In addition to mold, indoor bioaerosols also contain bacterial components that may have implications for human health. Endotoxin is a cell wall component in Gram-negative bacteria present at varying levels indoors that has been found to have respiratory health implications. Streptomyces is a large genus of Gram-positive bacteria, and some species have been shown to produce inflammatory reactions in vitro and in vivo. The aim of this study was to determine predictors of streptomycetes levels in house dust and to compare the variation in streptomycetes levels with that in endotoxin levels. Dust was collected by floor vacuuming from 178 homes in the Cincinnati metropolitan area. Streptomycetes levels were measured by quantitative PCR, and endotoxin was assayed by the Limulus amebocyte lysate method. Associations between home characteristics and bacterial contaminants, expressed as concentration and load, were investigated through multiple regression analyses. The presence of two or more dogs was a strong predictor of both streptomycetes and endotoxin levels. Season of dust collection and levels of outdoor molds were predictors of streptomycetes but not endotoxin levels. In contrast, number of inhabitants was a significant predictor of endotoxin load only. Neither streptomycetes nor endotoxin levels were associated with metrics of moisture damage. 相似文献

Graphene, a two-dimensional carbon allotrope, exhibits excellent optoelectronic properties. The assembly of graphene into films provides a platform to deepen the study of its interaction with varying surfaces, to engineer devices, and to develop functional materials. A general approach to produce graphene films consists of preparing a dispersion and laying it on a substrate of choice, followed by solvent evaporation. Here, we report the preparation of stable suspensions of new types of graphene nanomaterials namely, graphene nanoflowers (GNFs) and multi-layer graphene (MLG) flakes, in ethanol, N,N-dimethylformamide (DMF), and N-methyl-2-pyrrolidone (NMP). Sprayable suspensions of both GNFs and MLG were prepared in DMF/ethanol, which showed high stability, without addition of any surfactant. The stable suspensions were used to deposit micrometer-thick MLG/GNF films on glass substrates. Calculations of initial droplet size and of timescale of droplet evaporation are performed and possible thermophoretic effects on droplet deposition discussed as well. Coating glass substrates with a methacrylic acid–methyl methacrylate (MA) copolymer prior to the deposition significantly improved the adhesion of the nanomaterials to the substrate. With the MA coating, a substrate coverage of nearly 100% was achieved at 14-min spraying time for 0.05 wt% GNF and 0.1 wt% MLG suspensions. Raman spectra of the GNF and MLG films reveal that the films were made of MLG in which the individual graphene layers rotated from each other as in turbostratic graphene. This work provides a general approach to prepare graphene nanomaterial suspensions and to create films for a variety of applications. Chronic low-grade inflammation plays a central role in the pathogenesis of osteoarthritis (OA), and several pro- and anti-inflammatory cytokines have been implicated to mediate and regulate this process. Out of these cytokines, particularly IFNγ, IL-1β, IL-4 and IL-17 are associated with different phenotypes of T helper (TH) cells and macrophages, both examples of cells known for great phenotypic and functional heterogeneity. Chondrocytes also display various phenotypic changes during the course of arthritis. We set out to study the hypothesis of whether chondrocytes might adopt polarized phenotypes analogous to TH cells and macrophages. We studied the effects of IFNγ, IL-1β, IL-4 and IL-17 on gene expression in OA chondrocytes with RNA-Seq. Chondrocytes were harvested from the cartilage of OA patients undergoing knee replacement surgery and then cultured with or without the cytokines for 24 h. Total RNA was isolated and sequenced, and GO (Gene Ontology) functional analysis was performed. We also separately investigated genes linked to OA in recent genome wide expression analysis (GWEA) studies. The expression of more than 2800 genes was significantly altered in chondrocytes treated with IL-1β [in the C(IL-1β) phenotype] with a fold change (FC) > 2.5 in either direction. These included a large number of genes associated with inflammation, cartilage degradation and attenuation of metabolic signaling. The profile of genes differentially affected by IFNγ (the C(IFNγ) phenotype) was relatively distinct from that of the C(IL-1β) phenotype and included several genes associated with antigen processing and presentation. The IL-17-induced C(IL-17) phenotype was characterized by the induction of a more limited set of proinflammatory factors compared to C(IL-1β) cells. The C(IL-4) phenotype induced by IL-4 displayed a differential expression of a rather small set of genes compared with control, primarily those associated with TGFβ signaling and the regulation of inflammation. In conclusion, our results show that OA chondrocytes can adopt diverse phenotypes partly analogously to TH cells and macrophages. This phenotypic plasticity may play a role in the pathogenesis of arthritis and open new therapeutic avenues for the development of disease-modifying treatments for (osteo)arthritis. 相似文献

