We recently described a novel homogeneous assay principle based on upconversion fluorescence resonance energy transfer (UC-FRET), where an upconverting phosphor (UCP) is utilized as a donor. The UC-FRET has now been applied to a competitive homogeneous immunoassay for 17beta-estradiol (E2) in serum, using a small-molecular dye as an acceptor. The assay was constructed by employing an UCP coated with an E2-specific recombinant antibody Fab fragment as a donor and an E2-conjugated small-molecular dye, Oyster-556, as an acceptor. Standard curves for the assay were produced both in buffer and in male serum. Sensitized acceptor emission was measured at 600 nm under continuous laser diode excitation at 980 nm. In buffer, the IC50 value of the assay was 1 nM and in serum 3 nM. The lower limits of detection (mean of zero calibrators, 3 SD) were 0.4 and 0.9 nM, respectively. The measurable concentration range extended up to 3 nM in buffer and 9 nM in serum. Equilibrium in the assay was reached in 30 min. The novel principle of UC-FRET has unique advantages compared to present homogeneous luminescence-based methods and can enable an attractive assay system platform for clinical diagnostics and for high-throughput screening approaches. 相似文献

The aim of this study was to assess tissue reactions to bioabsorbable self-reinforced ciprofloxacin-releasing polylactide/polyglycolide (SR-PLGA) 80/20 screws in rabbits' cranial bone. Two screws were implanted in each rabbit, one screw on either side of the sagittal suture (n = 28 rabbits). Animals were sacrificed after 2, 4, 8, 16, 24, 54 and 72 weeks, four animals per group. On histological examination the number of macrophages, giant cells, active osteoblasts and fibrous tissue layers were assessed and degradation of the screws was evaluated. At 2 weeks, the highest number of macrophages and giant cells were seen near the heads of the screws. After 4 and 8 weeks, the number of giant cells decreased but that of macrophages decreased from 16 weeks and on. Screws were surrounded by fibrous tissue capsule that progressively was growing in thickness by time. Active osteoblasts were seen around the shaft of the screws with the highest number seen at 4 weeks postoperatively. At 16 weeks, compact fragmentation of the screw heads was seen with macrophages seen inside the screw matrices. After 24 weeks, no polarization of the screws was seen. After one year, PLGA screws had been replaced by adipose tissue, fibrous tissue and "foamy macrophages" which had PLGA particles inside them. After 1(1/2) years, the amount of biomaterial remaining had decreased remarkably. The particles of biomaterial were inside "foamy macrophages." Ciprofloxacin-releasing SR-PLGA 80/20 screws elicited a mild inflammatory reaction but did not interfere with osteoblast activity. A cochlear implant (CI)-associated local drug delivery system based on dexamethasone (DMS) was developed with the purpose to inhibit the growth of fibrotic tissue which influences the signal transmission from the CI to the neurons of the inner ear. For the realization of a targeted DMS delivery the following concepts were combined: modification of the silicone-based electrode carrier by incorporation of DMS and a DMS-containing polymeric coating chemically attached on the surface of the electrode carrier. It was demonstrated that the coated CI showed a high coating stability in a simulated implantation procedure. The in vitro drug release studies in a quasi-stationary model revealed a faster DMS release in the initial phase originating from the DMS-containing coatings and then a lower and sustained DMS release originating from the DMS-loaded silicone carrier. The performed in vitro biocompatibility study confirmed that the released DMS was non-toxic for cultured spiral ganglion cells. 相似文献

