The KlLYS2 gene, encoding the alpha-aminoadipate reductase of Kluyveromyces lactis, was isolated by complementation of a lysA1 mutant. The deduced amino acid sequence shared an identity of 73% with the LYS2 product of Saccharomyces cerevisiae. Despite the high sequence homology of the alpha-aminoadipate reductase genes, the two yeast species differently responded to the presence of alpha-aminoadipate in the medium. Wild-type S. cerevisiae is known to be sensitive to alpha-aminoadipate, but becomes resistant when mutated to lys2. In contrast, K. lactis strains were found to be naturally resistant to alpha-aminoadipate. Therefore, the positive selection procedure for the isolation of lys2 mutants on alpha-aminoadipate, as practised in S. cerevisiae, cannot be applied to K. lactis. A possible reason of this difference may be that the catalytic rate of the alpha-aminoadipate reductase differs in the two yeasts. Novel optically active methacrylic copolymers, bearing carbazole and azoaromatic moieties linked to the main chain with a rigid (S)-hydroxysuccinimide residue of one prevailing absolute configuration, have been prepared and characterized with the aim to establish the effects of the chiral group and of the dipolar interactions between the two side-chain chromophores on their properties.In particular, the observed chiroptical properties suggest the presence of ordered chiral conformations at least for chain segments of the macromolecules. Spectroscopic, thermal and chiroptical characterizations of these copolymers demonstrate the occurrence of significant electronic interactions between carbazole chromophores and azobenzene moieties. These interactions are related to the unusual partially alternating structure of the investigated copolymers and the relevant chiroptical and thermal properties. In fact, the polymeric derivates show high glass transition temperatures and good thermal stability, thus opening the way to a variety of potential applications based on photoconductive and optoelectronic properties.  相似文献

Most of the existing efforts for supporting the design, preparation, and deployment of accessible e-learning applications propose guidelines that primarily address technical accessibility issues. Little, if any, consideration is given to the real actors involved in the learning experience, such as didactical experts and disabled learners. Moreover, implementing artifacts addressed to the e-learning world requires a wide range of particular skills which are related not only to technical but also to didactical, pedagogical, usability, and accessibility aspects of the produced material. This paper argues that the know-how of a number of stakeholders should be blended into a joint design activity, and that it should be possible to determine the role of each participant in the successive phases of the development lifecycle of e-learning applications. The paper sketches the methodological guidelines of a design framework based on involving the users with disabilities, as well as pedagogical experts, in the development process. The novelty of this proposal mainly stems from being built up around the core of strategies and choices specifically bound to accessibility requirements. Characteristic elements of learner-centered design are then further integrated into processes and methodologies which are typical of participatory and contextual design approaches. Following such guidelines, it will be possible to gain a deeper understanding of the requirements and of the operational context of people needing accessible material, either as learners or educators. The underlying objective is to increase the potential to realize learning systems that better meet different user needs and that provide a more satisfying learning experience. Moreover, when people get involved in the development process, they gain a sense of ownership of the system and are therefore more likely to accept and “promote” it.  相似文献

Microstructural evolution at 1450°C of two 3 mol% Y2O3─ZrO2 tetragonal polycrystals (Y-TZP) containing different residual impurity amounts has been investigated by analytical and scanning electron microscopy in order to clarify the role of the amorphous intergranular phases. Phase partitioning is interpreted as a process based on dissolution/reprecipitation reactions involving mass transport via the liquid impurity phase. Grain growth is inhibited by yttria migration before the phase equilibrium is reached, then increases drastically.  相似文献

ETI is an Electronic Tool Integration platform designed for project-oriented, domain-specific, public or private interactive experimentation with heterogeneous tools. Its users can experiment at ease over the web with the integrated tools and functionalities, and build own prototypical problem solvers on the basis of components (tools and transformations) contained in the ETI repository. In this paper we describe the graph concept underlying ETIs flexibility, the significant role played by graphs in the ETI platform, and the kind of support ETI offers to deal with graph objects. This way we sketch the potential of a closer cooperation between the ETI community and the community of research on graph-based tools.  相似文献

The use of mobile sensors is of great relevance to monitor critical areas where sensors cannot be deployed manually. The presence of data collector sinks causes increased energy depletion in their proximity, due to the higher relay load under multi-hop communication schemes (sink-hole phenomenon). We propose a new approach towards the solution of this problem by means of an autonomous deployment algorithm that guarantees the adaptation of the sensor density to the sink proximity and enables their selective activation. The proposed algorithm also permits a fault tolerant and self-healing deployment, and allows the realization of an integrated solution for deployment, dynamic relocation and selective sensor activation. We formally prove the termination of our algorithm. Performance comparisons between our proposal and previous approaches show how the former can efficiently reach a deployment at the desired variable density with moderate energy consumption under a wide range of operative settings.  相似文献

Some organochlorine compounds, such as polychlorinated biphenyls (PCBs), have a tendency to bioaccumulate in humans and predators at the top of the food chain. We have recently confirmed the transplacental transfer of these compounds and the present study has been designed on the same material with the aim of investigating their potential health effects on newborns from 70 pregnant women, resident in a Northern Italy industrial town. Organochlorine compounds [namely, p,p′-dichlorodiphenyltrichloroethane (p,p′-DDT), p,p′-dichlorodiphenyldichloroethene (p,p′-DDE), hexachlorobenzene (HCB), and PCBs] have been analyzed both in cord and maternal serum, placenta, and maternal subcutaneous adipose tissue by GC-MSD. p,p′-DDT levels in the adipose tissue resulted significantly (p < 0.05) related to birth length. Mothers of neonates born by preterm programmed caesarean delivery showed significantly (p < 0.005 for both) higher serum p,p′-DDE serum concentrations and p,p′-DDT levels in the adipose tissue, as compared to mothers delivering at term.  相似文献

Embedded Systems, by their nature, constitute a meeting point for communities with extremely different background. In particular, the high demands for quality and reliability for embedded systems have led to complementary quality assurance efforts: hardware engineers have developed techniques for dynamic verification in terms of co-simulation, which, in particular, addresses the different nature of hardware and software components. Thus these techniques are tailored for the transactional level, which comprises dedicated models for the hardware and the software parts. On the other hand, there is a bulk of work on formal verification techniques, which typically address higher levels of abstraction. These techniques are exhaustive in the sense that they cover all the infinite possible paths of their models, however at the price of neglecting many of the low-level aspects treated by co-simulation. It is the goal of this paper to increase the mutual understanding between these communities and to animate research at this exciting borderline.  相似文献
