Here the interaction between a laminar two-phase, non-pre-mixed counterflow flame and a vortex is examined. Special emphasis is given to the influence of different flame and vortex parameters on the extinction behavior of the flame. Simultaneous planar laser-induced fluorescence of the CH radical layer produced by the flame and particle-image velocimetry measurements of the flowfield are used to characterize the flame-vortex interaction. These simultaneous diagnostics are used for the first time in this configuration. The extinction processes occurring during the flame-vortex interaction can be analyzed by this method, especially the influence of strain at the flame surface. The influence of the droplets on the extinction behavior appears clearly compared with a fully gaseous flame. The spray flame is weaker and extinguishes earlier than does a gaseous flame. In the measurements an additional broadband signal in the vicinity of the CH layer is probably due to the induced fluorescence of polycyclic aromatic hydrocarbons, excited at the same wavelength.  相似文献

An autonomous sensor for long-term in situ measurements of the pH of natural waters is described. The system is based upon spectrophotometric measurements of a mixture of sample and sulfonephthalein indicator. A simple plumbing design, using a small, low-power solenoid pump and valve, avoids the need for quantitative addition of indicator. A approximately 50-microL slug of indicator is pulled into the sample stream by the pump, and subsequent pumping and mixing results in a section of indicator and sample where absorbance measurements can be made. The design also permits direct determination of the indicator pH perturbation. Absorbances are recorded at three wavelengths (439, 579, and 735 nm) using a custom-built 1.7-cm path length fiber-optic flow cell. Solution blanks are obtained by periodically flushing the cell with sample. Field tests were performed in a local river over an 8-day period. The in situ accuracy, based on comparison with laboratory spectrophotometric pH measurements, was -0.003 pH unit (n = 16), similar to the measurement precision. No drift was observed during the 8-day period. The absorbance ratio used to calculate pH, in combination with a simple and robust optical design, imparts an inherent stability not achievable with conventional potentiometric methods, making the design feasible for long-term autonomous pH measurements.  相似文献

An approach for the characterization of glycosylation sites and oligosaccharide heterogeneity in glycoproteins based on a combination of nonspecific proteolysis, deglycosylation, and matrix-assisted laser desorption/ionization Fourier transform mass spectrometry (MALDI-FT MS) is described. Glycoproteins were digested with Pronase yielding primarily glycopeptides and amino acids. Nonglycosylated peptide fragments were susceptible to complete Pronase digestion to their constituent amino acids. Steric hindrance prohibited the digestion of the peptide moiety attached to the glycan. Glycopeptides were desalted and concentrated using solid-phase extraction and analyzed by MALDI MS. The oligosaccharides were also analyzed by MALDI MS after releasing the glycans from glycoproteins using PNGase F. The peptide moiety of the glycopeptides was identified by subtracting the masses of the glycans derived from PNGase F treatment from the masses of the glycopeptides. The experimental strategy was validated using glycoproteins with known oligosaccharide structures, ribonuclease B and chicken ovalbumin. This procedure was then used to determine the N-glycosylation sites and site heterogeneity of a glycoprotein whose glycosylation pattern was unknown, namely, the Xenopus laevis egg cortical granule lectin. This procedure is useful for determining protein site heterogeneity and structural heterogeneities of the oligosaccharide moiety of glycoproteins.  相似文献

The aim of this study was to investigate the effects of conjugated linoleic acid supplementation on the synthesis of milk fat in pasture-fed Friesian cows. In four cows, a commercial mixture containing 62.3% (wt/vol) conjugated linoleic acid was infused intraabomasally to avoid rumen fermentation and biohydrogenation. The design was a 4 x 4 Latin square in which each cow received infusions of 0, 20, 40, and 80 g/d of conjugated linoleic acid mixture for 4 d. Cows were fed freshly cut ryegrass/white clover pasture ad libitum. Milk fat concentration was decreased by 36, 43, and 62% and milk fat yield was decreased by 32, 36, and 60% by the 20, 40, and 80 g of conjugated linoleic acid/d treatments. Dry matter intake, milk protein concentration, and protein yield were unaffected by treatments; however, milk yield was increased by 11% during the 40-g conjugated linoleic acid/d treatment. The effects of conjugated linoleic acid infusion were most pronounced in reducing de novo fatty acid synthesis and desaturation. Results show that the inhibitory effect of this conjugated linoleic acid mixture on milk fat synthesis occurs in pasture-fed cows, and demonstrate the potential to dramatically alter gross milk composition. This technology could offer a management tool to manipulate milk composition and energy demands of pasture-fed cows.  相似文献

