The application of siRNA in gene therapy is mainly limited because of the problems with its transport into cells. Utilization of cationic dendrimers as siRNA carriers seems to be a promising solution in overcoming these issues, due to their positive charge and ability to penetrate cell membranes. The following two types of carbosilane dendrimers were examined: CBD-1 and CBD-2. Dendrimers were complexed with pro-apoptotic siRNA (Mcl-1 and Bcl-2) and the complexes were characterized by measuring their zeta potential, circular dichroism and fluorescence of ethidium bromide associated with dendrimers. CBD-2/siRNA complexes were also examined by agarose gel electrophoresis. Both dendrimers form complexes with siRNA. Moreover, the cellular uptake and influence on the cell viability of the dendrimers and dendriplexes were evaluated using microscopic methods and XTT assay on MCF-7 cells. Microscopy showed that both dendrimers can transport siRNA into cells; however, a cytotoxicity assay showed differences in the toxicity of these dendrimers.  相似文献

The Canadian Heart Health Initiative-Ontario Project (CHHIOP) investigates predisposition and capacity in Ontario public health departments to implement community-based heart health promotion activities. The research draws upon diffusion of innovations theory and recent work on ecological approaches to health promotion within which public health agencies are seen to play a central role. Mail-back surveys were completed by heart health staff in all 42 health departments in 1994 and 1996. Predisposition and capacity were measured as the importance and effectiveness ascribed to 18 organizational practices supportive of community heart health activities. Level of implementation was reported for 74 activities spanning a range of risk factors and settings. Predisposition, capacity and implementation scores increased between 1994 and 1996. The findings confirm positive correlations between predisposition and capacity and between capacity and implementation.  相似文献

The sex pheromone of feral sweet potato weevils Cylas formicarius elegantulus from Cuba was found, via solid-phase microextraction analysis, to be identical to (Z)-3-dodecenyl (E)-2-butenoate, a previously reported compound. Females emitted 20 pg pheromone d−1. In scanning electron microscopy studies carried out on the male antenna, we identified several types of sensilla: sensilla trichoidea of type 1 (ST1) as long hairs (100–150 μm), sensilla trichoidea of type 2 as short hairs (50–60 μm), sensilla basiconica of type 1 as thick pegs (20–25 μm), sensilla basiconica of type 2 as curved pegs (10–15 μm), and sensilla basiconica of type 3 as thin and straight short pegs (15–20 μm). The same types were observed in female antennae but ST1 were far less abundant than in males. Sensilla chaetica were also found on the flagellum subsegments in both sexes. In electrophysiological tests, the crotonate function in the pheromone structure proved to be critical for activity since regular depolarizations (0.6–0.8 mV) were obtained with puffs on 1 μg of the attractant, but not with puffs of the formate, acetate, propionate, or butyrate analogue of the pheromone. In a double dual-choice olfactometer, males showed maximum activity between the 4th and the 8th hr of scotophase at a dose of 50–1000 ng of pheromone. In field tests, a correlation between the contents of the Z,E isomer in the pheromone formulation with activity was noticed, and baits containing this isomer of stereomeric purity >94% showed the highest attractivity. The presence of 5% of the Z,Z isomer in the lure did not induce any synergistic or inhibitory effect, and the alcohol precursor of the pheromone was inactive. The results show that use of a stereomerically pure pheromone may not be necessary in pest control strategies.  相似文献

PDE7 inhibitors regulate pro‐inflammatory and immune T‐cell functions, and are a potentially novel class of drugs particularly useful for treatment of a wide variety of immune and inflammatory disorders. Structural optimization of thioxoquinazoline derivatives led to new compounds with very interesting profiles as PDE7 or PDE7/PDE4 dual inhibitors, which may be further developed as new drugs for inflammatory and neurological diseases.

