The bacterial community structure in bulk water and in rhizosphere fractions of giant duckweed, Spirodela polyrrhiza, was quantitatively and qualitatively investigated by PCR-based methods using 6 environmental water samples to elucidate the mechanisms underlying selective accumulation of aromatic compound-degrading bacteria in the rhizosphere of S. polyrrhiza. S. polyrrhiza selectively accumulated a diverse range of aromatic compound-degrading bacteria in its rhizosphere, regardless of the origin of water samples, despite no exposure to phenol. The relative abundances of the catechol 1,2-dioxygenase (C12O) gene (C12O DNA) and catechol 2,3-dioxygenase (C23O) gene (C23O DNA) were calculated as the ratios of the copy numbers of these genes to the copy number of 16S rDNA and are referred to as the rhizosphere effect (RE) value. The RE values for C12O DNA and C23O DNA were 1.0 × 101–9.3 × 103 and 1.7 × 102–1.5 × 104 times as high, respectively, in rhizosphere fractions as in bulk water fractions, and these higher values were associated with a notably higher sequence diversity of C12O DNA and C23O DNA. The RE values during phenol degradation were 3.6 × 100–4.3 × 102 and 2.2 × 100–1.7 × 102, respectively, indicating the ability of S. polyrrhiza to selectively accumulate aromatic compound-degrading bacteria in its rhizosphere during phenol degradation. The bacterial communities in the rhizosphere fractions differed from those in the bulk water fractions, and those in the bulk water fractions were notably affected by the rhizosphere bacterial communities. S. polyrrhiza released more than 100 types of phenolic compound into its rhizosphere as root exudates at the considerably high specific release rate of 1520 mg TOC and 214 mg phenolic compounds/d/g root (wet weight). This ability of S. polyrrhiza might result in the selective recruitment and accumulation of a diverse range of bacteria harboring genes encoding C12O and C23O, and the subsequent accelerated degradation of phenol in the rhizosphere.  相似文献

The film thickness, d of a p-hydroxymethylbenzene - C6H4CH2OH (HOMB) self-assembled monolayer (SAM) adsorbed on iron via a covalent bond was increased by multistep modification with tetraethoxysilane (C2H5O)4Si (TES) and 1,8-octanediol HO(CH2)8OH (C8DO) and subsequently with 1,2-bis(triethoxysilyl)ethane (C2H5O)3Si(CH2)2Si(OC2H5)3 (BTESE) and akyltriethoxysilane CnH2n+1Si(OC2H5)3 (CnTES, n = 8 or 18). The protective ability of the film against iron corrosion was examined by polarization measurement of the iron electrode coated with the film in an oxygenated 0.5 M NaCl after immersion in the solution for many hours. The values of protective efficiency, P for the films, the HOMB SAM modified with TES and C8DO twice and subsequently with BTESE and CnTES were high, more than 81% at n = 8 and 85% at n = 18 in the range of the immersion time, t up to 240 h, respectively. The maximal P values of the respective films were 88.0% and 92.2%, of which approximate d values were 4.6 and 5.8 nm. The high protection of iron against corrosion was attributed to increases in the film thickness and interconnection between the adsorbed molecules with Si-O polymer linkages. The protective ability of the film was remarkably persistent during immersion in the solution for many hours. A slight enhancement of P was observed by additional modification of the modified HOMB SAM with C8TES due to increases in interconnection and close packed arrangement. The formation of strong adsorption bonds, σ-covalent bond and back-donating π-bond, between the carbon atom of HOMB moiety and iron atom was discussed.  相似文献

