For detection and quantification of food proteins triggering hypersensitivity reactions, reliable, validated analytical methods are necessary. At present, the most commonly used methodology for this purpose is ELISA. The lack of incurred reference materials (RM) complicates the fulfilling of method validation. Today, a well identifiable research direction is the development of methods (immunoanalytical, PCR and MS methods) suitable for multi-component analysis. As a consequence of these R+D efforts, the development of multi-component RMs is reasonable. Our work presented here is focusing on the development and investigation of multi-component model food matrices that can be suitable for supporting the validation processes. On the basis of the relative incidence of hypersensitivity reactions, four components—wheat, milk, egg and soy—were selected for our preliminary work. The target proteins were determined in single- and multi-component matrices with different ELISA methods, and the results were compared by statistical evaluation. The main direction of our investigation was to identify the effects of the potential interactions of these proteins on the analytical results, and the influence of food processing was also investigated. The results showed that the presence of multiple allergenic components caused much less uncertainty in the analytical results than analytical and technological steps, such as sample preparation, repetition or even heat treatment. However, probably due to the relatively complex matrices, some discrepancies from the expected outcome could also be observed. 相似文献

Meat contaminating bacteria may be the direct cause of foodborne diseases and represent a potential cause for the drug resistance of human pathogenic agents. The prevalence and resistance to 17 antimicrobial drugs of isolates of selected bacterial species were investigated in 70 swabs of beef carcasses and 70 subsequent samples of beef meat. Molecular techniques (coagulase gene typing Staphylococcus aureus and original gene typing Escherichia coli) were used in the differentiation of isolates. Carcasses were already contaminated after evisceration, least frequently with S. aureus strains (7.5% of samples), most frequently with coagulase-negative staphylococci strains (52.2% of samples). During carcass processing, contamination with resistant or polyresistant strains of S. aureus and E. coli significantly increased (P<0.01). Gene typing isolates of S. aureus and E. coli indicated that the strains probably originated in the processing plant. 相似文献

Abstract We discuss the performance of “closed-end” fluctuation tests, used to detect changes in the structural stability of a model, in the case where the time horizon is large and the observations do not possess high enough moments. It is demonstrated via a simulation study that, in the latter case, the choice of the boundary function should take into account the number of moments existing; otherwise the actual size of the test may exceed the nominal level. We suggest an alternative “open-end” fluctuation procedure and study its asymptotic behavior. It turns out that if an improper boundary function is chosen, then the null hypothesis may be rejected even if it is true. This means that an appropriate choice of the boundary function requires some prior information about the tail heaviness of the observations. This is different from the cumulative sum-based monitoring schemes suggested by Chu et al. (1996) and further studied by Horváth et al. (2004). 相似文献

Uncertainties in flood predictions complicate the planning of mitigation measures. There is a consensus that many possible incident scenarios should be considered. For each scenario, a specific response plan should be prepared which is optimal with respect to criteria such as protection, costs, or realization time. None of the existing software tools is capable of creating large scenario pools, nor do they provide means for quick exploration and assessment of the associated plans. In this paper, we present an integrated solution that is based on multidimensional, time‐dependent ensemble simulations of incident scenarios and protective measures. We provide scalable interfaces which facilitate and accelerate setting up multiple time‐varying parameters for generating a pool of pre‐cooked scenarios. In case of an emergency, disaster managers can quickly extract relevant information from the pool to deal with the situation at hand. An interactive 3D‐view conveys details about how a response plan has to be executed. Linked information visualization and ranking views allow for a quick assessment of many plans. In collaboration with flood managers, we demonstrate the practical applicability of our solution. We tackle the challenges of planning mobile water barriers for protecting important infrastructure. We account for real‐world limitations of available resources and handle the involved logistics problems. 相似文献

The authors are discussing hepatic and extrahepatic pathologic processes caused by hepatitis C virus (HCV) infection and they focus their interest to the skin disorders appearing in the presence of chronic, active HCV infections. The trigger of the immunologic processes leading to dermatologic manifestations are the activated T cells (CD8 + cytotoxic T lymphocytes), cytokins, and also the expansion of certain B cells. Pathologic immunologic phenomena may initiate various dermatologic manifestations. Immunoglobulins, immuncomplexes generated by the disease itself are manifested as various forms of cutan vasculitis. In the present series of patients (pts), HCV related skin disorders known from the literature were diagnosed in eleven cases and they were representing 7 different disease entities. These were palpable purpura (3 pts), urticaria, prurigo and alopecia areata (2-2 pts), lichen ruber planus, pruritus and vitiligo (1-1 patient respectively). Newborn A mice were injected either with a single i.v. dose (Group A) or with repeated doses of (B10 x A)F1 semiallogeneic spleen cells (SSC) (Group B). A similar degree of partial transplantation tolerance (TT) to B10 skin allografts was revealed in both groups. No signs of acute, rapidly fatal host-versus-graft disease (HVGD) (anemia, leukocytosis, severe thrombocytopenia, hepatic infarcts, gastrointestinal bleedings) were found, rather a chronic HVGD developed [moderate thrombocytopenia, autoimmune antithymocyte antibodies (ATA)] in both groups. The mortality due to lymphoproliferative disorders (LPD) was significantly higher in Group A. Thus, repeated perinatal injections of (B10 x A)F1 SSC into A mice did not increase the tolerogenic and the LPD-inducing effect either, and they did not elicit acute HVGD in contrast to observations in other F1 donor-recipient combinations [1]. We develop tests of normality for time series of functions. The tests are related to the commonly used Jarque–Bera test. The assumption of normality has played an important role in many methodological and theoretical developments in the field of functional data analysis. Yet, no inferential procedures to verify it have been proposed so far, even for i.i.d. functions. We propose several approaches which handle two paramount challenges: (i) the unknown temporal dependence structure and (ii) the estimation of the optimal finite‐dimensional projection space. We evaluate the tests via simulations and establish their large sample validity under general conditions. We obtain useful insights by applying them to pollution and intraday price curves. While the pollution curves can be treated as normal, the normality of high‐frequency price curves is rejected. 相似文献