Four algorithms used to simulate pasture intake in grazing dairy cows in a dairy decision support system were proposed and evaluated with data from the literature. The algorithms proposed were: 1) an algorithm combining the approach used in a published model to determine dry matter intake based on neutral detergent fiber intake as a percentage of the BW, energy requirements, pasture availability and a standard supplementation (PIest), 2) the previous algorithm modified to consider the type and amount of supplementation (PIsup), 3) an algorithm which considers the effect of selection of pasture (PIsel), and 4) the combination of algorithms 2 and 3 (PIsupsel). Pasture intake data of 27 grazing experiments from the literature were used to evaluate those algorithms. Two methods of evaluation were used: 1) simple linear regression between reported and simulated values, and 2) analysis of variance for the difference between reported and simulated values considering pasture availability and type of supplementation. The R2 of the linear regression and average proportional bias between reported values and simulated values were 0.24 and 19% for PIest, 0.42, and 23% for PIsup, 0.45 and 2% for PIsel and 0.41 and 10% for PIsupsel. Those results showed that PIsel had the lower variability and the values closer to pasture intake. The algorithm PIsup had low variability but tended to underpredict pasture intake. The algorithm PIest values were closer to reported values for low pasture availability. The modeling results show the influence of pasture selection in grazing systems.  相似文献

High-resolution nuclear magnetic resonance (NMR) spectroscopy has provided a new means for detection and recognition of metabolic changes in biological systems in response to pathophysiological stimuli and to the intake of toxins or nutrition. To identify meaningful patterns from NMR spectra, various statistical pattern recognition methods have been applied to reduce their complexity and uncover implicit metabolic patterns. In this paper, we present a genetic algorithm (GA)-based feature selection method to determine major metabolite features to play a significant role in discrimination of samples among different conditions in high-resolution NMR spectra. In addition, an orthogonal signal filter was employed as a preprocessor of NMR spectra in order to remove any unwanted variation of the data that is unrelated to the discrimination of different conditions. The results of k-nearest neighbors and the partial least squares discriminant analysis of the experimental NMR spectra from human plasma showed the potential advantage of the features obtained from GA-based feature selection combined with an orthogonal signal filter.  相似文献

针对国家提出的加快煤层气地面开发和推广煤矿"先抽后建"理念,分析了新建突出煤矿"先抽后建"的基本特征,构建了"先抽后建"的时间-空间-安全条件模式模型与政策落实-技术研究-工程实施路径图,并对官寨煤矿工程实例进行了研究。研究得出:煤层气地面井抽采是煤矿区域瓦斯治理的创新举措,是推动实现新建突出煤矿"先抽后建"的有效途径;"先抽后建"模式为科学规划煤层气抽采与矿井建设的工程分区,统筹衔接和协调实施煤层气与煤炭开发;实施路径是在煤矿筹建阶段明确落实"先抽后建"政策的总体方案,根据项目可行性研究的成果,研究编制"先抽后建"的技术实施方案,按照地面钻井-瓦斯先抽-矿井后建的步骤进行"先抽后建"的工程实施。现场工程实践显示,官寨井田煤层气开发与矿井建设工程"先抽后建"方案可行,具有典型的代表性和很好的借鉴意义。  相似文献

BACKGROUND: Chloroform, ethyl acetate and methanol extracts of a sample of red propolis from the state of Alagoas (northeast Brazil) were analyzed by gas chromatography–mass spectrometry and high‐performance liquid chromatography–diode array detection–electrospray ionization‐mass spectrometry. Antimicrobial and antioxidant activities were also obtained. RESULTS: The propolis sample contained low content of narigenin‐8‐C‐hexoside, this being the first report of a C‐glycoside in propolis. The main constituent found was characterized as 3,4,2′,3′‐tetrahydroxychalcone. Other important constituents were the chalcone isoliquiritigenin, the isoflavans (3S)‐vestitol, (3S)‐7‐O‐methylvestitol, the pterocarpan medicarpin, the phenylpropenes trans‐anethol, methyl eugenol, elimicin, methoxyeugenol and cis‐asarone, and the triterpenic alcohols lupeol and α‐ and β‐ amyrins. The methanol extract exhibited high antioxidant activities by 2,2‐diphenyl‐1‐picrylhydrazyl and β‐carotene/linoleic acid assay methods, and antimicrobial activity toward Gram‐positive and Gram‐negative bacteria. CONCLUSION: Structures are suggested for new substances never before seen in any kind of propolis. This is the first report of 3,4,2′,3′‐tetrahydroxychalcone and a flavone C‐glycoside in a propolis sample. Copyright © 2011 Society of Chemical Industry  相似文献