The dynamic behaviour of an industrial copper solvent extraction mixer–settler cascade is modelled to develop an advanced process control system. First, the process is introduced and the dynamical models are formulated. The testing environment is described and the successful results presented. Only industrially measured variables are required and plant-specific McCabe-Thiele diagrams are utilized to predict copper concentrations. The results with constant and adapted parameters are compared and the importance of parameter adaptation is discussed. Testing the simulator with adapted parameters over a period of 1 month of industrial operating data gave data that followed the real process measurements closely. In the future, the mechanistic models will be used for control system development and testing. The model can be used on all copper solvent extraction plants by modifying the flow configuration and adapting parameters. 相似文献

Indoor air bacteria and fingal spore levels were studied in 71 non-complaint homes. lk data were analyzed according to the season and the higher limit of the range within which 95% of the cases fall was computed. On the basis of the data the following highest normal levels are proposed for winter: for bacteria 5000 cfulm3 and for fingal spores 500 cfulm3. The recommended levels apply in a subarctic climate for urban and suburban homes when the measurements are made using the same method as in this study. We recommend that if abnormal indoor sources are suspected, indoor samples should be taken in winter when the ground is frozen and covered with snow. At that time, the background levels are at their lowest and the abnormal indoor sources are most easily detected. lk recommended levels should not be used as an indicator of a health risk, but as an indication of abnormal indoor sources or insufficient ventilation. 相似文献

Three different techniques for reducing exposure to microorganisms were tested during remediation of moldy buildings. Concentrations of spores (fungi and actinomycetes) were determined by filter sampling before, during, and after remediation. The local exhaust method used for asbestos dismantling was the most effective control method. In the construction zone, concentrations of microorganisms were 4-25 times higher during remediation than before it. In the adjacent area no increase in concentrations was seen. When the construction zone was placed under negative pressure with a fan and isolated with a plastic barrier, concentrations of microorganisms were about 100 times higher there during remediation work. Nevertheless, levels remained low in the adjacent area. The use of a portable exhaust fan with a side-draft hood decreased concentrations of fungi to one-tenth compared with demolition without the control technology. Furthermore, this method prevented the migration of fungal spores from the construction zone to the adjacent area, although it was less effective in prevention of actinomycete spore migration. It also decreased the levels of microorganisms in the construction zone below the preconstruction level within 2 hours. This study showed that levels of airborne microorganisms, including from the working area to adjacent area, can be reduced with commonly used dust control methods during demolition work. However, microorganism levels in the construction zone remained elevated. Therefore, personal protection of construction workers is needed even with control techniques. 相似文献

Abstract. Information technology is a key factor in competitiveness. Business managers increasingly have to make decisions in this field. Their involvement in planning and decision making should be active and genuine. There are numerous methods to support information systems planning and implementation. This study focuses on the organizational approach to generating plans. An interactive framework for evolutionary information systems planning is presented together with the results of applying this framework in six organizations. The author has been working with these organizations very intensively, using an action research approach. The main emphasis is on the success of the organizational method of creating information management strategies. The main finding is that an interactive way of generating strategies is not enough. The role of internal 'project champions' in both planning and implementation is extremely important. Outside facilitators cannot replace their contribution. 相似文献
