Theoretical pelagic primary production of phytoplankton and benthic primary production of periphyton were modelled for two small lakes in Estonia (Northeast Europe). Although located only 500 m apart, the water colour and light attenuation of these two lakes differed markedly. The Secchi depth (SD) in the clear‐water lake was 4.5 m and only 0.47 m in the dark‐water lake. The total phosphorus (TP) concentrations were, respectively, 15 μg/l and 28 μg/l. An empirical model whose inputs were morphometric, light conditions and dissolved organic carbon parameters obtained from in situ measurements was employed for the present study. The model calculated primary production with a time‐step of 10 min, and a spatial resolution of 10 cm, from sunrise to sunset and from lake surface to lake bottom. The primary production of periphyton and phytoplankton was almost equal in the clear lake, whereas only phytoplankton contributed to whole‐lake primary production in the dark lake because of the stronger light attenuation in the water column. The results of the present study indicated the depth‐distribution profiles differed dramatically between the two lakes. The clear lake had a deep, U‐shaped curve, with the productive layer reaching considerable depth soon after sunrise and maintaining a similar profile throughout the light hours. In contrast, the dark lake production declined rapidly with increasing depth, whereas the profile changed over the day reaching the greatest depth at noon.  相似文献

Single-chain antibodies consist of the variable, antigen-bindingdomains of antibodies joined to a continuous polypeptide bygenetically engineered peptide linkers. We have used the flexibleinterdomain linker region of a fungal cellulase to link togetherthe variable domains of an anti-2-phenyloxazolone IgGl and showhere that the resulting single-chain antibody is efficientlysecreted and released to the culture medium of Escherichia coli.The yield of affinity-purified single-chain antibody is 1 -2mg/1 of culture medium and its affinity and stability are comparableto those of the corresponding native IgG.  相似文献

Addition of nitrogen (N) to the field layer of boreal forests has been shown to increase the occurrence of the parasitic fungus Valdensia heterodoxa on Vaccinium myrtillus plants. We investigated whether N addition to soil alters the levels of polyamines in V. myrtillus shoots, and discuss here whether such changes could promote the spread of the parasitic fungus on V. myrtillus. Using HPLC, we analyzed the concentrations of free and conjugated polyamines in healthy and naturally V. heterodoxa-infected V. myrtillus plants, which had received a moderate or high dose of N fertilizer, or no additional N. Fertilization with N increased the concentrations of free diamines (putrescine and diaminopropane), but had no significant effect on conjugated amines. Thus, N-induced changes in the constitutive levels of soluble conjugated amines do not seem to explain the increased parasite susceptibility of V. myrtillus under N enrichment. Generally, the concentrations of free diamines and insoluble conjugated putrescine were higher in diseased than in healthy shoots, suggesting parasite-induced accumulation of diamines. Free spermine seemed to accumulate in unfertilized, diseased plants, but in fertilized plants this induction was dampened, suggesting that N-induced alterations in spermine metabolism may promote the spread of parasites on V. myrtillus under N-enrichment.  相似文献

The triboelectric charging of fungal spores was experimentally characterized during rebound and resuspension. A fungal spore source strength tester (FSSST) was used as a primary aerosol generator for spores of three fungal species and two powders (silicon carbide and silver). The critical velocity of rebound was determined using a variable nozzle area impactor (VNAI), and the charging state of particles after resuspension and rebound was measured using the FSSST, different impactor setups, electrometers, and optical particle counters. In the impactor setups and the FSSST, five different surface materials relevant for indoor environments were used (steel, glass, polystyrene, paper, and polytetrafluoroethylene). The critical velocity of rebound was determined to be 0.57 m/s for fungal spores, which is relatively low compared to silicon carbide and previous results for micron-sized aerosol particles. Based on the rebound impactor measurements, we were able to define the crucial parameters of charge transfer for different particle–surface material pairs. A contact charge parameter, which describes the triboelectric charging during rebound, was found to have a negative correlation with the charging state of the particles after the resuspension from an impactor. This connects the triboelectric charging during rebound and resuspension to each other. Based on the contact charge parameter values, quantified triboelectric series could be formed. The results of this work show that fungal spores can be charged both positively and negatively during rebound and resuspension depending on the fungal species and surface material.

