The intensity of amyloid-bound thioflavine T fluorescence was studied in crude lysates of yeast strains carrying mutations in the ADE1 or ADE2 genes and accumulating the red pigment (a result of polymerization of aminoimidazoleribotide), and in white isogenic strains--either adenine prototrophs or carrying mutations at the first stages of purine biosynthesis. We found that the red pigment leads to a drop of amyloid content. This result, along with the data on separation of protein polymers of white and red strains in PAGE, suggests that the red pigment inhibits amyloid fibril formation. The differences in transmission of the thioflavine T fluorescence pattern by cytoduction and in blot-hybridization of pellet proteins of red and white [PSI(+) ] strains with Sup35p antibodies confirmed this conclusion. Purified red pigment treatment also led to a decrease of fluorescence intensity of thioflavine T bound to insulin fibrils and to yeast pellet protein aggregates from [PSI(+) ] strains. This suggests red pigment interaction with amyloid fibrils. Comparison of pellet proteins from red and white isogenic strains separated by 2D-electrophoresis followed by MALDI analysis has allowed us to identify 48 pigment-dependent proteins. These proteins mostly belong to functional classes of chaperones and proteins involved in glucose metabolism, closely corresponding to prion-dependent proteins that we characterized previously. Also present were some proteins involved in stress response and proteolysis. We suppose that the red pigment acts by blocking certain sites on amyloid fibrils that, in some cases, can lead in vivo to interfere with their contacts with chaperones and the generation of prion seeds.  相似文献

The study was designed to test the effects of feeding fish meal (FM) and specific n-3 fatty acids on milk yield and composition, dry matter intake, plasma concentrations of metabolic hormones and metabolites, and liver triglyceride accumulation in early lactating cows. From 5 to 50 d in milk (DIM), cows were fed diets that were isonitrogenous, isoenergetic, and isolipidic containing none (control), 1.25, 2.5, or 5% menhaden FM or 2.3% Ca salts of fish oil fatty acids (CaFOFA). Milk yield (48.2, 49.8, 48.6, 53.5, and 52.2 ± 1.0 kg/d, respectively) and dry matter intake (22.7, 22.8, 23.0, 23.8, and 24.7 ± 0.5 kg/d, respectively) differed among diets. Average daily plasma glucose concentration (53.4, 55.3, 51.1, 57.6, and 57.3 ± 1.3 mg/dL, respectively) was also affected by diet, and plasma insulin concentration was increased by 5% FM and 2.3% Ca-FOFA. At 25 and 50 DIM, blood was collected before feeding and hourly for 11 h after feeding. Plasma glucose concentrations in cows during the day were similar among diets at 25 DIM, but differed at 50 DIM (54.6, 54.4, 52.4, 60.5, and 58.3 ± 1.4 mg/dL for 0, 1.25, 2.5, and 5% FM or 2.3% CaFOFA, respectively). Plasma insulin was increased in cows fed 5% FM and 2.3% CaFOFA at 25 DIM and was similar among diets at 50 DIM. Dietary treatments had no significant effect on milk composition, energy balance, or on daily plasma concentrations of nonesterified fatty acids, β-hydroxybutyrate, and urea. Plasma aspartate aminotransferase and hepatic triglyceride concentration in cows did not differ among diets at 21 DIM. Results from this experiment demonstrate that dietary supplementation with FM or n-3 polyunsaturated fatty acids in early lactating dairy cows significantly increased milk yield and DMI with no change in milk composition.  相似文献

Our study investigated the separate and combined effects of branched-chain amino acids (AA) and sodium caseinate on milk protein concentration and yield. Four Holstein cows (112 d in milk) were abomasally infused with water, branched-chain AA (150 g/d), sodium caseinate (600 g/d), or branched-chain AA plus sodium caseinate (44 and 600 g/d, respectively) according to a 4 x 4 Latin square design with 8-d treatment periods. Cows were fed a dry diet based on alfalfa hay and concentrates for ad libitum intake. The ration was formulated to exceed requirements for metabolizable energy and protein using the Cornell Net Carbohydrate and Protein System. Neither daily dry matter intake (24.2 +/- 0.4 kg/d; X +/- SEM) nor milk yield (32.9 +/-; 0.4 kg/d) was affected by any of the infusion treatments. Infusion of branched-chain AA had no effect on any milk production parameters, despite a 50% increase in their concentrations. Modest increases in milk protein concentration (0.1%) and milk protein yield (62 g/d) resulted from the infusion of sodium caseinate or branched-chain AA plus sodium caseinate. True protein and whey protein concentrations in milk were also marginally increased by infusion of sodium caseinate and branched-chain AA plus sodium caseinate, and infusion of branched-chain AA, sodium caseinate, or both elevated milk nonprotein N content. Plasma urea N concentrations were elevated by the sodium caseinate and branched-chain AA plus sodium caseinate treatments. No treatment effects on other plasma metabolites or hormones were observed. Our results show no benefit of supplementation with branched-chain AA and only modest effects of sodium caseinate on milk protein concentration and yield in well-fed cows.  相似文献