A film of two-dimensional polymer was prepared on an iron electrode passivated in a borate buffer solution at pH 8.49, derivatized with 16-hydroxyhexadecanoate ion and then modified with 1,2-bis(triethoxysilyl)ethane (C2H5O)3Si(CH2)2Si(OC2H5)3 and octadecyltriethoxysilane C18H37Si(OC2H5)3. The protective ability of the polymer film adsorbed on passivated iron was examined by polarization measurement in oxygenated 0.1 M KClO4 solutions with and without 1 × 10−4 and 1 × 10−3 M of Cl. The values of the open-circuit potential, Eoc were monitored with the immersion time, t in these solutions. The Eoc value of the passivated electrode in 0.1 M KClO4 was maintained high, more than −0.2 V/SCE in the initial region of t up to 10 h, indicating the presence of a passive film on the electrode. Thereafter, Eoc decreased to −0.4 V/SCE abruptly, exhibiting breakdown of the passive film. The value of the passivated electrode covered with the polymer film remained almost constant around −0.04 V/SCE during immersion for 45 h. The protective efficiency, P (%) of the polymer film on passivated iron was extremely high, more than 99.9% unless the passive film was broken down, indicating complete protection of iron against corrosion. The times for breakdown on the passivated electrode and polymer-coated one diminished with an increase in the concentration of Cl. The polymer-coated surface was analyzed by electron-probe microanalysis after immersion in 0.1 M KClO4 for 24 h.  相似文献

A side-chain liquid-crystalline polymer (LCP) was synthesized by the addition of the mesogenic monomer to poly(methyl siloxane) in presence of a Pt-catalyst. When an aqueous solution of 10wt% ethanol was permeated through a LCP membrane by pervaporation at various temperatures, the permeation rate increased with increasing temperature and drastically changed at glass-nematic (Tg) and nematic-isotropic (TNI) transition temperatures of the LCP membrane. The LCP membrane exhibited the waterpermselectivity in the glassy and liquid-crystalline states. The ethanol concentration in the permeate increased with increasing permeation temperature and the LCP membrane changed from the waterpermselectivity to the ethanol-permselectivity around TNI. These results suggested that the permselectivity was influenced by the change of the LCP membrane structure, that is, its state transformation. It was found that a balance of the orientation of mesogenic groups and flexibility of siloxane chains is very important for the permeability and selectivity.  相似文献

An ultrathin, ordered and two-dimensional polymer coating was prepared on passivated iron by modification of 16-hydroxyhexadecanoate ion HO(CH2)15CO2 self-assembled monolayer (SAM) with 1,2-bis(triethoxysilyl)ethane (C2H5O)3Si(CH2)2Si(OC2H5)3 and octadecyltriethoxysilane C18H37Si(OC2H5)3. Protection of passivated iron against passive film breakdown and corrosion of iron was examined by monitoring of the open-circuit potential and repeated polarization measurements in an aerated 0.1 M NaNO3 solution during immersion for many hours. Passive film breakdown on the polymer-coated electrode in the solution was not observed during immersion for 480 h, whereas that of the passivated one occurred at 18.1 h, indicating protection of the passive film from breakdown by coverage with the polymer coating. The protective efficiencies of the passive film covered with the coating were extremely high, around 99.9% in the initial region of the immersion time up to 72 h and more than 98.3% thereafter, indicating prominent cooperative suppression of iron corrosion in 0.1 M NaNO3 by coverage with the passive film and polymer coating. The polymer-coated surface was characterized by contact angle measurement and electron-probe microanalysis.  相似文献

Terpendole E is first natural product found to inhibit mitotic kinesin Eg5, but its inhibitory mechanism remains to be revealed. Here, we report the effects of terpendole E and 11ketopaspaline (a new natural terpendole E analogue) on the Eg5–microtubule interaction and in several Eg5 mutants. 11‐Ketopaspaline is a shunt product from terpendole E, and it shows potent inhibitory activity against the microtubule‐stimulated ATPase activity of Eg5. Unlike other Eg5 inhibitors, such as S‐trityl‐L ‐cysteine (STLC) and GSK‐1, both terpendole E and 11‐ketopaspaline only partially inhibited Eg5–microtubule interaction. Furthermore, terpendole E and 11‐ketopaspaline inhibited several Eg5 mutants that are resistant to STLC (Eg5D130A, Eg5L214A) or GSK‐1 (Eg5I299F, Eg5A356T), but with the same extent of inhibition against wild‐type Eg5. Because Eg5D130A and Eg5L214A show cross‐resistance to most known Eg5 inhibitors, which bind the L5 loop, these results suggest that terpendole E and its analogues have a different binding site and/or inhibitory mechanism to those for L5 loop‐binding type Eg5 inhibitors.  相似文献