The aim of this study was to examine the response of the autonomic nervous system in younger (mean age 31 yrs, n=14) and older (mean age 54 yrs, n=14) healthy female teachers during work periods of perceived high and low stress. In the younger participants, heart rate, cortisol excretion rate and psychosomatic symptoms were significantly higher during the high work stress period. The older participants experienced no decrease in their heart rate and cortisol excretion during the low stress period and they exhibited no significant decrease in blood pressure after the work in the evening during both periods. It may be concluded that the recovery from the stress in the older teachers was insufficient particularly in view of their elevated diastolic blood pressure during the low work stress period. Ergonomic and individually tailored measures in terms of work time control, specific relaxation techniques, and a part-time retirement may improve the stress management of older teachers.  相似文献

A two level control strategy that stabilizes and optimizes the production of an industrial copper solvent extraction process is presented. The stabilizing layer consists of a multi-input–multi-output controller or two single-input–single-output controllers with additional four feedforward compensators that regulate the flow rates in the copper solvent extraction process. The optimization layer consists of an optimizer that maximizes the production of the copper solvent extraction process and gives setpoints to the controllers at the stabilizing level. The mechanistic plant models, verified with industrial data, are linearized by identifying first and higher order transfer function models from simulated PRBS data. On the basis of the linear models, the interactions of the controlled variables, and the pairing of the controlled and manipulated variables are studied and the optimizer and the controllers designed. The control strategy employing two PI-control loops or a model predictive controller and additionally four feedforward control loops is successfully tested against simulated disturbances and setpoint changes. The control strategy is also compared to the data collected from the industrial plant under manual control. With this two level control strategy the production of the copper solvent extraction process is increased by 3–5% and the process variation is decreased by 70–90% compared to the manual operation of the case industrial plant. The results gained in simulation environment are successful and encouraging for further testing in an industrial plant.  相似文献

Several studies indicate that virtual reality (VR) systems are useful for end-user participation in an environmental design process. However, these systems can be costly and thus support for the decision whether to invest in a VR of some type is useful. This study presents a novel method for analysing the usefulness of a VR system for the purpose of end-user participation. We collected qualitative end-user opinion data in the real environment and then contrasted this data with the capabilities of a VR system. Additionally, to better understand the capabilities of the VR used, we examined how the end-users perceive the used virtual environment, which in this case was CAVE, an immersive VR system where projectors are directed to the walls of a room-sized cube. In this way, we analysed whether the same functions and elements identified by end-users on the actual wards could also be evaluated in the CAVE. Eleven nurses and 11 patients participated in the study by evaluating a bathroom and/or four patient rooms modelled by the CAVE and the actual hospital wards. The CAVE was convenient for evaluating most issues identified by the study participants in the actual hospital wards, i.e. aesthetics; correct location of equipment, supplies and materials; distraction by or the good companion of other patients as well as window position and size and living/workspace. However, it was not possible to evaluate with full certainty the possibilities for bracing against grab bars or other objects in the VR, and this was found to be relevant to the independent functioning of patients with limited mobility. Also, due to the relatively low luminance levels of projectors, evaluations regarding lighting were considered unreliable. Moreover, end-users were not always certain about the sizes and sufficiency of space in the CAVE. Solutions to overcome these limitations were proposed.  相似文献

This work presents experimental [infrared (IR) thermography] and computational (finite element model) results of temperature distributions of an electrokinetic separation chip. Thermal characteristics of both the electrolyte solution and the polymer chip (SU-8) are taken into account in modeling temperature distributions during electrokinetic flow. Multiphysics and multiscale simulation couples electrostatics, heat transfer, and fluid dynamics. The accompanying IR thermography is a non-contact method, which can measure fractional temperature differences with sub-second time resolution. Any structures or temperature marker molecules interfering with the experiment are not needed. Nominal spot size in the IR measurements is 30 μm with a field of view of several millimeters enabling both local and chip-scale temperature monitoring simultaneously. As a result, we present a computer model for electrokinetic chips, which enables simulation of fractional temperature changes during electrophoresis under real operating conditions. The accuracy of the model is within ±1°C when the deviation in electrochemical processes is taken into account. The simulation results also suggest that the temperature on the chip surface qualitatively reflects the temperature inside the microchannel with an average offset of 1–2°C.  相似文献