Biomarkers for placental dysfunction are currently lacking. We recently identified SPINT1 as a novel biomarker; SPINT2 is a functionally related placental protease inhibitor. This study aimed to characterise SPINT2 expression in placental insufficiency. Circulating SPINT2 was assessed in three prospective cohorts, collected at the following: (1) term delivery (n = 227), (2) 36 weeks (n = 364), and (3) 24–34 weeks’ (n = 294) gestation. SPINT2 was also measured in the plasma and placentas of women with established placental disease at preterm (<34 weeks) delivery. Using first-trimester human trophoblast stem cells, SPINT2 expression was assessed in hypoxia/normoxia (1% vs. 8% O2), and following inflammatory cytokine treatment (TNFα, IL-6). Placental SPINT2 mRNA was measured in a rat model of late-gestational foetal growth restriction. At 36 weeks, circulating SPINT2 was elevated in patients who later developed preeclampsia (p = 0.028; median = 2233 pg/mL vs. controls, median = 1644 pg/mL), or delivered a small-for-gestational-age infant (p = 0.002; median = 2109 pg/mL vs. controls, median = 1614 pg/mL). SPINT2 was elevated in the placentas of patients who required delivery for preterm preeclampsia (p = 0.025). Though inflammatory cytokines had no effect, hypoxia increased SPINT2 in cytotrophoblast stem cells, and its expression was elevated in the placental labyrinth of growth-restricted rats. These findings suggest elevated SPINT2 is associated with placental insufficiency.  相似文献

Multi-phase ac motor drives are nowadays considered for various applications, due to numerous advantages that they offer when compared to their three-phase counterparts. In principle, control methods for multi-phase machines are the same as for three-phase machines. Variable speed induction motor drives without mechanical speed sensors at the motor shaft have the attractions of low cost and high reliability. To replace the sensor, information of the rotor speed is extracted from measured stator currents and voltages at motor terminals. Vector controlled drives require estimating the magnitude and spatial orientation of the fundamental magnetic flux waves in the stator or in the rotor. Open-loop estimators and closed-loop observers are used for this purpose. They differ with respect to accuracy, robustness, and sensitivity against model parameter variations. This paper analyses operation of an open-loop and model reference adaptive system (MRAS)-based sensorless control of vector controlled five-phase induction machine with current control in the stationary reference frame. The MRAS-based sensorless operation of a three-phase induction machine is well established and the same principle is extended in this paper for an IRFOC five-phase induction machine. Performance, obtainable with hysteresis current control, is illustrated for a number of operating conditions on the basis of simulation results. Full decoupling of rotor flux control and torque control is realised. Dynamics, achievable with a five-phase vector controlled induction machine, are shown to be essentially identical to those obtainable with a three-phase induction machine. This article proposes a novel framework for the recognition of six universal facial expressions. The framework is based on three sets of features extracted from a face image: entropy, brightness, and local binary pattern. First, saliency maps are obtained using the state-of-the-art saliency detection algorithm “frequency-tuned salient region detection”. The idea is to use saliency maps to determine appropriate weights or values for the extracted features (i.e., brightness and entropy).We have performed a visual experiment to validate the performance of the saliency detection algorithm against the human visual system. Eye movements of 15 subjects were recorded using an eye-tracker in free-viewing conditions while they watched a collection of 54 videos selected from the Cohn-Kanade facial expression database. The results of the visual experiment demonstrated that the obtained saliency maps are consistent with the data on human fixations. Finally, the performance of the proposed framework is demonstrated via satisfactory classification results achieved with the Cohn-Kanade database, FG-NET FEED database, and Dartmouth database of children’s faces.  相似文献

This article presents fabrication, characterization, and performance evaluation of polyetherimide (PEI)/polyvinyl acetate (PVAc) blend membranes. Polymer blend membranes with various blend ratios of PEI/PVAc were prepared by solution casting and evaporation technique. Morphology and miscibility of polymer blend membranes were characterized by field emission scanning electron microscope (FESEM) and differential scanning calorimetry (DSC), respectively. The interaction between blend polymers was analyzed by FTIR analysis. Gas separation performance was evaluated in terms of permeability and selectivity. FESEM results revealed that pure polymer and blend membranes were homogeneous and dense in structure. A single glass transition temperature of polymer blend membranes was found in DSC analysis which indicated the miscibility of PEI/PVAc blend. FTIR analysis confirmed the presence of molecular interaction between blend polymers. The permeation results showed that the presence of PVAc (3 wt%) in blend membranes has improved CO2 permeability up to 95% compared to pure PEI membrane. In addition, CO2/CH4 selectivity was found to be 40% higher than pure PEI membrane. This study shows that blending a small fraction of PVAc can improve the gas separation performance of PEI/PVAc blend membranes. POLYM. ENG.
