Five monoclonal antibodies (mAbs) that recognize human glutamate dehydrogenase (GDH) have been selected and designated as monoclonal antibodies hGDH60-6, hGDH60-8, hGDH63-10, hGDH63-11, and hGDH91-14. A total of five mAbs recognizing different epitopes of the enzyme were obtained, two of which inhibited human GDH activity. When total proteins of human homogenate separated by SDS- PAGE, were probed with mAbs, a single reactive protein band of 55 kDa, which co-migrated with purified recombinant human GDH was detected. When the purified GDH was incubated with each of the mAbs, its enzyme activity was inhibited by up to 58%. Epitope mapping analysis identified, two subgroups of mAbs recognizing different peptide fragments. Using the individual anti-GDH antibodies as probes, the cross reactivities of brain GDH obtained from human and other animal brain tissues were investigated. For the human and animal tissues tested, immunoreactive bands on Western blots appeared to have the same molecular mass of 55 kDa when hGHD60-6, hGHD60-8, or hGHD91-14 mAbs were used as probes. However, the anti-human GDH mAbs immunoreactive to bands on Western blots reacted differently on the immunoblots of the other animal brains tested, i.e., the two monoclonal antibodies hGDH63-10 and hGDH63-11 only produced positive results for human. These results suggest that human brain GDH is immunologically distinct from those of other mammalian brains. Thorough characterization of these anti-human GDH mAbs could provide potentially valuable tool as immunodiagnostic reagents for the detection, identification and characterization of the various neurological diseases related to the GDH enzyme. 相似文献

A biphasic effect of ethanol was observed on Ca2+-uptake activity, Ca2+-uncoupled respiratory control ratio (CURCR), and State 3 oxygen uptake. At low concentrations (0.1-0.5%) ethanol enhanced these activities, probably as the result of an increase in membrane fluidity. Cyclosporin A (CsA) acts in two distinct modes against Schistosoma mansoni (S. mansoni) infection. It behaves in part as an antihelminthic and also as an immunomodulator. In this study, two methods were performed to increase the efficacy of the subcurative dose of CsA (30 mg/kg). These methods were, passive immunization where the drug was given simultaneously with rabbit antisera post infection and active immunization where the worm surface antigen was given prior to infection. The results showed that both methods led to enhancement of the curative effect of the CsA. However, the active immunization gave more significant reduction in liver worm and egg loads. Histopathological study showed small size and few number of hepatic granulomas in the passively immunized group while in the active group, there was just few and small collection of inflammatory cells. Moreover, the indirect fluorescent antibody test visualized damage to the worm tegument in both types of immunization. Thus, it could be concluded that active immunization increased the immunomodulatory effect of CsA in treatment of S. mansoni with less toxic effects by using a subcurative dose. 相似文献

Microbiological analyses of workers' hands were made for the common indicators, including aerobic mesophilic plate counts (APC), as well as the common food pathogens. Opportunities were observed for cross-contamination of roast beef by workers' hands during slicing operations. Workers' hands showed APC counts of up to 10(7) CFU/hand and the presence of S. aureus and C. perfringens. Salmonella spp were not isolated from hands. These results show that handling of these foods by such workers would be a risk in transmitting pathogenic microorganisms to the foods and is apparent that it is necessary for these workers to take care of personal hygiene. Decimal reductions obtained in the microbiological counts after washing and antisepsis of workers' hands were at 2,6 logs cycles and still demonstrated the importance of this practice in food services by the fact that pathogens such as S. aureus and C. perfringens were inhibited or killed. 相似文献

This case report details a single patient with pure red cell aplasia (PRCA) associated with clonal CD3+, TCRalphabeta+, TCR-Vbeta8+, CD8+, CD57+ large granular lymphocytosis whose anaemia did not respond to conventional immunosuppressive therapy but did respond to cyclosporin A (CsA).
