We developed an in vitro autoradiographic method to localize and quantify active renin in primate tissues. Active renin in monkey kidney sections was labeled with the primate specific renin inhibitor, 3H-CGP29287, and quantitated with autoradiography and computerized densitometry. Microscopic emulsion autoradiography was carried out to clarify the detailed localization of the binding. Non-specific binding to aspartyl proteases other than renin was blocked using 1 mumol/L of N-acetyl-pepstatin. To assess the usefulness of this procedure, binding of 3H-CGP29287 was examined both by film and emulsion autoradiography in the kidneys of monkeys (Macaca fuscata) that were given chronically either an angiotensin converting enzyme inhibitor (trandolapril), an angiotensin II receptor antagonist (E4177), or vehicle. 3H-CGP29287 was found to bind very selectively to the juxtaglomerular apparatus (JGA) under control conditions. In monkeys treated with trandolapril or E4177, 3H-CGP29287 binding was increased in proportion to the increase in renal renin concentration determined enzymatically; in these kidneys, emulsion autoradiography revealed radioinhibitor binding extending far from the JGA. The potency of a series of unlabeled renin inhibitor in competing for 3H-CGP29287 binding in the autoradiographic system closely paralleled their potencies, as determined in inhibiting renin by an enzymatic assay. This technique permits specific labeling of the catalytic site of renin in the monkey kidney sections.  相似文献

A ceramic/metal nanocomposite in tetragonal zirconia-molybdenum system possesses a novel microstructural feature composed of a mutual intragranular nanostructure, in which either nanometer sized Mo particles or equivalent sized zirconia particles are located within the zirconia grains or Mo grains, respectively. A toroidal simulator is used to study the magnetomechanical behavior of a superconducting D-shaped coil set at the center between sectored ferromagnetic plates. Since the magnetic field distribution between the two plates is expected to be almost the same as that created by toroidal field coils arrayed in a torus, the toroidal simulator may be expected to show almost the same magnetomechanical behavior as that of the full torus coils. To verify its validity experimentally, buckling and vibration tests were carried out in a liquid helium cryostat. In addition numerical analyses were performed for the magnetic field distribution to reveal its similarity and for the experimental results from the toroidal simulator to confirm its validity. Its validity is fully demonstrated in the present study.  相似文献

The design and fabrication of a GaAs varactor diode for u.h.f. t.v. tuners were carried out using a GaAs double epitaxial layer. The capacitance ratio of diode. C3/C25, is 6.0 and the series resistance at 470 MHz is 0.24 ?. These characteristics are much better than those of an Si diode and well satisfy the requirements for t.v. tuners.  相似文献

The chemoselective hydrogenation of unsaturated bonds and azide functionalities is achieved in the presence of nitro groups by a heterogeneous palladium catalyst supported on molecular sieves (MS3A). The present method shows a wide‐range of applicability with regard to substrates and the catalyst can be easily prepared and reused at least three times without any loss of activity.  相似文献

This study conducted in vivo and in situ experiments with rats to investigate the glucagon‐like peptide‐1 (GLP‐1) secretion in response to oral or ileal administration of α‐glucosyl‐isoquercitrin (20–40 mmol in 2 mL; Q3G), fructooligosaccharides (200 mmol in 2 mL; FOS) and Q3G + FOS. Direct effects on GLP‐1‐producing l ‐cells were also examined by an in vitro study using a murine enteroendocrine cell line, GLUTag. To evaluate the plasma GLP‐1 level, blood samples from jugular cannula for in vivo and portal cannula for in situ experiments were obtained before and after administration of Q3G, FOS, or Q3G + FOS. We found tendencies for increases but transient stimulation of GLP‐1 secretion by Q3G in in vivo and in situ experiments. Although FOS alone did not have any effects, Q3G + FOS enhanced and prolonged high plasma GLP‐1 level in both experiments. In addition, application of Q3G on GLUTag cells stimulated GLP‐1 secretion while FOS enhanced the effect of Q3G. Our results suggest that Q3G + FOS possess the potential for the management or prevention of Type 2 diabetes mellitus (T2DM) by enhancing and prolonging the GLP‐1 secretion via direct stimulation of GLP‐1 producing l ‐cell.  相似文献

The cells of Streptomyces sp. YB-1 adsorbed 4-6 mg ytterbium (Yb) per g dry weight. The Yb contents of the cell wall fraction, cell-free extract, and cell membrane fraction were 11%, 2%, and 87%, respectively. The Yb content in the cell membrane fraction was 20-25 mg per g dry weight. The adsorbed Yb could be quantitatively desorbed by treating the cell membrane fraction with 1 mM EDTA and 1 M HCl at 37 degrees C for 4 h. Treatment with 1 M NaOH caused Yb desorption to some extent. Treatments with proteinase K, lysozyme, 0.5% Triton X-100, 0.4% sodium dodecyl sulfate, and 1 M NaCl did not cause Yb desorption. Elemental analysis of Yb-adsorbed materials after removal of proteins and then extraction of lipids from the membrane fraction revealed that the molar ratio of Yb and P in the materials was about 1:1. The cells and the membrane fraction could be used repeatedly as a bioadsorbent for Yb.  相似文献

An agarolytic bacterium was isolated from soil in Gifu prefecture, Japan, and identified as Bacillus sp. strain MK03. The strain secreted neoagarooligosaccharide hydroluse into the culture medium. The enzyme was purified 49.7-fold from the culture fluid by ammonium sulfate precipitation and anion-exchange and gel-filtration column chromatographic methods. The purified enzyme appeared as a single band on polyacrylamide gel electrophoresis (PAGE) and sodium dodecyl sulfate (SDS)-PAGE. Estimations of the molecular mass by gel filtration and SDS-PAGE gave values of 320 kDa and 42 kDa, respectively, indicating that the enzyme is octametric. The enzyme cleaved the alpha-1,3 linkage in neoagarobiose to produce 3,6-anhydro-L-galactose and D-galactose. It also selectively cleaved the alpha-1,3 linkage at the nonreducing end in neoagarotetraose or neoagarohexaose to give 3,6-anhydro-L-galactose and agarotriose or agaropentaose. The optimum temperature and pH for the enzyme were 30 degrees C and 6.1, respectively. The N-terminal amino acid sequence showed no homology to sequences of other known neoagarooligosaccharide hydrolases and agarases.  相似文献

This communication reports on the preparation and electric properties of dense nanocrystalline ZnO ceramics. By spark plasma sintering, nanocrystalline (∼100 nm) ZnO ceramics with a high density of 98.5% were obtained at a very low temperature of 550°C. Electric property measurement revealed a novel conduction nonlinearity in the sample sintered at 500°C. This phenomenon is due to the nanometerization of ZnO crystal and the grain boundary layer with an amorphous interfacial layer.  相似文献
