Membranes were prepared from solutions containing Udel‐type polysulfone (PSf) and sulfonated poly(2,6‐dimethyl‐1,4‐phenylene oxide) (SPPO). Polymer solutions in 1‐methyl‐2‐pyrrolidone were cast on a nonwoven textile and precipitated in a water bath. The permeabilities and selectivities of the prepared membranes depended on the concentrations of both polymers in the casting solution. The higher the concentration of PSf, the lower were the permeabilities to water and average pore sizes of the membranes. On the other hand, a very small amount of SPPO in the casting solution (about 1–4 wt % relative to the casting solution weight) brought about a considerable increase in water permeabilities and had a small influence on the average pore sizes. The effects were most pronounced if SPPO with a degree of sulfonation of 20–40% was used. The considerable increase in water permeabilities was explained by separation of the PSf and SPPO phases during precipitation in water and by the concentration of hydrophilic SPPO on the surface of the membrane and its pores. The determinations of the oriented concentration potentials proved the presence of a negative surface charge in the membranes. © 2001 John Wiley & Sons, Inc. This report describes the clinical and histologic features of a pulmonary tumor in a 14-year-old girl that is most consistent with a rare entity described in the literature as "pulmonary endodermal tumor resembling fetal lung" (PET). This tumor is composed of glycogen-rich columnar cells forming complex glands with focal festooning and mitotic activity, admixed with solid "morules" of cells with eosinophilic cytoplasm and focal nuclear clearing. Patchy tumor necrosis and a bland stroma were also present. Immunoreactivity for carcinoembryonic antigen (CEA), alpha 1-antichymotrypsin, and 12E7 was present in glandular cells and for human chorionic gondatropin (HCG), alpha 1-antichymotrypsin, and 12E7 in morular cells. Ultrastructural features are those of an epithelial tumor. Related entities have been called "pulmonary blastoma lacking sarcomatous elements" and "adenocarcinoma of fetal lung type." Most cases of PET have occurred in adults, and the histologic features thought to have prognostic significance in small published series are applied to our case, in which the patient remains well and without evidence of tumor recurrence or metastasis for 28 months following local resection as the sole treatment.  相似文献

The 'small' (43 kDa) sialidase of Clostridium perfringens is inhibited by low concentrations of mercury ions. For the investigation of possible functional roles of the enzyme's four cysteine residues at the amino acid positions 2, 282, 333 and 349, they were separately altered to serine by site-directed mutagenesis. The four mutant sialidases expressed in E. coli and purified by metal chelate chromatography were markedly reduced in specific activity when compared to the wild-type enzyme but with the exception of C282S exhibited similar K(M)-values indicating an unchanged mode of substrate binding. The substrate specificity was also conserved for C2S, C282S, and C333S. Only the C349S sialidase exhibited a higher relative activity with colominic acid and the alpha2,6-linked sialic acid of sialyllactose compared to the alpha2,3-linked isomer than the other mutants. Chemical modifications with the thiol-blocking reagents N-ethylmaleimide (NEM), p-chloromercuribenzoate (pCMB) and HgCl2 had little effect on the C282S sialidase, e.g., 6% inhibition by 5 mM NEM compared to reductions in activity between 65 and 90% for the wild-type and other mutant enzymes, supporting the idea that among the enzyme's cysteines, Cys-282 has the highest structural or functional significance. The results also explain the higher mercury tolerance of Salmonella typhimurium and Clostridium tertium sialidases, which have the positions equivalent to Cys-282 altered to Val and Thr, respectively, indicating that the thiol group of Cys-282, despite being situated near the active site, is not involved in catalysis.  相似文献

PURPOSE: To investigate the development of fluoroquinolone resistance among Neisseria gonorrhoeae isolates in Japan and the frequency and patterns of mutations involving the GyrA and ParC proteins, which confer quinolone resistance to the bacteria, in isolates. MATERIALS AND METHODS: Antimicrobial susceptibilities of 145 gonococcal isolates, including 79 isolated from February 1992 through February 1993 and 66 isolated from February 1995 through February 1996, to six fluoroquinolones and several other antibiotics were compared with those of 27 isolates obtained from 1981 through 1984. To identify mutations in gyrA and parC genes of the isolates, the quinolone resistance-determining regions of the gyrA and parC genes were PCR-amplified and the PCR products were directly sequenced. RESULTS: The minimum inhibitory concentration for 90% of strains (MIC90) values of norfloxacin for the isolates from 1992 to 93 (4 microg./ml.) and 1995 to 96 (8 microg./ml.) were 16- and 32-fold, respectively, higher than those for isolates from 1981 to 84 (0.25 microg./ml.). The MIC90 values of ciprofloxacin for isolates from 1992 to 93 (0.5 microg./ml.) and 1995 to 96 (1 microg./ml.) showed increase of 8- and 16-fold, respectively, in comparison with those from 1981 to 84 (0.063 microg./ml.). The isolates from 1992 to 93 and 1995 to 96 were also less susceptible to newer fluoroquinolones including levofloxacin, sparfloxacin, DU-6859a and AM-1155, as compared with those from 1981 to 84. In 46 (67.6%) and 16 (23.5%) of the 68 gonococcal strains sequenced, GyrA and ParC mutations were identified, respectively. No ParC substitutions were identified in any isolates without co-existence of the GyrA mutation. A Ser-91 to Phe mutation, which was detected in 30 (65.2%) of the 46 isolates with GyrA mutations, was the most common GyrA mutation. Mutants with the single Ser-91 to Phe substitution in GyrA were 12-fold and at least 13-fold, respectively, less susceptible to norfloxacin and ciprofloxacin than the wild type. CONCLUSIONS: The results obtained in this study suggest that a high prevalence of gonococcal isolates with the Ser-91 to Phe mutation in GyrA has reduced the susceptibility of this organism to fluoroquinolones in Japan.  相似文献

