespective of packaging treatments. Injected steaks and those stored at ‐1 °C had significantly (P < 0.05) higher OMB levels compared with non‐injected counterparts and those stored at +4 °C, respectively. MAP is an excellent option for short‐term storage due to its positive effects on meat color, but for longer storage, VP may be necessary. Storing meat under vacuum and then placing it under MAP just before retail display might be another option to increase shelf life.  相似文献

The combined effect of incorporation of four non-muscle proteins, NMP (blood plasma, BP; sodium caseinate, SC; soy protein isolate, SPI; gelatin, G) at 2 g/100 g levels on hydration and textural characteristics of pork gels processed without or with 0.6 g/100 g microbial transglutaminase preparation (MTG) was investigated. Addition of SC and BP most favourably affected hydration properties and thermal stability, yielding lower cooking loss and expressible moisture for pork gels. Interactions between NMP and MTG were observed. Improvement of gel strength by addition of transglutaminase was observed for treatments containing SC and BP but not G nor soy isolate. Of the four proteins tested SC was found to be a superior substrate for MTG in enhancing textural properties of a gelled meat system. None of the tested ingredients was able to yield gel cohesiveness equivalent to the control containing 8% muscle proteins. Results of this study indicate a potential for using MTG to improve or modify the functional and textural properties of investigated food proteins (SC and BP in particular) in comminuted meat products.  相似文献

The paper presents the determination of iron forms in food products. The procedure of sample extraction was developed and optimized, preserving the content of particular forms of iron. The colorimetric method using 2,2′-bipirydyl (measurements at 520 nm) was applied in Fe(II) determinations, while in Fe(III) determinations, the colorimetric method with potassium thiocyanate (measurements at 470 nm) was applied. The total content of iron was determined by the technique of atomic absorption spectrometry, which allowed for the determination of iron content in organic and inorganic complex compounds. Detection limits of 1 mg kg?1 were obtained for all determined iron forms, with the precision ranging between 0.7 % and 1.5 % for 10 mg kg?1 concentration. The optimized analytical procedure was applied in the determinations of iron forms in iron-fortified food products.  相似文献
