The ability of sodium polymethacrylate salts with various molecular weights (1200 to 30000 g·mol−1) to deflocculate concentrated aqueous suspensions (58 wt%) of two clays, widely used for stoneware ceramic tile production, was investigated. Rheological measurements were made in distilled water or in water containing calcium ions (0 to 6 × 10−3 mol·dm−3) to evaluate the practical applications of these salts as deflocculants in the ceramic tile industry. All slips presented a non-Newtonian behavior. Polymethacrylates with low molecular weights were the most efficient deflocculants for clay suspensions. The salts acted by an essentially electrosteric stabilization mechanism, in combination with a high sequestering ability toward flocculating cations. For their efficient deflocculating capabilities toward concentrated aqueous clay suspensions, sodium(I) polymethacrylates may be considered for practical industrial application, in particular for the preparation of liquid deflocculants.  相似文献

Amyloid aggregation of human ataxin-3 (ATX3) is responsible for spinocerebellar ataxia type 3, which belongs to the class of polyglutamine neurodegenerative disorders. It is widely accepted that the formation of toxic oligomeric species is primarily involved in the onset of the disease. For this reason, to understand the mechanisms underlying toxicity, we expressed both a physiological (ATX3-Q24) and a pathological ATX3 variant (ATX3-Q55) in a simplified cellular model, Escherichia coli. It has been observed that ATX3-Q55 expression induces a higher reduction of the cell growth compared to ATX3-Q24, due to the bacteriostatic effect of the toxic oligomeric species. Furthermore, the Fourier transform infrared microspectroscopy investigation, supported by multivariate analysis, made it possible to monitor protein aggregation and the induced cell perturbations in intact cells. In particular, it has been found that the toxic oligomeric species associated with the expression of ATX3-Q55 are responsible for the main spectral changes, ascribable mainly to the cell envelope modifications. A structural alteration of the membrane detected through electron microscopy analysis in the strain expressing the pathological form supports the spectroscopic results.  相似文献

Deletion of phenylalanine at position 508 (F508del) in the CFTR chloride channel is the most frequent mutation in cystic fibrosis (CF) patients. F508del impairs the stability and folding of the CFTR protein, thus resulting in mistrafficking and premature degradation. F508del-CFTR defects can be overcome with small molecules termed correctors. We investigated the efficacy and properties of VX-445, a newly developed corrector, which is one of the three active principles present in a drug (Trikafta®/Kaftrio®) recently approved for the treatment of CF patients with F508del mutation. We found that VX-445, particularly in combination with type I (VX-809, VX-661) and type II (corr-4a) correctors, elicits a large rescue of F508del-CFTR function. In particular, in primary bronchial epithelial cells of CF patients, the maximal rescue obtained with corrector combinations including VX-445 was close to 60–70% of CFTR function in non-CF cells. Despite this high efficacy, analysis of ubiquitylation, resistance to thermoaggregation, protein half-life, and subcellular localization revealed that corrector combinations did not fully normalize F508del-CFTR behavior. Our study indicates that it is still possible to further improve mutant CFTR rescue with the development of corrector combinations having maximal effects on mutant CFTR structural and functional properties.  相似文献

The use of graphene nanomaterials (GNMs) for biomedical applications targeted to the central nervous system is exponentially increasing, although precise information on their effects on brain cells is lacking. In this work, the molecular changes induced in cortical astrocytes by few‐layer graphene (FLG) and graphene oxide (GO) flakes are addressed. The results show that exposure to FLG/GO does not affect cell viability or proliferation. However, proteomic and lipidomic analyses unveil alterations in several cellular processes, including intracellular Ca2+ ([Ca2+]i) homeostasis and cholesterol metabolism, which are particularly intense in cells exposed to GO. Indeed, GO exposure impairs spontaneous and evoked astrocyte [Ca2+]i signals and induces a marked increase in membrane cholesterol levels. Importantly, cholesterol depletion fully rescues [Ca2+]i dynamics in GO‐treated cells, indicating a causal relationship between these GO‐mediated effects. The results indicate that exposure to GNMs alters intracellular signaling in astrocytes and may impact astrocyte–neuron interactions.  相似文献

In many applicative fields, there is the need to model and design complex systems having a mixed discrete and continuous behavior that cannot be characterized faithfully using either discrete or continuous models only. Such systems consist of a discrete control part that operates in a continuous environment and are named hybrid systems because of their mixed nature. Unfortunately, most of the verification problems for hybrid systems, like reachability analysis, turn out to be undecidable. Because of this, many approximation techniques and tools to estimate the reachable set have been proposed in the literature. However, most of the tools are unable to handle nonlinear dynamics and constraints and have restrictive licenses. To overcome these limitations, we recently proposed an open‐source framework for hybrid system verification, called Ariadne , which exploits approximation techniques based on the theory of computable analysis for implementing formal verification algorithms. In this paper, we will show how the approximation capabilities of Ariadne can be used to verify complex hybrid systems, adopting an assume–guarantee reasoning approach. This paper studies the microstructure, sliding wear behavior and corrosion resistance of high-velocity oxygen fuel (HVOF)-sprayed FeVCrC-based coatings. Various process parameters were tested to evaluate their effects on the coating properties, which were also compared to those of HVOF-sprayed NiCrBSi and Stellite-6 coatings. The Fe alloy coatings are composed of flattened splats, originating from molten droplets and consisting of a super-saturated solid solution, together with rounded particles, coming from partially unmolten material and containing V- and Fe-based carbide precipitates. All process parameters, apart from “extreme” settings with excess comburent in the flame, produce dense coatings, indicating that the feedstock powder is quite easily processable by HVOF. These coatings, with a microhardness of 650-750 HV0.3, exhibit wear rates of ≈2 × 10?6 mm3/(Nm) in ball-on-disk tests against sintered Al2O3 spheres. They perform far better than the reference coatings, and better than other Fe- and Ni-based alloy coatings tested in previous research. On the other hand, the corrosion resistance of the coating material (tested by electrochemical polarization in 0.1 M HCl solution) is quite low. Even in the absence of interconnected porosity, this results in extensive, selective damage to the Fe-based matrix. This coating material is therefore unadvisable for severely corrosive environments.  相似文献

Airborne particulate matter samples were collected in an urban and a rural–suburban monitoring stations of the city of Rome, Italy, and the particles were analyzed through the Raman microspectroscopy. A careful examination of the spectral bands, performed with a five-(Voigt) curve deconvolution model previously described by the literature and here adapted to the purpose, lead to the characterization of the graphitic and carbonaceous material plus the identification of the mineral particles associated with it. Statistical analysis of the full-width at half-maximum (FWHM) values of the bands, as well as of their intensity ratio, revealed the presence of two classes of soot particles that can be ascribed to a different degree of crystallinity. The population of soot collected at the urban site, where the vehicular emission component prevails, exhibits mostly crystalline characteristics (with a D1 FWHM of 150–155 cm?1), whereas the population collected at the rural–suburban site, particularly the coarse fraction, shows a prevailing amorphous nature (with a D1 FWHM of ~175 cm?1). A similar aspect emerges for the pure black carbon particles, mainly crystalline, and the black carbon particles associated with minerals, generally disordered. These results add useful information and characterization of the soot, a relevant component of the ambient air, and its different features with respect to the urban or rural–suburban areas.