Biodegradation of a mixture containing seven commercial textile dyes with different structures and color properties has been investigated by an ecofriendly strain--Brevibacillus laterosporus MTCC 2298. It showed 87% decolorization in terms of ADMI removal (American Dye Manufacturing Institute) within 24h. The effective decolorization of dye mixture was attained in the presence of metal salt--CaCl(2) and nitrogen sources. The induction of oxido-reductive enzymes such as veratryl alcohol oxidase, tyrosinase, NADH-DCIP reductase and azo reductase was found to be responsible for biotransformation of dyes. High performance thin layer chromatography exposed the mechanism of preferential biodegradation of dyes at different time periods. Significant change in the high pressure liquid chromatography and Fourier transform infrared spectroscopy of sample before and after treatment confirmed the biodegradation of dye mixture. Phytotoxicity study revealed the much less toxic nature of the metabolites produced after the degradation of dyes mixture.  相似文献

A lightweight (60 g), personal nanoparticle respiratory deposition (NRD) sampler was developed to selectively collect particles smaller than 300 nm similar to their typical deposition in the respiratory tract. The sampler operates at 2.5 Lpm and consists of a respirable cyclone fitted with an impactor and a diffusion stage containing mesh screens. The cut-point diameter of the impactor was determined to be 300 nm with a sharpness σ = 1.53. The diffusion stage screens collect particles with an efficiency that matches the deposition efficiency of particles smaller than 300 nm in the respiratory tract. Impactor separation performance was unaffected by loading at typical workplace levels (p-value = 0.26). With chemical analysis of the diffusion media, the NRD sampler can be used to directly assess exposures to nanoparticles of a specific composition apart from other airborne particles. The pressure drop of the NRD sampler is sufficiently low to permit its operation with conventional, belt-mounted sampling pumps.  相似文献

This study presents the life cycle assessment (LCA) of three batteries for plug-in hybrid and full performance battery electric vehicles. A transparent life cycle inventory (LCI) was compiled in a component-wise manner for nickel metal hydride (NiMH), nickel cobalt manganese lithium-ion (NCM), and iron phosphate lithium-ion (LFP) batteries. The battery systems were investigated with a functional unit based on energy storage, and environmental impacts were analyzed using midpoint indicators. On a per-storage basis, the NiMH technology was found to have the highest environmental impact, followed by NCM and then LFP, for all categories considered except ozone depletion potential. We found higher life cycle global warming emissions than have been previously reported. Detailed contribution and structural path analyses allowed for the identification of the different processes and value-chains most directly responsible for these emissions. This article contributes a public and detailed inventory, which can be easily be adapted to any powertrain, along with readily usable environmental performance assessments.  相似文献

Advances in nanophotonics have shown the potential of colloidal metal nanoparticles with sharp tips, such as rods, to focalize plasmonic electromagnetic fields. We report on the synthesis of Au nanorods via a seed mediated approach and the influence of silver ions on the aspect ratio of the Au nanorods. The longitudinal surface plasmon resonance (LSPR) of the Au nanorods was successfully tuned with the concentration of silver ions. The surface enhanced Raman scattering (SERS) effect of 2-aminothiophenol (2-ATP) as a probe molecule on Au nanorods was systematically studied by varying the longitudinal surface plasmon resonance of the nanorods. The highest electromagnetic enhancement was observed when the longitudinal surface plasmon resonance of the Au nanorods overlapped with the laser excitation wavelength. The variation of the SERS enhancement factor with the longitudinal surface plasmon resonance and laser excitation lines is also discussed in detail.  相似文献

The ability to resolve isomeric protonated dipeptides was investigated with the new technique of differential ion mobility mass spectrometry that uses "modifier" molecules to enhance differential mobility. Two pairs of protonated peptides [glycine-alanine (GlyAla) and alanine-glycine (AlaGly), glycine-serine (GlySer) and serine-glycine (SerGly)] and eight different modifiers (water, 2-propanol, 1,5-hexadiene, 2-chloropropane, chlorobenzene, dichloromethane, acetonitrile, and cyclohexane) were used in the initial study. Separation of the protonated peptides was found to be dependent on the mass and proton affinity of the modifier and combinations of functionalities present in the modifier and the analyte ion. Six of the eight modifiers (water, 2-propanol, chlorobenzene, cyclohexane, dichloromethane, and acetonitrile) were able to separate the protonated isomeric peptide pairs, and generally, modifiers with electron-rich groups performed the best. In the presence of some modifiers, a reduction of ion current was observed under the highest field conditions (>115 Td). Dopant-catalyzed isomerization, likely by proton-transport catalysis, and field-induced fragmentation may have contributed to these losses. Two high vapor pressure modifiers, 1,5-hexadiene and 2-chloropropane, significantly influenced ion formation leading to the formation of stable cluster populations that could be observed in the mass spectrometer. Although not a major concern, both fragmentation and influence of modifier evaporation warrant further studies in order to fully understand and possibly eliminate them.  相似文献
