We found 11 genes (FAO1-11) encoding putative oxidoreductases in the Aspergillus oryzae genome, which are similar to fungal fructosyl-amino acid oxidases. The cDNAs corresponding to the genes were cloned and expressed in Escherichia coli. rFao2 had fructosyl-amino acid oxidase activity, whereas rFao1 did not show any enzyme activity, even though the deduced amino acid sequence of Fao1 is identical to that of one of the fructosyl-amino acid oxidase isozymes from Aspergillus oryzae. rFao7 and rFao8 showed oxidase activity toward sarcosine, L-pipecolate, and L-proline. rFao10 was active toward only sarcosine, of the substrates tested. The functions of the other proteins were also predicted from a phylogenetic analysis.  相似文献

Offshore wind simulations were performed with the Weather Research and Forecasting (WRF) model driven by three different sea surface temperature (SST) datasets for Japanese coastal waters to investigate the effect of the SST accuracies on offshore wind simulations. First, the National Centers for Environmental Prediction Final analysis (FNL) (1° × 1° grid resolution) and the Operational Sea Surface Temperature and Sea Ice Analysis (OSTIA) (0.05° × 0.05° grid resolution) datasets were compared with in situ measurements. The results show a decrease in accuracy of these datasets toward the coast from the open ocean. Aiming at an improved accuracy of SST data, we developed a new high‐resolution SST dataset (0.02° × 0.02° grid resolution). The new dataset referred to as MOSST is based on the Moderate Resolution Imaging Spectroradiometer (MODIS) product, provided by the Japan Aerospace Exploration Agency (JAXA). MOSST was confirmed to be more accurate than FNL and OSTIA for the coastal waters. Then, WRF simulations were carried out for 1 year with a 2 km grid resolution and by using the FNL, OSTIA and MOSST datasets. The use of the OSTIA dataset for a WRF simulation was found to improve the accuracy when compared with the FNL dataset, and further improvement was obtained when the MOSST dataset was applied. The sensitivity of wind speed and wind energy density to SST is also discussed. We conclude that the use of an accurate SST is a key factor not only for realistic offshore wind simulations near the surface but also for accurate wind resource assessments at the hub height of wind turbines. The statistical scatter of fracture toughness in the ductile-brittle transition temperature range was experimentally examined on a 500 MPa class low carbon steel. Fracture toughness tests were replicatedly performed at −60 °C, −20 °C and −10 °C. The tests at −60 °C resulted in a single modal Weibull distribution with a shape parameter of 4 for the critical stress intensity factor converted from J-integral, whereas the Weibull distributions of the critical stress intensity factor at −20 °C and −10 °C showed a bilinear pattern with an elbow point, which caused a wider scatter than that at −60 °C. Such scatter transition behavior was discussed with reference to stable crack initiation. A model of the statistical scatter transition has been proposed in this work and the model reasonably explains the experimental results.  相似文献

Colorful rutile TiO2 was prepared by heating Ti2O3 at 550–900 °C to develop novel visible-light-sensitive and eco-friendly photocatalysts for environmental remediation under visible-light irradiation. The colors of the prepared samples, which ranged from grayish green to yellowish off-white via yellow differed from the reported colors of reduced TiO2, such as blue and black. The TiO2 prepared in this study was characterized by X-ray diffraction, Raman spectroscopy, X-ray photoelectron spectroscopy, and UV–Visible spectroscopy. These measurements showed that the TiO2 contained Ti3+-interstitial sites. The TiO2 was sensitive to visible light, and calculation of the band diagram demonstrated that this visible-light absorption is caused mainly by formation of Ti3+-interstitial sites in rutile TiO2. Among the prepared samples, the TiO2 prepared by heating Ti2O3 at 700 °C shows the highest photocatalytic activity under visible-light irradiation. In addition, the sample was further and mildly ground using a bead-milling machine. The ground sample possessed higher surface area and better photocatalytic activity.  相似文献

We propose wedge type light guide plate (LGP) adopting twin inclined cylindrical surfaces (TICS) at the incident part of a LGP. It has higher optical efficiency than conventional wedge type LGP. In dimension of LGP which the thickness of incident part is 1.5 mm and that of luminance part is 0.7 mm, the optical efficiency of wedge type LGP adopting TICS is expected to be 96% in comparison with a normal flat LGP. In addition, there is an advantage to reduce the non‐uniformity of vicinity of light emitted diodes (LEDs) by TICS reflecting light laterally. However, it is necessary that the LED centers are aligned with the intersections of TICS. The misalignment of LED causes non‐uniformity of optical profile that is slightly larger than a normal LGP.  相似文献