The stellarator Wendelstein 7-X is under construction at the Max-Planck-Institut für Plasmaphysik in Greifswald. Its superconducting coil system is fixed by a massive structure. During machine operation the coils exert high forces and moments against each other and the central support structure (CSS). Therefore, the detailed analysis of the coil to CSS connections, the so-called central support elements (CSE), is a critical item. The major details of the design have been frozen; nevertheless, there is still need for detailed analysis of the CSEs due to assembly issues, and later on for exploring operational limits of the machine. These analyses have to be performed quickly, reliably, and shall provide results in a standardized form to enable timely responses to the assembly team. Special numerical tools – finite element (FE) parametric models of CSEs – have been developed for the purpose of such analyses. In the models, the geometry, material properties, contact conditions, loads as well as results presentation are defined in a parametric way. The use of the developed models for the definition of the final weld parameters, bolt preloads, assessment of acceptable tolerances, and optimal positions of the CSE-wedges before welding is also discussed.  相似文献

A new single-crystal scintillator intended for applications in a scanning electron microscope (SEM) is presented. It is a single-crystal disc of yttrium aluminium perovskite activated by trivalent cerium free of traces of Fe ions. The single crystals of YAlO3: Ce3+ (YAP: Ce3+) were prepared by the Czochralski method in a reducing atmosphere of Ar and H2 with an excess amount of Y3+ ions. Effective methods of purification and purity control of the raw material are described. The highest achievable concentration of Ce3+ ions in as grown crystals amounted to 0.3 wt%, concentration in the melt was 8–9 times higher. The best properties were found with samples of the maximum possible concentration of Ce3+. Compared with the previous aluminate Y3 Al5 O12: Ce3+ (YAG: Ce3+) these samples showed higher efficiency, a shorter decay time of luminescence (40 ns) and an emission band in a more advantageous spectral region (378 nm). Because of high resistance to radiation damage, high chemical resistance and applicability to ultra-high vacuum it is also suitable for detection of other kinds of ionizing radiation.  相似文献

BACKGROUND: Although the anticancer effects of progesterone therapy for patients with endometrial carcinoma are widely acknowledged, a detailed assessment of the resultant morphologic alterations in tumor tissue kinetics has hitherto been lacking. METHODS: Biopsy and hysterectomy specimens of 14 endometrial carcinomas (endometrioid-type) before and during progesterone therapy were studied to clarify changes in apoptosis and cell proliferation and their relation to morphologic alterations. The extent of squamous differentiation within tumor lesions was also examined. RESULTS: In the good-response group, tumor cells took on characteristics of normal endometrial gland cells in the secretory phase. A positive correlation between reduction in the mitotic index and the degree of morphologic alterations during hormone therapy was observed, but the frequency of apoptotic cells did not vary. In both the good-response and poor-response groups, development or enlargement of squamous areas was observed, in comparison with the initial biopsy specimens. CONCLUSIONS: These results suggest that prolonged progesterone administration can suppress cell proliferation in endometrial carcinomas through tumor cell differentiation without altering apoptosis, resulting in a shift in tissue kinetics toward a relative predominance of cell deletion. In addition, increases in the occurrence of squamous areas within tumors do not always appear to be related to treatment efficacy.  相似文献

Little is known about the usefulness of psychotherapeutic approaches for traumatized refugees who continue to live in dangerous conditions. Narrative exposure therapy (NET) is a short-term approach based on cognitive-behavioral therapy and testimony therapy. The efficacy of narrative exposure therapy was evaluated in a randomized controlled trial. Sudanese refugees living in a Ugandan refugee settlement (N = 43) who were diagnosed as suffering from posttraumatic stress disorder (PTSD) either received 4 sessions of NET, 4 sessions of supportive counseling (SC), or psychoeducation (PE) completed in 1 session. One year after treatment, only 29% of the NET participants but 79% of the SC group and 80% of the PE group still fulfilled PTSD criteria. These results indicate that NET is a promising approach for the treatment of PTSD for refugees living in unsafe conditions. (PsycINFO Database Record (c) 2010 APA, all rights reserved)  相似文献

Aerosol mass (PM10 and PM2.5) and detailed elemental composition were measured in monthly composites during the calendar year of 2007 at a site in Lahore, Pakistan. Elemental analysis revealed extremely high concentrations of Pb (4.4 μg m− 3), Zn (12 μg m− 3), Cd (0.077 μg m− 3), and several other toxic metals. A significant fraction of the concentration of Pb (84%), Zn (98%), and Cd (90%) was contained in the fine particulate fraction (PM2.5 and smaller); in addition, Zn and Cd were largely (≥ 60%) water soluble. The 2007 annual average PM10 mass concentration was 340 μg m− 3, which is well above the WHO guideline of 20 μg m− 3. Dust sources were found to contribute on average (maximum) 41% (70%) of PM10 mass and 14% (29%) of PM2.5 mass on a monthly basis. Seasonally, concentrations were found to be lowest during the monsoon season (July-September). Principle component analysis identified seven factors, which combined explained 91% of the variance of the measured components of PM10. These factors included three industrial sources, re-suspended soil, mobile sources, and two regional secondary aerosol sources likely from coal and/or biomass burning. The majority of the Pb was found to be associated with one industrial source, along with a number of other toxic metals including As and Cr. Cadmium, another toxic metal, was found at concentrations 16 times higher than the maximum exposure level recommended by the World Health Organization, and was concentrated in one industrial source that was also associated with Zn. These results highlight the importance of focusing control strategies not only on reducing PM mass concentration, but also on the reduction of toxic components of the PM as well, to most effectively protect human health and the environment.  相似文献