Accelerated weathering tests were carried out on wood–polypropylene composites containing minerals. Three different mineral fillers were studied: calcium carbonate, wollastonite and talc. Colour changes were evaluated after distinct periods; the total time of exposure of the composites to UV irradiation was 2000 h. The weathering resulted in significant colour fading of the composites. The composites containing mineral fillers had higher changes of colour (lightness) than the reference composite. Scanning electron microscopy analysis revealed deterioration of the polymer surface layer in all weathered composites. Exposure of the reference composite to UV irradiation resulted in the disappearance of the polypropylene surface layer and disclosure of wood fibres, which led to a higher drop in the lignin content of this composite compared to mineral-containing composites. A substitution of part of the wood with mineral fillers resulted in decreased water absorption and thickness swelling of mineral-containing composites, compared to the reference composite. Exposure to water immersion-freeze–thaw cyclic treatment and UV irradiation led to a decrease in the Charpy impact strength of the composites, except for the composite containing talc. 相似文献

Initially discussed are some of Alan Turing's wonderfully profound and influential ideas about mind and mechanism-including regarding their connection to the main topic of the present study, which is within the field of computability-theoretic learning theory. Herein is investigated the part of this field concerned with the algorithmic, trial-and-error inference of eventually correct programs for functions from their data points. As to the main content of this study: in prior papers, beginning with the seminal work by Freivalds et al. in 1995, the notion of intrinsic complexity is used to analyse the learning complexity of sets of functions in a Gold-style learning setting. Herein are pointed out some weaknesses of this notion. Offered is an alternative based on epitomizing sets of functions-sets that are learnable under a given learning criterion, but not under other criteria that are not at least as powerful. To capture the idea of epitomizing sets, new reducibility notions are given based on robust learning (closure of learning under certain sets of computable operators). Various degrees of epitomizing sets are characterized as the sets complete with respect to corresponding reducibility notions! These characterizations also provide an easy method for showing sets to be epitomizers, and they are then employed to prove several sets to be epitomizing. Furthermore, a scheme is provided to generate easily very strong epitomizers for a multitude of learning criteria. These strong epitomizers are the so-called self-learning sets, previously applied by Case & K?tzing in 2010. These strong epitomizers can be easily generated and employed in a myriad of settings to witness with certainty the strict separation in learning power between the criteria so epitomized and other not as powerful criteria! 相似文献

The relationships of micro-pores and cracks in metallurgical coke have been investigated by optical microscope and field emission scanning electron microscope, using surface section samples. The pores have circular, elliptical and irregular shapes with smooth outlines, formed during the thermoplastic stage of the coking process. They often associate with connecting cracks between neighbouring pores. In case of elliptical pores, the connecting cracks are usually oriented along the longer axis of the pore. The connecting cracks can be developed between the pores, depending on their size and the distance between them. The coke with a large number of small pores rather than with a small number of larger pores will have lower strength due to the increased amount of connecting cracks. When compared with circular pores, elliptical and flattened pores have a lower ability to resist load pressure. Nano-sized pores have polygonal outlines, indicating an ‘explosion’-type formation in the solidified matrix. 相似文献

Abstract Experimental observations of the power transfer effect between two writing beams in dry methylene-blue-sensitized gelatin films are presented. A low-power He-Ne laser is used for the realization of two-wave mixing. The influences of the writing angles and the polarization of the writing beams on the holographic enhancement coefficient, which shows the magnitude of the power transfer, are demonstrated. The electrochemical reduction of oxygen on thin-film platinum electrodes in 0.1 M HClO4 and 0.05 M H2SO4 solutions has been investigated using the rotating disk electrode (RDE) method. Thin films of Pt (0.25-20 nm thick) were prepared by vacuum evaporation onto glassy carbon substrate. The surface morphology of Pt films was examined by transmission electron microscopy (TEM). The specific activity of O2 reduction was higher in HClO4 and decreased with decreasing film thickness. In H2SO4, the specific activity was lower and appeared to be independent of the Pt loading. The values of Tafel slopes close to −120 mV dec−1 in high current density range and −60 mV dec−1 in low current density range were obtained for all electrodes in both solutions, indicating that the mechanism of O2 reduction is the same for thin-film electrodes as for bulk Pt. The number of electrons transferred per O2 molecule was close to four for all thin Pt films studied. 相似文献