Self-organized rhodamine 6G particles prepared by wetting/dewetting process of an ethanol solution on a hydrophilic glass surface exhibits fluorescence without quenching, showing a sharp linewidth of 2nm with a large redshift, which indicates an existence of dye aggregates, similar to J-aggregates, inside the particle. Polarized evanescent field excitation showed that the dye molecule's transition moment along the pi-conjugation was oriented unidirectionally within particles and parallel to the substrate surface. This deduced dye orientation showed correlation between adjacent, but separated, particles and pointed roughly 45 degrees off the dewetting direction. In contrast, the particles of another pi-conjugated NK1420 dye, J-aggregates of which grows easily from an oversaturated solution, showed dye orientation along the dewetting direction preferably, still indicating the effect of self-organization, however based on a different mechanism. An annealing procedure revealed that both aggregates are in quasi-stable states, which is consistent with the rapidness of the dewetting process that may lead to crystallization in nonequilibrium.  相似文献

Mutations around His15 which lie far away from the active site,stimulated glycol chitin activity of lysozyme at physiologicaltemperature. Del-Argl4Hisl5 lysozyme, a mutant lysozyme whoseArgl4 and Hisl5 were deleted together, and has the highest activityamong these mutant lysozymes, had a similar binding abilityto a trimer of N-acetyl-glucosamine, a substrate analogue, relativeto native lysozyme. This suggests that the increased activitywas due to an increased kcat in the catalysis reaction. TheH-D exchange rate of the N-1 proton in the Trp63 which is locatedin the active site cleft, was enhanced in the Del-Argl4Hisl5lysozyme, while 2-D proton NMR analysis revealed no conformationalchange around Trp63. We conclude that some sort of fluctuationat the active site might be required for the manifestation ofactivity. This theory is supported by the finding that the Del-Argl4Hisl5lysozyme showed a shift in temperature dependency of activityto lower temperatures compared with that of native lysozyme.  相似文献

Gel-derived titania coating on commercial pure (c.p.) titanium induces hydroxyapatite formation onto its surface from a simulated body fluid (SBF, a metastable calcium phosphate solution). The induced apatite is similar to bone apatite in that it is poorly crystallized, calcium-deficient, and carbonate-containing. Furthermore, the carbonate (CO2–3) groups go into the apatite lattice and lie at the positions of PO3–4 and OH to replace these ionic groups, resembling the (CO2–3) groups of bone apatite. Therefore, the apatite induced by the gel-derived titania is said to be bonelike. A chemical stimulation, stemming from abundant hydroxyl groups and negative charges at the surface of the titania gel, is believed to be responsible for the bonelike apatite induction. The potential of bone-bonding is predicted for the gel-derived titania, for it is an efficient bonelike apatite inducer in the SBF.  相似文献

研究了超声波探头位置变化对Al-5.7%Cu合金铸锭凝固组织的影响,考察了探头水平位置变化对等轴晶占有率的影响,探讨了超声波作用下晶粒细化机理。结果表明:当探头浸入深度相同且位于铸型中心线时,得到的细等轴晶区最大,且呈对称分布;当探头的位置贴近铸型壁时,细等轴晶区向探头端面附近集中并缩减,柱状晶区进一步扩大。超声波的细化效果来自于声空化造成微区瞬时过冷,以及动态形核;而声流造成凝固的枝晶破碎,提供结晶核心。  相似文献