Many efforts have been paid to realize the superior anodes for future Li batteries in either the dry Ar atmosphere or the dry air atmosphere. In this work, in order to clarify the effects of such atmospheres, the most reactive anodes of Li were freshly electrodeposited under the dry Ar or under the dry air condition. The Solid Electrolyte Interface (SEI) formed during the electrodeposition of Li anodes is revealed to have a different chemical composition and protective feature. The Li deposited under the dry air was revealed to have longer cycle life in the electrolyte than that deposited in Ar, even in the electrolyte containing ionic liquid. From the XPS results, the SEI formed in dry air is proved to be different from that formed in Ar gas atmospheres, that is, the SEI formed in dry air consists of Li2CO3 and Li nitride. In order to improve the performance of the anodes, the atmosphere for the initial preparation of the anode/electrolyte interface should be tuned.  相似文献

Sn2+-incorporated potassium titanate (K2Ti6O13) nanoribbons were prepared by a facile acid-free ion-exchanged method in a dehydrated methanol solution at room temperature. XRD patterns suggested that K2Ti6O13 (KTO) and Sn2+-incorporated KTO (SKTO) are well crystallized with monoclinic phase structures. The mole ratio of incorporated Sn2+ to K+ in SKTO was estimated to be 2. X-ray photoelectron spectrum showed that the Sn species of SKTO consisted of 90% of Sn2+ and 10% of Sn4+, suggesting that part of Sn2+ was oxidized to Sn4+ in the incorporation process. The band gap of SKTO was 0.7 eV narrower than that of KTO, which was derived from lift of the top of the valence band due to the hybridization of Sn5s and O2p orbitals. The SKTO nanoribbons showed remarkable photocatalytic activities for H2 evolution and rhodamine B degradation under visible light irradiation (λ ≥ 420 nm). The photocatalytic mechanism and durability were studied in detail. The advantage of this acid-free ion-exchange method is ease of ion-exchange of K+ with H+ and maintenance of the integrity of the 1D nanoribbon structures. This method can be applied to preparation of other Sn2+-incorporated compounds with special nanostructures.  相似文献

A temperature-jump (T-jump) time-resolved X-ray crystallographictechnique using the Laue method was developed to detect small,localized structural changes of proteins in crystals exposedto a temperature increase induced by laser irradiation. In achimeric protein between thermophilic and mesophilic 3-isopropylmalatedehydrogenases (2T2M6T), the initial structural change uponT-jump to a denaturing temperature (~90°C) was found tobe localized at a region which includes a ß-turn and aloop located between the two domains of the enzyme. A mutant,2T2M6T-E110P/S111G/S113E, having amino acid replacements inthis ß-turn region with the corresponding residues ofthe thermophilic enzyme, showed greater stability than the originalchimera (increase of Tm by ~10°C) and no T-jump-inducedstructural change in this region was detected by our method.These results indicate that thermal unfolding of the originalchimeric enzyme, 2T2M6T, is triggered in this ß-turn region.  相似文献

Silk is a protein fiber used to weave fabrics and as a biomaterial in medical applications. Recently, genetically modified silks have been produced from transgenic silkworms. In the present study, transgenic silkworms for the mass production of three colors of fluorescent silks, (green, red, and orange) are generated using a vector originating from the fibroin H chain gene and a classical breeding method. The suitability of the recombinant silks for making fabrics is investigated by harvesting large amounts of the cocoons, obtained from rearing over 20 thousand silkworms. The application of low temperature and a weakly alkaline solution for cooking and reeling enables the production of silk fiber without loss of color. The maximum strain tolerated and Young's modulus of the fluorescent silks are similar to those of ordinary silk, although the maximum stress value of the recombinant silk is slightly lower than that of the control. Fabrics with fluorescent color are demonstrated using the recombinant silk, with the color persisting for over two years. The results indicate that large amounts of genetically modified silk can be made by transgenic silkworms, and the silk is applicable as functional silk fiber for making fabrics and for use in medical applications.  相似文献
