The mechanical reliability of products must be assured for scaling up and production of complex-shaped components by spark plasma sintering (SPS) of spray-dried granules. The evolution of morphologies of pores and defects, which control the mechanical strength, is investigated by using synchrotron X-ray multiscale tomography during SPS of alumina granules at 1300 °C. While large defects arising from the hierarchical granule packing structure cannot be removed by pressureless sintering, crack-like defects and branched rodlike defects are almost eliminated by SPS at stresses higher than 30 and 50 MPa, respectively. But, small ellipsoidal porous regions, which may arise from aggregates or dimples of granules, cannot be removed even at a pressure of 50 MPa. A very large defect is also found by using micro-CT. It is supposed that this defect is formed from a large void in loosely packed granules. The shrinkage of large voids and the elimination of crack-like defects are explained by the theoretical prediction based on the continuum theory of sintering.  相似文献

Biosynthetic genes are not only responsible for the formation of bioactive substances but also suited for other applications including gene therapy. To test the feasibility of human cells producing antibiotics in situ when provided with a heterologous biosynthetic gene, we focused on cytochrome P450, the class of enzymes important in conferring bioactivity to natural product precursors. We selected Fma-P450 that plays a central role in the fumagillin antimicrobial biosynthesis in Aspergillus fumigatus to examine fungal metabolite production by HeLa cells that express fma-P450 heterologously. Here we show that HeLa cells harboring fma-P450 can biosynthesize 5-hydroxyl-β-trans-bergamoten and cytotoxic 5-epi-demethoxyfumagillol when supplemented with the nontoxic precursor β-trans-bergamotene. While the production level was insufficient to effect cell death, we demonstrate that programming human cells to autogenerate antibiotics by introducing a heterologous biosynthetic gene is feasible.  相似文献

In situ observation of sintering of ceramics is important to understand the sintering behavior, including the development of internal structures, and to fabricate ceramics with superior properties. However, there has been no studies thus far on high-speed in situ observation of the internal structure of green and sintered bodies at high temperatures with micrometer-scale resolution. Here, we applied swept-source optical coherence tomography (OCT) to observe the evolution of the internal structure in Al2O3 green bodies during sintering. In situ OCT observations revealed that internal structure changes during sintering, such as the development of density distribution and growth of coarser pores. We could also obtain the sintering shrinkage ratio and relative density from the OCT images. The OCT observation of the internal structure during sintering of green bodies with added or stacked granules of different primary particle sizes confirmed that inhomogeneous regions developed as the densification progressed at high temperatures. These results indicate that in situ observation of the sintering behavior of ceramic green bodies using swept-source OCT is a novel technique that facilitates real-time observation of the evolution of the internal structure during sintering.  相似文献

In this study, the yield stress and fracture strength of β $\ubeta$ -Si3N4 single crystals were directly measured by bending tests of microcantilever beam specimens that were prepared by a focused ion beam method. The β $\ubeta$ -Si3N4 single crystals were plastically deformed at room temperature under high bending stress, and the yield stress depended on the crystal orientation. Transmission electron microscopy observation of the specimens after bending tests indicates that the plastic deformation resulted from dislocations in the primary slip system { 10 1 ¯ 0 } $\{ 10\bar{1}0\} $ <0001>, and the critical resolved shear stress of this slip system determined from the yield stress was 1.34 GPa. The fracture strength of β $\ubeta$ -Si3N4 single crystals ranged approximately up to 20 GPa, depending on the crystal orientation as with the yield stress. The fracture behavior of β $\ubeta$ -Si3N4 single crystals was discussed in terms of the accumulation of dislocations.  相似文献

Heavy metals such as zinc are widely used in plating plants for the processing of material surfaces; therefore, an adequate wastewater treatment is required afterwards to comply with wastewater regulations. However, the presence of chemicals such as degreasing agents in the wastewater generated from the plating process inhibits the formation of hydroxides, thereby making treatment of wastewater and compliance with the stipulated regulations difficult. Herein, the effects of fine bubbles on oil cleaning and wastewater treatment were investigated for developing an alternative cleaning technology to degreasing agents. The concentration of fine bubbles significantly affected the oil cleaning. The fine bubbles demonstrated remarkable cleaning of highly viscous oil, which was difficult to remove using pure water. The high cleaning effect of fine bubbles on oil was achieved because of the hydrophobic interaction of the bubbles. Furthermore, a decrease in cleaning effect was observed for samples with uneven shapes, which was attributed to the difficulty in penetration of fine bubbles in materials with irregularities. Furthermore, fine bubble cleaning exhibited better efficiency for wastewater treatment than degreaser cleaning. Therefore, fine bubble cleaning is speculated to be an excellent cleaning technology with high oil cleaning effectiveness and improved wastewater treatability.  相似文献
