Ca2+ signaling has been involved in controling critical cellular functions such as activation of proteases, cell death, and cell cycle control. The endoplasmatic reticulum plays a significant role in Ca2+ storage inside the cell, but mitochondria have long been recognized as a fundamental Ca2+ pool. Protozoan parasites such as Plasmodium falciparum, Toxoplasma gondii, and Trypanosoma cruzi display a Ca2+ signaling toolkit with similarities to higher eukaryotes, including the participation of mitochondria in Ca2+-dependent signaling events. This review summarizes the most recent knowledge in mitochondrial Ca2+ signaling in protozoan parasites, focusing on the mechanism involved in mitochondrial Ca2+ uptake by pathogenic protists.  相似文献

Visceral adipose tissue (VAT) metabolic profiling harbors the potential to disentangle molecular changes underlying obesity-related dysglycemia. In this study, the VAT exometabolome of subjects with obesity and different glycemic statuses are analyzed. The subjects (n = 19) are divided into groups according to body mass index and glycemic status: subjects with obesity and euglycemia (Ob+NGT, n = 5), subjects with obesity and pre-diabetes (Ob+Pre-T2D, n = 5), subjects with obesity and type 2 diabetes under metformin treatment (Ob+T2D, n = 5) and subjects without obesity and with euglycemia (Non-Ob, n = 4), used as controls. VATs are incubated in culture media and extracellular metabolite content is determined by proton nuclear magnetic resonance (1H-NMR). Glucose consumption is not different between the groups. Pyruvate and pyroglutamate consumption are significantly lower in all groups of subjects with obesity compared to Non-Ob, and significantly lower in Ob+Pre-T2D as compared to Ob+NGT. In contrast, isoleucine consumption is significantly higher in all groups of subjects with obesity, particularly in Ob+Pre-T2D, compared to Non-Ob. Acetate production is also significantly lower in Ob+Pre-T2D compared to Non-Ob. In sum, the VAT metabolic fingerprint is associated with pre-diabetes and characterized by higher isoleucine consumption, accompanied by lower acetate production and pyruvate and pyroglutamate consumption. We propose that glucose metabolism follows different fates within the VAT, depending on the individuals’ health status.  相似文献

Industrial competition and environmental concerns lead to the exploration of alternative and energy-efficient technologies for ceramic materials processing. The main objective of this work was to present microwave heating as a viable option for stoneware processing. Stoneware functional properties are presented and discussed, with emphasis on impact strength, water absorption, porosity, and color. Microstructure analyses show that microwave- and gas-fired samples have higher densifications than electrically fired samples. A relevant finding for processing conditions is that microwave firing requires temperatures approximately 100°C lower than those required by conventional firing. Microwave-fired samples’ rupture energies are approximately twice (0.57 ± 0.06 (J)) those of the reference samples (0.26 ± 0.03 (J)), and their water absorptions are approximately one-half (1.5% at 1170°C and 0.8% at 1190°C) of those of the reference samples (2.0%), whereas the water absorption of electrically fired samples at 1180°C has been estimated to be 7.5%. Color analysis also evidences a shift to lower microwave firing temperatures, what is attributed to the enhanced transformations promoted by microwave heating when comparing with the transformations promoted by conventional (gas or electric) heating.  相似文献

Ray tracing technique is an important tool not only to forward but also for inverse problems in Geophysics, which most of the seismic processing steps depend on. However, implementing ray tracing codes can be very time consuming. This article presents a computer library to trace rays in 2.5D media composed by a stack of layers. The velocity profile inside each layer is such that the eikonal equation can be analytically solved. Therefore, the ray tracing within such profile is made fast and accurate. The great advantage of an analytical ray tracing library is the numerical precision of the quantities computed and the fast execution of the implemented codes. Even though ray tracing programs exist for a long time, for example the seis88 package by ?ervený, most of those programs use a numerical approach to compute the ray. Regardless of the fact that numerical methods can solve more general problems, the analytical ones could be part of a more sophisticated simulation process, where the ray tracing time is completely relevant. We demonstrate the feasibility of our codes using several examples (Miqueles et al., 2013)  [1]. The library can also be used for other applications besides seismic, e.g., optics and tomography.  相似文献

