Two experiments were conducted to evaluate the effect of isopropyl ester of 2-hydroxy-4-(methylthio) butanoic acid (HMBi) on lactation performance of dairy cows. Experiment 1 evaluated the effect of HMBi in diets with 15.3% crude protein (CP) and with different proportions of rumen-degradable and undegradable protein. Variation in rumen-degradable and undegradable protein was achieved by replacing raw with heated soybeans. Experiment 2 was an on-farm trial to evaluate HMBi with a large number of observations and using a farm-formulated diet (17.2% CP). In experiment 1, 20 Holsteins at 100 ± 41 d in milk were allocated to 5 replicated 4 × 4 Latin squares with 21-d periods. Treatments were formed by a 2 × 2 factorial arrangement of raw or heated soybeans with or without HMBi. Paper capsules with HMBi were orally administered twice daily to each cow. Dosage of HMBi was 7.6 g of digestible Met/cow per day. There was no interaction between soybean type and HMBi. Heat-treated soybeans increased the yields of milk, protein, fat, and lactose, and reduced urea N in milk and plasma (PUN) compared with raw soybeans. Rumen microbial yield, dry matter intake (DMI), and the total-tract apparent digestibility of nutrients did not differ between soybean types. There was no evidence for HMBi-driven effects on DMI, milk and components yield, or diet digestibility. Urinary purine derivative excretion and PUN concentration were reduced in HMBi-fed cows compared with cows fed diets without HMBi. In experiment 2, 294 Holstein cows were blocked by parity and milk yield, and randomly assigned to HMBi (8.9 g of digestible Met/cow per day) or control. The final data set had 234 cows (215 ± 105 days in milk; 96 primiparous and 138 multiparous; 114 on control and 120 on HMBi) housed in 4 freestall groups (1 group/treatment per parity). The freestall group was the experimental unit for DMI, diet and orts composition, and feed availability. The HMBi supplement was top dressed for 28 d on the first daily meal of each cow, immediately after feed delivery of the same batch of feed to all 4 freestall groups (3 times per day). Sample collection and feed analysis occurred during the last 5 d. Spot urine samples and blood samples from each cow were obtained for analysis of the urinary allantoin to creatinine ratio and PUN. Feed availability, the contents of CP and neutral detergent fiber in diets and orts, and DMI did not differ. Cows fed with HMBi had greater milk protein yield and concentration compared with control and had no change in milk fat and lactose. Rumen microbial yield was greater and PUN was lower in HMBi-fed cows compared with control. In experiment 1, HMBi decreased rumen microbial yield and did not affect lactation performance, but it increased ruminal microbial yield and the secretion of milk protein in experiment 2. These results suggest that lactation response to HMBi may be partially mediated by ruminal events. Heated soybeans increased the efficiency of N utilization and the yields of milk, protein, fat, and lactose, but did not interact with HMBi supplementation.  相似文献

This study reports on the development and characterization of bovine serum albumin (BSA) nanospheres containing Silicon(IV) phthalocyanine (NzPc) and/or maghemite nanoparticles (MNP), the latter introduced via ionic magnetic fluid (MF). The nanosized BSA-loaded samples were designed for synergic application while combining Photodynamic Therapy and Hyperthermia. Incorporation of MNP in the albumin-based template, allowing full control of the magnetic content, was accomplished by adding a highly-stable ionic magnetic fluid sample to the albumin suspension, following heat denaturing. The material's evaluation was performed using Zeta potential measurements and scanning electron microscopy. The samples were characterized by steady-state techniques and time-resolved fluorescence. The in vitro assay, using human fibroblasts, revealed no cytotoxic effect in all samples investigated, demonstrating the potential of the tested system as a synergistic drug delivery system.  相似文献

trans-Resveratrol, antioxidant activity, phenolic compounds, proximate composition, and fatty-acid composition were quantified in different parts (peel, pulp, and seeds) of four grape varieties: Brazil (Vitis vinifera), Benitaka (V. vinifera), Isabel (Vitis labrusca) and Niagara (V. labrusca). Large concentrations of polyunsaturated fatty acid (18.84 and 9204.27 mg 100 g− 1), linoleic acid (LA, 18:2n-6), from 5890.87 to 9148.93 mg 100 g− 1, and alpha-linolenic (LNA, 18:3n-3), from 5.25 to 56.85 mg 100 g− 1, were found in different parts of the four grape varieties. The lowest concentrations necessary for 50% inhibition of DPPH were (9.26 to 126.91 ??g mL− 1) in seeds; the pulp did not have antioxidant activity. The total phenolic content varied between 0.04 and 122.35 mg EAG g− 1; seeds had the highest concentrations (89.83 to 122.35 mg EAG g− 1), especially in Niagara and Isabel varieties. trans-Resveratrol was detected in peels (1.17 to 12.96 ??g g− 1) and seeds (2.03 to 2.44 ??g g− 1) of the four varieties; however, it was absent in the pulp.  相似文献

