In conventional whole‐tooth culture systems, limitation exists regarding maintenance of the vitality of the dental pulp, because this tissue is encased in rigid dentin walls that hinder nutrition supply. We here report a whole tooth‐in‐jaw‐bone culture system of rat mandibular first molars, where transcardiac perfusion with culture medium was carried out before placement of the jaw bone into culture medium, aiming to facilitate longer time preservation of the dental pulp tissue. Following 7 days of culture, the pulp tissues were analyzed by histology and immunohistochemistry to ED2 (antiresident macrophage). ED2‐positive macrophages were also analyzed for their Class II MHC, interleukin‐6 (IL‐6), and p53 mRNA expression levels by means of immune‐laser capture microdissection (immune‐LCM). Dentin sialophosphoprotein (DSPP) mRNA expression in odontobalstic layer was also examined by LCM. Teeth cultured following saline‐perfusion and nonperfusion served as cultured controls. Normal teeth also served as noncultured controls. Histological examination demonstrated that the structure of the pulp tissue was well preserved in the medium‐perfused explants in contrast to the cultured control groups. The Class II MHC, IL‐6, and p53 mRNA expression levels of ED2‐positive cells and DSPP expression levels of odontoblastic layer tissues in the pulp of medium‐perfused explants were not significantly different from those in the noncultured normal teeth. In conclusion, the structural integrity and mRNA expression in the pulp were maintained at the in vivo level in the ex vivo whole tooth‐in‐jaw‐bone culture system. We have developed a new fabrication method of single-mode self-written waveguide by controlling the propagation mode in an optical fiber. This method is very appropriate for repeatable fabrication of the single-mode self-written waveguide. Since a Gaussian-like near-field pattern is required for the fabrication of a tiny and uniform waveguide core, the propagation mode in a conventional optical communication fiber was controlled by coupling with an optical fiber having 3-/spl mu/m core, which shows a single-mode operation at visible wavelength region. Single-mode propagation at optical communication wavelength was confirmed for the fabricated self-written waveguide. The evaluated core diameter of the self-written waveguide was /spl sim/9.5 /spl mu/m. 相似文献

We have previously demonstrated that the preendosomal compartment in addition to clathrin-coated vesicles, comprises distinct nonclathrin coated endocytic vesicles mediating clathrin-independent endocytosis (Hansen, S. H., K. Sandvig, and B. van Deurs. 1991. J. Cell Biol. 113:731-741). Using K+ depletion in HEp-2 cells to block clathrin-dependent but not clathrin-independent endocytosis, we have now traced the intracellular routing of these nonclathrin coated vesicles to see whether molecules internalized by clathrin-independent endocytosis are delivered to a unique compartment or whether they reach the same early and late endosomes as encountered by molecules internalized with high efficiency through clathrin-coated pits and vesicles. We find that Con A-gold internalized by clathrin-independent endocytosis is delivered to endosomes containing transferrin receptors. After incubation of K(+)-depleted cells with Con A-gold for 15 min, approximately 75% of Con A-gold in endosomes is colocalized with transferrin receptors. Endosomes containing only Con A-gold may be accounted for either by depletion of existing endosomes for transferrin receptors or by de novo generation of endosomes. Cationized gold and BSA-gold internalized in K(+)-depleted cells are also delivered to endosomes containing transferrin receptors. h-lamp-1-enriched compartments are only reached occasionally within 30 min in K(+)-depleted as well as in control cells. Thus, preendosomal vesicles generated by clathrin-independent endocytosis do not fuse to any marked degree with late endocytic compartments. These data show that in HEp-2 cells, molecules endocytosed without clathrin are delivered to the same endosomes as reached by transferrin receptors internalized through clathrin-coated pits. 相似文献

We propose a spiral-groove aerodynamic thrust bearing functioning as a viscous vacuum pump in a laser scanner motor to reduce the windage power loss of a polygon mirror. The proposed bearing pumps out the air in the scanner housing using the pumping effect of the spiral-groove thrust bearing, reducing the inner pressure of the housing. The pumping performances and the static characteristics of the spiral-groove thrust bearings were investigated numerically and experimentally. Two numerical calculation methods were used to study the pumping characteristics of the spiral-groove thrust bearing. It was found that a bearing with 15 spiral grooves reduced the inner pressure of the housing to <0.01 MPa at 20,000 rpm and at bearing clearance of 2.5 μm, and this has sufficient load-carrying capacity to support the rotating shaft and the mirror. 相似文献

Enhancement of the Seebeck coefficient (S ) without reducing the electrical conductivity (sigma) is essential to realize practical thermoelectric materials exhibiting a dimensionless figure of merit (ZT=S2 x sigma x T x kappa-1) exceeding 2, where T is the absolute temperature and kappa is the thermal conductivity. Here, we demonstrate that a high-density two-dimensional electron gas (2DEG) confined within a unit cell layer thickness in SrTiO(3) yields unusually large |S|, approximately five times larger than that of SrTiO(3) bulks, while maintaining a high sigma2DEG. In the best case, we observe |S|=850 microV K-1 and sigma2DEG=1.4 x 10(3) S cm-1. In addition, by using the kappa of bulk single-crystal SrTiO(3) at room temperature, we estimate ZT approximately 2.4 for the 2DEG, corresponding to ZT approximately 0.24 for a complete device having the 2DEG as the active region. The present approach using a 2DEG provides a new route to realize practical thermoelectric materials without the use of toxic heavy elements. 相似文献