In 2011, a vertical‐slot fish pass was built at the Coimbra Açude‐Ponte dam (Mondego River, Portugal), approximately 45 km upstream from the river mouth. The performance of this infrastructure for sea lamprey passage was evaluated between 2011 and 2015 using several complementary methodologies, namely radio telemetry [conventional and electromyogram (EMG)], passive integrated transponder (PIT) telemetry and electrofishing surveys. During the study period, the electrofishing revealed a 29‐fold increase in the abundance of larval sea lamprey upstream of the fish pass. Of the 20 radio‐tagged individuals released downstream from the dam, 33% managed to find and successfully surpass the obstacle in less than 2 weeks, reaching the spawning areas located in the upstream stretch of the main river and in one important tributary. Fish pass efficiency was assessed with a PIT antenna installed in the last upstream pool and revealed a 31% efficiency, with differences between and within migratory seasons. Time of day and river flow significantly influenced the attraction efficiency of the fish pass, with lampreys negotiating it mainly during the night period and when discharge was below 50 m3 s?1. Sea lampreys tagged with EMG transmitters took 3 h to negotiate the fish pass, during which high muscular effort was only registered during passage, or passage attempts, of the vertical slots. The use of complementary methodologies provided a comprehensive passage evaluation for sea lamprey, a species for which there is a considerable paucity of valuable data concerning behavioural, physiological and environmental influences on obstacle negotiation. Clinical practice guidelines in paper format are still the preferred form of delivery of medical knowledge and recommendations to healthcare professionals. Their current support and development process have well identified limitations to which the healthcare community has been continuously searching solutions. Artificial intelligence may create the conditions and provide the tools to address many, if not all, of these limitations.. This paper presents a comprehensive and up to date review of computer-interpretable guideline approaches, namely Arden Syntax, GLIF, PROforma, Asbru, GLARE and SAGE. It also provides an assessment of how well these approaches respond to the challenges posed by paper-based guidelines and addresses topics of Artificial intelligence that could provide a solution to the shortcomings of clinical guidelines. Among the topics addressed by this paper are expert systems, case-based reasoning, medical ontologies and reasoning under uncertainty, with a special focus on methodologies for assessing quality of information when managing incomplete information. Finally, an analysis is made of the fundamental requirements of a guideline model and the importance that standard terminologies and models for clinical data have in the semantic and syntactic interoperability between a guideline execution engine and the software tools used in clinical settings. It is also proposed a line of research that includes the development of an ontology for clinical practice guidelines and a decision model for a guideline-based expert system that manages non-compliance with clinical guidelines and uncertainty.  相似文献

Traditional cloud computing providers enable developers to program and deploy applications in the cloud by using platform as a service (PaaS) providers. Despite the benefits of such an approach, this service model usually comes with a high level of the lock in effect into the service provider. The lock in effect occurs when a software developer needs to change its cloud provider. When this happens, it usually means a major application rewrite, specific for the new PaaS. This paper details the initial results of a project whose goal is to build a PaaS where vendor lock in does not occur. With this PaaS, developers wishing to deploy their applications into the cloud may continue to program using their usual development environments. There are no necessary changes required to make the application PaaS compatible. As a proof of concept, we developed an open source PaaS management application as a regular web application and then deployed it on the cloud.  相似文献

The sustainability framework has a strong influence on the policies, practices and procedures of companies. Green creativity plays a pivotal role in the development of initiatives and innovations for the environmental pillar of sustainability. Based on a survey applied to a Portuguese company with an accredited environmental management system (N = 146), the findings reveal that green creativity is influenced in different ways when analysed from distinct perspectives. The leader's green creativity is positively influenced by the subordinate's green self-efficacy and personal identification with the leader, whereas the subordinate's green creativity is positively influenced by their green passion and green self-efficacy, and the leaders' green creativity. This study shows that green creativity develops differentially according to distinct job positions and expectations that in practice must be complementary and synergistic for its effective development.  相似文献