Abstract

Shewhart control charts are known to be somewhat insensitive to shifts of small and moderate size. Expectedly, alternative control schemes such as the exponentially weighted moving average (EWMA) charts have been proposed to speed up the detection of such shifts. Unfortunately, applying the ordinary EWMA recursion to count data leads to a control statistic no longer with a fixed discrete range. Therefore, we propose a novel chart which relies on a EWMA control statistic where the usual scalar product is replaced by a thinning operation. Actually, we use the new fractional binomial thinning to avoid the typical over-smoothing ascribable to ceiling, rounding, and flooring operations. The properties of this discrete statistic are similar to the ones of its continuous EWMA counterpart and the run length (RL) performance of the associated chart can be computed exactly using the Markov chain approach for independent and identically distributed (i.i.d.) counts. Moreover, this chart is set in such way that: the average run length (ARL) curve attains a maximum in the in-control situation, i.e., the chart is ARL-unbiased; and the in-control ARL is equal to a pre-specified value. We use the R statistical software to provide compelling illustrations of this unconventional EWMA chart and to compare its RL performance with the ones of a few competing control charts for the mean of i.i.d. Proximate composition (ash, moisture, total protein, and fat contents) and fatty acid profile of Balistes capriscus (grey triggerfish) were assessed over a 6‐months period (April to September, 2011) in animals captured along the coast of Peniche (Portugal). High protein (18.9% to 21.4%) and low lipid (0.5% to 0.8%) contents were determined. The highest lipid level was found in June‐captured animals. High proportions of n‐3 polyunsaturated fatty acids (n‐3 PUFA) were observed (53.87 ± 2.00%) together with substantial quantities of oleic acid (27.58 ± 1.09%) and palmitic acid (16.52 ± 0.93%). Docosahexaenoic acid accounted for 77% of the total PUFA and was 7 times more abundant than eicosapentaenoic acid, and the n‐3/n‐6 ratio was 7.2. Gender did not influence lipid levels and the fatty acid profile, but seasonal variations were observed for PUFA and monounsaturated fatty acid contents. Overall, the nutritional properties of this underexploited species may be comparable to those of other lean fish species with higher economic value.  相似文献

A previous study showed that the use of nitrate by Dekkera bruxellensis might be an advantageous trait when ammonium is limited in sugarcane substrate for ethanol fermentation. The aim of the present work was to evaluate the influence of nitrate on the yeast physiology during cell growth in different carbon sources under oxygen limitation. If nitrate was the sole source of nitrogen, D. bruxellensis cells presented slower growth, diminished sugar consumption and growth‐associated ethanol production, when compared to ammonium. These results were corroborated by the increased expression of genes involved in the pentose phosphate (PP) pathway, the tricarboxylic acid (TCA) cycle and ATP synthesis. The presence of ammonium in the mixed medium restored most parameters to the standard conditions. This work may open up a line of investigation to establish the connection between nitrate assimilation and energetic metabolism in D. bruxellensis and their influence on its fermentative capacity in oxygen‐limited or oxygen‐depleted conditions. Food lipid major components are usually analyzed by individual methodologies using diverse extractive procedures for each class. A simple and fast extractive procedure was devised for the sequential analysis of vitamin E, cholesterol, fatty acids, and total fat estimation in seafood, reducing analyses time and organic solvent consumption. Several liquid/liquid-based extractive methodologies using chlorinated and non-chlorinated organic solvents were tested. The extract obtained is used for vitamin E quantification (normal-phase HPLC with fluorescence detection), total cholesterol (normal-phase HPLC with UV detection), fatty acid profile, and total fat estimation (GC-FID), all accomplished in <40 min. The final methodology presents an adequate linearity range and sensitivity for tocopherol and cholesterol, with intra- and inter-day precisions (RSD) from 3 to 11 % for all the components. The developed methodology was applied to diverse seafood samples with positive outcomes, making it a very attractive technique for routine analyses in standard equipped laboratories in the food quality control field.  相似文献
